Impact of Treatment Effect Heterogeneity on the Estimation of Individualized Treatment Rules for Count Outcomes.

There is growing interest in tailoring treatment decisions to individual patient characteristics, but few studies have examined the implementation and performance of individualized treatment rules (ITRs) for count data. Our objective was to compare ITR methods in randomized trials with count outcomes and explore the impact of sample size and distribution of heterogeneity of treatment effect (HTE) on the validity of treatment recommendations. We conducted a simulation study where patients were randomized to receive one of two treatments and created five responder strata to reflect different HTE

Dressing patterns in healthy individuals and brain-damaged patients: the MIlan Dressing Assessment.

BACKGROUND: The ability to dress appropriately is a fundamental daily living skill that is often impaired by neurological disorders and can manifest as a specific form of apraxia (dressing apraxia). Despite its prevalence and impact on functional independence, standardized and reliable assessment tools for dressing skills remain limited. The aim of this study was to identify dressing patterns in healthy individuals and use them as a reference for introducing a new clinical tool to assess dressing skills. METHODS: We conducted two studies, including seventy healthy participants (mean age = 48.

Deep Learning to Quantify Body Composition in Lung Cancer Screening-Eligible Individuals.

PURPOSE: Most low-dose computed tomography (LDCT) scans in lung cancer (LC) screening are negative, yet contain prognostic information. We developed a fully automated 3D deep learning model for thoracic body composition (BC) quantification from LDCT and evaluated its prognostic value for overall and cause-specific mortality. MATERIALS AND METHODS: Baseline and first follow-up LDCTs from the National Lung Screening Trial were analyzed. A 3D convolutional neural network was trained for full thoracic volumetric segmentation of subcutaneous adipose tissue (SAT) and skeletal muscle (SM) and extrac

In vivo detection of HIV-1 antisense transcripts in untreated and ART-treated individuals.

Natural antisense transcripts are expressed in eukaryotes, prokaryotes, and viruses and can possess regulatory functions at the transcriptional and/or post-transcriptional levels. In vitro studies have shown that HIV-1 antisense transcripts (AST) promote viral latency through epigenetic silencing of the proviral 5' long terminal repeat (LTR). However, expression of HIV-1 AST in vivo have not been convincingly demonstrated. Here, we used single RNA template amplification, detection, and sequencing to demonstrate expression of AST in unstimulated PBMC collected from people with HIV-1 (PWH). We f

Integrated omics profiling of individual variations in intestinal damage to the soybean allergen in piglets.

INTRODUCTION: A small number of soybean allergens [including Glycinin (11S) and β-Conglycinin (7S)] in the commercially available corn-soybean meal diet can still cause allergy in some weaned piglets, which may be the result of the interaction of genetic, and nutrition, but the specific mechanism is still unclear. METHODS: In this study, 20 allergic piglets and 20 non-allergic piglets were selected from 92 weaned piglets by skin sensitization tests, which were used to examine the whole sequence genome. The indicators related to humoral and cellular immunity, transcriptomics, and metabolomics

Individual Coordinative Structures During Single-Legged Stance Present Lateral Asymmetry.

This work investigated the effect of lateral preference in angular coordination during single-legged postural control, considering individual coordination patterns rather than assuming collective motion modes. Nineteen healthy individuals (8 women), aged between 18-40 years, with a consistent preference for the right side, performed a single-legged stance, while a Vicon System captured data of 39 markers to calculate 30 angles. Two trials of 60s for each lower limb were performed. Principal Component Analysis (PCA) was performed individually for each participant´s data. The involvement of a gi

A database of upper limb surface electromyogram signals from demographically diverse individuals.

Upper limb based neuromuscular interfaces aim to provide a seamless way for humans to interact with technology. Among noninvasive interfaces, surface electromyogram (EMG) signals hold significant promise. However, their sensitivity to physiological and anatomical factors remains poorly understood, raising questions about how these factors influence gesture decoding across individuals or groups. To facilitate the study of signal distribution shifts across individuals or groups of individuals, we present a dataset of upper limb EMG signals and physiological measures from 91 demographically diver

Single-cell transcriptomic reveals network topology changes of cancer at the individual level.

