Ribosomal protein L11 mutation in zebrafish leads to haematopoietic and metabolic defects.

Mutations in ribosomal proteins are associated with a congenital syndrome, Diamond-Blackfan anaemia (DBA), manifested by red blood cell aplasia, developmental abnormalities and increased risk of malignancy. Recent studies suggest the involvement of p53 activation in DBA. However, which pathways are involved and how they contribute to the DBA phenotype remains unknown. Here we show that a zebrafish mutant for the rpl11 gene had defects both in the development of haematopoietic stem cells (HSCs) and maintenance of erythroid cells. The molecular signature of the mutant included upregulation of p5

[Changes in the activity of amyloid-degrading metallopeptidases leads to disruption of memory in rats].

In old male Wistar rats (older than 12 months), or adult males (3-4 months) subjected to prenatal hypoxia (7% 02, 3 h, E14), a disruption of short-term memory was observed. The prenatal hypoxia also led to a decrease in the brain cortex expression of metallopeptidases neprilysin (NEP) and endothelin-converting enzyme (ECE-1) which regulate some neuropeptides and are the main beta-amyloid-degrading enzymes. Moreover, a significant decrease (by 2.7 times) in NEP activity in the sensorimotor cortex of old and adult rats subjected to prenatal hypoxia (by 1.7 times) was observed. To confirm possibl

Beta-amyloid causes depletion of synaptic vesicles leading to neurotransmission failure.

Alzheimer disease is a progressive neurodegenerative brain disorder that leads to major debilitating cognitive deficits. It is believed that the alterations capable of causing brain circuitry dysfunctions have a slow onset and that the full blown disease may take several years to develop. Therefore, it is important to understand the early, asymptomatic, and possible reversible states of the disease with the aim of proposing preventive and disease-modifying therapeutic strategies. It is largely unknown how amyloid beta-peptide (A beta), a principal agent in Alzheimer disease, affects synapses i…

Treatment-resistant meningitis leading to the diagnosis of Currarino syndrome: a case report.

A 20-day-old female infant developed fever. Sepsis evaluation revealed cerebrospinal fluid (CSF) pleocytosis and a modestly decreased CSF glucose. After 48 hours of broad-spectrum antibiotics, the infant remained febrile. Repeat CSF analysis showed increased pleocytosis and a very low glucose value. Subsequently, a rectothecal fistula and sacral abnormalities were found, leading to the diagnosis of Currarino syndrome. Parameningeal foci should be considered in treatment-resistant meningitis.

Dual atrioventricular-nodal physiology, elicited by pacing and leading to a reversible cardiomyopathy.

Atrioventricular nodal re-entry tachycardia is the most common form of regular paroxysmal tachycardia in the adult population. This tachycar … ed characteristic dual atrioventricular-nodal physiology that was suppressed by amiodarone therapy, leading to a reversal of the cardiomyopathy. We discuss the mechanisms that surround such phenomena.

Fetal asphyxia leads to the loss of striatal presynaptic boutons in adult rats.

Fetal asphyxic insults in the brain are known to be associated with developmental and neurological problems like neuromotor disorders and cognitive deficits. Little is known, however, about the long-term consequences of fetal asphyxia contributing to the development of different neurological diseases common in the adult or the aging brain. For that reason the present study aimed to investigate the long-term effects of fetal asphyxia on synaptic organization within the adult rat brain. Fetal asphyxia was induced at embryonic day 17 by 75-min clamping of the uterine and ovarian arteries. Presyna

Laser-assisted extraction of pacemaker and defibrillator leads: the role of the cardiac surgeon.

BACKGROUND: The development of laser-assisted extraction techniques for chronically implanted pacemaker and defibrillator leads has reduced the need for open surgical removal. Reports of the mortality from laser-assisted extraction range from 1.9% to 3.4%. The purpose of this study was to determine the rate of major cardiovascular injury and emphasize the need for cardiothoracic surgical participation in this procedure. METHODS: A retrospective cohort study was performed of 112 consecutive laser-assisted lead extractions at a single university medical center during a 6-year period. Patient an…

Maternal vitamin D deficiency leads to cardiac hypertrophy in rat offspring.

The aim of this study was to determine the effect of vitamin D deficiency from conception until 4 weeks of age on the development of the hea … 4 weeks of age. Our findings suggest that exposure to vitamin D deficiency in utero and early life leads to delayed maturation and subsequent enhanced growth (proliferation and hypertrophy) of cardiomyocytes in the LV. This may lead to altered cardiac function later in life.

Chronic fungal sinusitis leading to disastrous cerebral aspergillosis: a case report.

Cerebral angioinvasion is a fatal complication of disseminated aspergillosis and has been rarely described in diabetic population in the absence of ketoacidosis. We present a case of invasive fungal sinusitis in a 79 year old diabetic man who presented with chronic frontal headaches. Despite appropriate medical and surgical treatment, the disease progressed and the patient eventually succumbed to a fatal ruptured mycotic aneurysm. We emphasize that clinicians should consider this in the differential diagnoses of all diabetics who present with chronic sinusitis, as early diagnosis could be the

Transient TWEAK overexpression leads to a general salivary epithelial cell proliferation.

