Glucose selectively drives a rapid oxidative burst and immunometabolic reprogramming in human neutrophils during Mycobacterium tuberculosis infection.

Neutrophil functions have been linked to tuberculosis (TB)-associated tissue damage; however, the mechanisms driving immunopathology in the human TB lung remain poorly understood, due partly to the scarcity of human tissue for study. Here, we examine the metabolic and bioenergetic reprogramming of human neutrophils in response to Mycobacterium tuberculosis ( Mtb ) infection. In human necrotic TB granulomas, levels of NETosis-associated proteins are increased and co-localize with GLUT3, linking nutrient uptake to tissue damage. In vitro , Mtb elicits an immediate, contact-dependent oxidative bu…

Bedaquiline Binding at the Leading Site of Mycobacterium tuberculosis ATP Synthase Induces Distinct Structural and Dynamic Changes.

ATP synthase (ATPase) is a crucial molecular motor in Mycobacterium tuberculosis (Mtb), essential for energy production and oxygen-dependent pathogenesis. The enzyme consists of two distinct rotors: a membrane-embedded Fₒ unit and a cytosolic catalytic F1 unit, along with a stator, a central stalk, and a heterodimeric peripheral stalk (PS). As the F0 region hosts critical drug-binding pockets, it has gained significant interest. This study focuses on local structural dynamics at the leading site in the presence of bedaquiline (BDQ). All-atom molecular dynamics simulations were performed using …

National genomic survey of drug-resistance and multi-jurisdictional clusters of Mycobacterium tuberculosis in Australia 2015-2023.

BACKGROUND: Tuberculosis is a global public health priority. In Australia, genomic analysis of Mycobacterium tuberculosis (Mtb) occurs at state or territory level, thus multijurisdictional genomics of Mtb remains poorly understood. We present the first national assessment of Mtb using nine years of genomic data from all Australian jurisdictions, as a part of the Australian Pathogen Genomics program. METHODS: Our national Mtb sequence dataset spanned 2015-2023 and represented culture-confirmed cases. AusTrakka was optimised to allow secure, national contribution of Mtb genomes and metadata. Dr…

Construction of Recombinant Mycobacterium smegmatis Expressing the Pro-Apoptotic Protein BIK and Its Impact on Macrophage Apoptosis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis, remains a major global health threat. A key virulence mechanism involves the pathogen's ability to inhibit apoptosis in infected macrophages, thereby facilitating immune evasion and persistent infection. The pro-apoptotic protein Bcl-2-interacting killer (BIK) represents a critical mediator of macrophage apoptosis. In this study, we engineered a recombinant Mycobacterium smegmatis strain (rMs-BIK) designed to secrete BIK via fusion with the Ag85B signal peptide (ASP). Secretion and expression of the BIK protein were confirmed by Western …

Inhibition of type II NADH dehydrogenase in Mycobacterium tuberculosis reveals collateral vulnerability in the electron transport chain.

To sustain the anti-tuberculosis drug development pipeline, novel antibiotics must be developed both as standalone agents and as components of future combination regimens. To this end, tricyclic-spirolactams (TriSLa)-based inhibitors of mycobacterial type II NADH dehydrogenase (Ndh-2) represent a promising novel class of antibiotics, though activity is carbon source dependent. Initial studies established an enhanced understanding of the impact of TriSLa on mycobacteria using metabolomics and transcriptomics. Then, CRISPRi chemical-genetics and extensive in vitro TriSLa-combination studies show

Radiological factors associated with the recurrence of Mycobacterium avium complex pulmonary disease: a multicentre retrospective cohort study.