Network biology facilitates a better understanding of complex diseases. Single-sample networks retain individual information and have the potential to distinguish disease status. Previous studies mainly used bulk RNA sequencing data to construct single-sample networks, but different cell types in the tissue microenvironment perform significantly different functions. In this study, we investigated whether network topology features of cell-type-specific networks varied in different pathological states at the individual level. Protein-protein interaction network (PPI) and co-expression network of

Individualized cerebellar damage predicts the presence of behavioral disorders in children with brainstem tumors.

BACKGROUND: Brainstem tumors often cause intractable neurobehavioral issues, which can be a challenge for patients and surgeons. Research on cerebellar changes in these patients is limited, despite symptoms similar to cerebellar injuries. This study aims to investigate cerebellar damage pattern resulting from brainstem tumors and its association with behavioral disorders. METHODS: This study enrolled 147 children with brainstem tumors. A U-Net-based segmentation algorithm is used to divide their cerebellums into 26 lobules. And these lobules are then used to build a normative model for assess

Identification of human remains in case of transgender individuals in forensic anthropology: a systematic review.

During anthropological analysis of human skeletal remains, sex assessment is a key step in forensic identification of individuals. For transgender individuals, complications can arise during the estimation of biological sex, as physical characteristics may not correspond with the documented gender recorded at registration of the missing person with authorities. In this systematic review, we analyzed the current literature focusing on methods employed for state-of-the-art identification of skeletal remains of transgender and gender-diverse individuals in forensic anthropology, including the eff

Individual differences in fear memory expression engage distinct functional brain networks.

Fearful stimuli elicit a mix of active (e.g., evasion) and passive (e.g., freezing) behaviors in a wide range of species, including zebrafish (Danio rerio). However, it is not clear if individual differences in fear responses exist and, if so, what parts of the brain may underlie such differences. To probe these questions, we developed a contextual fear conditioning paradigm for zebrafish that uses conspecific alarm substance (CAS) as an unconditioned stimulus where fish associate CAS administration with a specific tank. To identify individual differences, we collected behavioral responses fro

Optimizing the mirror illusion during mirror therapy: evidence from unimpaired individuals.

INTRODUCTION: Mirror therapy has demonstrated functional benefits for patients recovering from hemiparetic stroke, with its effectiveness primarily attributed to the induction of a compelling visual illusion that engages sensorimotor networks. Although previous research has identified various intervention parameters influencing therapeutic outcomes, a comprehensive understanding of their effects on the illusory experience remains limited. This study investigated how four critical parameters-mirror size (large vs. small), object manipulation (present vs. absent), task complexity (simple vs. com

Individualization of Testicular Cells for Single-Cell Subcellular Immunocytochemistry and Cell Culture Applications.

The use of tissue dissociations to produce single-cell suspensions is foundational to a long history of research applications. More recently, increasingly sensitive and complex down-stream single-cell assays and tissue culture applications are being developed that require refined tissue-specific dissociation protocols that produce consistent, high-quality, single-cell populations. To dissociate tissues, mechanical, enzymatic, and chemical methods are optimized in a tissue-specific manor dependent largely on the histomorphological arrangement of extracellular matrix and intercellular junctions.

Genome-wide identification and analysis of recurring patterns of epigenetic variation across individuals.

Epigenetic mapping studies across individuals have identified many positions of epigenetic variation across the human genome. However the relationships between these positions, and in particular global patterns that recur in many regions of the genome, remains understudied. In this study, we use a stacked chromatin state model to systematically learn global patterns of epigenetic variation across individuals and annotate the human genome based on them. We apply this framework to histone modification data across individuals in lymphoblastoid cell lines and across autism spectrum disorder cases