OBJECTIVES: Tumor necrosis factor-like weak inducer of apoptosis (TWEAK) is a multifunctional cytokine that has pro-apoptotic, pro-angiogenic and pro-inflammatory effects. In liver, TWEAK leads to proliferation of progenitor oval cells, but not of mature hepatocytes. This study evaluated the hypothesis that TWEAK overexpression in salivary glands would lead to the proliferation of a salivary progenitor cell. METHODS: A recombinant, serotype 5 adenoviral vector encoding human TWEAK, AdhTWEAK, was constructed, initially tested in vitro, and then administered to male Balb/c mice via cannulation …

Ectopic varices leading to occult haemorrhage in portal hypertension: a surgical quandary.

When portal venous pressure exceeds 12 mmHg, portosystemic collaterals develop. Common sites include the oesophago-gastric junction, bare area of the liver, anterior abdominal wall, and the anorectal junction. Variceal haemorrhage from the oesophago-gastric junction is voluminous, overt, and usually easily accessible, however occult bleeding from ectopic varices presents the ultimate challenge to those managing gastrointestinal haemorrhage. In this article we describe two cases of ectopic varices with an outline of the management strategy for such patients.

Neurofibromatosis 2 leads to higher incidence of strabismological and neuro-ophthalmological disorders.

PURPOSE: Ophthalmic features of neurofibromatosis 2 (NF2) include juvenile cataract, retinal hamartomas and tumours of the cranial nerves. We hypothesize that these tumours lead to strabismological and neuro-ophthalmological symptoms, including palsies of cranial nerves III, IV and VI, nystagmus and gaze palsies. METHODS: We carried out a retrospective review of 73 patients with known genotype. They underwent ophthalmic, neuro-ophthalmological and strabismological examination. Statistical analysis was performed by calculating odds ratios and their 95% confidence intervals. RESULTS: Mean best…

A maternal epimutation of GNAS leads to Albright osteodystrophy and parathyroid hormone resistance.

CONTEXT: Pseudohypoparathyroidism (PHP) type Ia is a rare maternally transmitted disease due to maternal loss-of-function mutations of GNAS, the gene encoding Galphas, the alpha-stimulatory subunit of the G protein. Affected individuals display hormonal resistance (mainly PTH and TSH resistance) and Albright hereditary osteodystrophy. PHP type Ib (PHP-Ib), usually defined by isolated renal resistance to PTH and sometimes mild TSH resistance, is due to a maternal loss of GNAS exon A/B methylation, leading to decreased Galphas expression in specific tissues. OBJECTIVE AND RESULTS: We report a g…

Exposure of the mouse perinatal testis to radiation leads to hypospermia at sexual maturity.

The first round of mouse spermatogenesis begins from 3 to 4 days after birth through differentiation of gonocytes into spermatogonial-stem cells and type A spermatogonia. Consequently, this step of differentiation may determine generation of the original population of stem cells and the fertility potential of the adult mouse. We aimed to determine the effect of perinatal exposure to ionizing radiation on the testis at the end of the first wave of spermatogenesis and at sexual maturity. Our results show that, radiation sensitivity of the testis substantially decreases from late foetal life to t

Spontaneous hemorrhagic angiomyolipoma present with massive hematuria leading to urgent nephrectomy.

Managing acute abdomen is a challenge for physicians in the emergency department. An immediate surgery is sometimes required for the hemorrhagic uropathy of renal angiomyolipoma. We present a case with left flank pain and findings of hematuria and pyuria. Abdominal and pelvic computed tomographic scans showed a 3 x 5-cm, fat-containing tumor at the left lower pole of the kidney and a bleeding and perirenal hematoma in the left perirenal space. An urgent left nephrectomy was lifesaving for the postoperative finding of renal angiomyolipoma. It is important to consider the possibility of the pres

Severe shoulder dystocia leading to neonatal injury: a case control study.

BACKGROUND AND OBJECTIVES: Severe shoulder dystocia (SD) is associated with neonatal brachial plexus injuries and skeletal fractures, with the former being the commonest cause for litigation related to birth trauma. The aim of this case-control study was to evaluate risk factors for birth injuries in cases presenting with SD. METHODS: Between January 2000 and December 2006, 22 babies who sustained brachial nerve paralysis or skeletal fractures following severe SD and requiring admission to Special Care Baby Unit (SCBU) were identified. The control group (n = 22) comprised the next infant deli

Studies leading to potent, dual inhibitors of Bcl-2 and Bcl-xL.

Overexpression of the antiapototic proteins Bcl-2 and Bcl-xL provides a common mechanism through which cancer cells gain a survival advantage and become resistant to conventional chemotherapy. Inhibition of these prosurvival proteins is an attractive strategy for cancer therapy. We recently described the discovery of a selective Bcl-xL antagonist that potentiates the antitumor activity of chemotherapy and radiation. Here we describe the use of structure-guided design to exploit a deep hydrophobic binding pocket on the surface of these proteins to develop the first dual, subnanomolar inhibitors