Mycobacterium avium complex pulmonary disease (MAC-PD) frequently recurs after treatment but predictors of recurrence remain unclear. We retrospectively analysed patients with successfully treated MAC-PD from two tertiary centres. Chest CT findings-including bronchiectasis, cellular bronchiolitis, cavity, nodule and consolidation scores-were assessed before and after treatment. Multivariable competing risk regression analyses were performed to identify factors associated with recurrence. Among 538 patients, 216 (40.1%) experienced recurrence. A higher post-treatment cellular bronchiolitis scor…

The Flexible Peripheral Segments Modulate the Catalytic Activity of the FAD-Containing Monooxygenase EthA from Mycobacterium tuberculosis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a major global health concern, particularly due to the emergence of drug-resistant strains. FAD-containing monooxygenase EthA activates the antitubercular prodrug ethionamide (ETH) in Mtb. However, the structural and functional mechanisms of Mtb EthA are not fully understood. Here, we report an AlphaFold2-predicted structural model of Mtb EthA, validated using Ramachandran analysis (91.7% residues in favored regions) and a ProSA Z-score of -10.96. Based on the results of molecular docking, site-directed mutagenesis was cond…

Long-Segment Ventral Spinal Epidural Abscesses Caused by Mycobacterium Tuberculosis: A Report of 2 Cases and Review.

» Abscesses caused by spinal tuberculosis typically occur in the anterior and lateral regions of the vertebral bodies, often extending to the paraspinal muscles as cold abscesses. Although intraspinal abscesses due to spinal tuberculosis are very common, cases with longitudinally extensive abscesses in the ventral epidural space of the spinal cord are not well-documented. The disease is characterized by insidious onset, rapid progression, and a high risk of irreversible neurological damage, highlighting the necessity for early diagnosis and prompt treatment. » This review presents 2 rare cases

Ongoing evolution of PE/PPE genes in Mycobacterium tuberculosis associated with drug resistance and host immune response.

The Proline-Glutamate/Proline-Proline-Glutamate (PE/PPE) gene family comprises approximately 10% of the Mycobacterium tuberculosis (Mtb) genome and is characterized by GC-rich, highly repetitive sequences. As a result, these genes are usually excluded from short-read-based whole-genome sequencing analyses, leaving their sequence diversity and evolutionary dynamics poorly characterized. Recently, a genome masking approach demonstrated that roughly 54% of PE/PPE sequences are recoverable from short-read data, providing an opportunity to examine the evolution of this gene family at a population l…

miR-1236-3p targets Toll-like receptor 4 to suppress the anti-Mycobacterium tuberculosis activity of macrophage.

Mycoba cterium tuberculosis (Mtb) modulates host innate immunity via Toll-like receptor 4 (TLR4), associated with the susceptibility to Mtb. Bioinformatics predicted miR-1236-3p could be a potential target for the 3'-UTR of the TLR4 gene. However, the clinical significance and underlying mechanisms remain unclear. To validate this, we analyzed miR-1236-3p levels in 81 subjects and observed that both active tuberculosis (ATB) and latent tuberculosis infection (LTBI) patients exhibited elevated miR-1236-3p levels compared to healthy control (HC) subjects. In vitro dual-luciferase reporter assays

Design and synthesis of isoxazole-functionalized benzene sulphonamides as novel inhibitors of Mycobacterium tuberculosis β-carbonic anhydrases.

The escalating prevalence of multidrug-resistant tuberculosis (MDR-TB) underscores the urgent need for new classes of antitubercular agents targeting novel pathways. Carbonic anhydrase, a ubiquitous metalloenzyme, catalyses the reversible hydration of carbon dioxide in the CO2 + H2O ⇋ HCO3 - + H+ reaction. Suppressing this enzymatic activity has recently been identified as a new pathway for the treatment of Mycobacterium tuberculosis. To address this, a series of isoxazole-sulphonamides was rationally designed, incorporating an isoxazole pharmacophore as the aromatic tail, amide as a linker, a…

Long short-term memory-based deep learning model for the discovery of antimicrobial peptides targeting Mycobacterium tuberculosis.

MOTIVATION: Tuberculosis, caused by Mycobacterium tuberculosis, remains a global health challenge driven by rising antibiotic resistance. Antimicrobial peptides offer a promising alternative due to membrane-disruptive activity and low resistance potential, yet the scarcity of TB-specific AMP data constrains targeted development. We present a reproducible deep learning protocol that integrates long short-term memory networks with transfer learning to classify and generate TB-active peptides. RESULTS: Classifiers were pretrained on a large corpus of general AMPs and fine-tuned on curated TB-spe…
