Recent evidence has suggested that peripheral blood suppressor cell populations can modulate immune responsiveness. Absence of suppressor cell activity has been noted in diseases of presumed autoimmune basis. We determined inducible suppressor cell activity in patients with primary biliary cirrhosis (PBC) as compared to age and sex-matched controls. There was a significant loss of suppressor cell activity for mitogen response in patients with PBC (-4.5 +/- 8.0%) versus controls (43.7 +/- 7.8%). Peripheral blood lymphocytes transformed significantly to denatured type I collagen in 22 of 38 patients with rheumatoid arthritis (58%), 1 of 9 patients with Reiter's disease (P less than 0.05), and 9 of 32 controls (28%) (P less than 0.01). Determination of HLA antigens revealed significant differences between 24 rheumatoid arthritis (RA) patients and a control population for HLA-DRw4 (75% versus 23%). Of 30 bacterial species tested 18 stimulated DNA synthesis in human blood lymphocytes. The maximum response was after 3-4 days of culture suggesting a mitogenic effect. This was confirmed by the induction of polyclonal antibody production shown by a plaque assay. Cultures of human lymphocytes were incubated with 8-methoxypsoralen (8-MOP) and irradiated with long wave ultraviolet light (UVA). Subsequently the mitogenic response to PHA was determined by 3H-thymidine incorporation. In the present study, 15 children with non-Hodgkin lymphoma have been immunologically evaluated by the following parameters on peripheral blood (PB) lymphocytes: phytohemagglutinin responsiveness (PHA-r); non-immune rosette formation with sheep red blood cells (RF-L) as T-cell marker; presence of surface immunoglobulins (sIg-L) as B-cell marker; serum immunoglobulins levels (IgA, IgM, IgG).
