Enteric nervous system in the European beaver (Castor fiber) pylorus - an immunohistochemical study.

European beaver (Castor fiber), the largest rodent species inhabiting a wide area of Eurasia, feeds mainly on dry parts of plants, bark or wood. Such kind of nourishment needs to be properly digested in each part of the gastrointestinal tract. The time of stomach digestion, which directly influences all the following steps of the digestion process, is precisely controlled by the pylorus and its innervation. However, virtually no data is available on the organization of the enteric nervous system in most of the wild animal species, including beavers. On the other hand, a pecu- liar diet consume

Continuous Vagal Neuromonitoring Using the Laryngeal Adductor Reflex: Can Preincision Dyssynchrony Predict Intraoperative Nerve Behavior?

OBJECTIVE: The laryngeal adductor reflex (LAR) is an airway-protective response triggered by sensory laryngeal receptors and resulting in bilateral vocal fold adduction. The normal morphology of the early R1 response resembles that of the compound muscle action potential (CMAP). However, in a small subset of patients, the morphology is dyssynchronous with multiple peaks. This study investigates whether preoperative LAR dyssynchrony predicts intraoperative nerve behavior during thyroid surgeries. STUDY DESIGN: Retrospective case-control study. SETTING: US academic health center. SUBJECTS AND

Differential Expression of Neuroinflammatory mRNAs in the Rat Sciatic Nerve Following Chronic Constriction Injury and Pain-Relieving Nanoemulsion NSAID Delivery to Infiltrating Macrophages.

The neuroinflammatory response to peripheral nerve injury is associated with chronic pain and significant changes in the molecular expressio … apy to explore the impact on mRNA expression changes in both pain and pain-relieved states. Sciatic nerve tissue recovered from CCI animals is used to evaluate the mRNA expression profiles utilizing quantitative PCR. We observe mRNA changes consistent with the reduced recruitment of macrophages evident by a reduction in chemokine and cytokine expression. Furth …

Granulocyte-macrophage colony-stimulating factor improves mouse peripheral nerve regeneration following sciatic nerve crush.

Peripheral nerve injuries severely impair patients' quality of life as full recovery is seldom achieved. Upon axonal disruption, the distal nerve stump undergoes fragmentation, and myelin breaks down; the subsequent regeneration progression is dependent on cell debris removal. In addition to tissue clearance, macrophages release angiogenic and neurotrophic factors that contribute to axon growth. Based on the importance of macrophages for nerve regeneration, especially during the initial response to injury, we treated mice with granulocyte-macrophage colony-stimulating factor (GM-CSF) at variou

Soluble P-selectin promotes retinal ganglion cell survival through activation of Nrf2 signaling after ischemia injury.

Retinal ischemic injuries play an important role in the pathogenesis of several eye disorders. Inflammation and oxidative stress are key pla … ction, maintained retinal nerve fiber layer thickness, and reduced macrophage infiltration in optic nerve tissue in the chronic phase (day 28). Increased NAD(P)H quinone dehydrogenase 1 (NQO1) and heme oxygenase 1(HO-1) expression levels, along with increased transcription factor Nrf2, suggesting an antioxidant role of P-sel-IgG via the Nrf2 signaling pathway. …

The importance of nerve microenvironment for schwannoma development.

Schwannomas are predominantly benign nerve sheath neoplasms caused by Nf2 gene inactivation. Presently, treatment options are mainly limited to surgical tumor resection due to the lack of effective pharmacological drugs. Although the mechanistic understanding of Nf2 gene function has advanced, it has so far been primarily restricted to Schwann cell-intrinsic events. Extracellular cues determining Schwann cell behavior with regard to schwannoma development remain unknown. Here we show pro-tumourigenic microenvironmental effects on Schwann cells where an altered axonal microenvironment in cooper

The medial sural artery perforator flap: a versatile donor site for hand reconstruction.

BACKGROUND: The challenge of modern hand reconstruction goes beyond simple coverage. Thanks to the advances of microsurgery, there are ever-improving standards of functional and esthetic outcomes in hand reconstruction. The versatile donor site of the medial sural artery perforator flap can fulfill this purpose. MATERIALS: Between June 2006 and October 2008, we used free medial sural artery perforator flaps for hand reconstruction in 14 cases. The sites of reconstruction included digits (n=7), dorsal hand (n=3), palmar hand (n=2), and wrist (n=2). Associated tendon and nerve defects were foun

Fascial feeder and perforator-based V-Y advancement flaps in the reconstruction of lower limb defects.

The principle of the V-Y advancement flap has been used since its first description by Blasius (1848) for reconstruction of smaller defects. We wish to describe V-Y advancement flaps, the design of which includes distinct perforator or fascial feeder vessels, which may originate from periosteum, muscle, cutaneous nerve or from large tendon sheaths. These flaps are planned in an oblique manner when there is a defect over the anterior, antero-lateral or antero-medial aspect, and in a vertical manner when there is a defect over the posterior aspect of the leg. The main advantage of this design is

The development of the optic nerve in rodents.

In mammals, normal visual function depends upon both the retinotopic organization of visual nuclei and their interconnections. We have investigated in rodents some developmental mechanisms contributing to this organization. On embryonic day 14, in the rat, retinal ganglion cells first project axons through glial channels on the retinal surface before reaching the optic stalk. We suggest that the sequence in which axons enter the stalk (central before peripheral) and their prominent fasciculation impose some retinotopic order amongst the emerging optic nerve fibres. At birth (embryonic day 21)

Structural reconstruction of spinal cord white matter using a biomimetic zoned fiber-hydrogel scaffold.

Following spinal cord injury (SCI), the zonal architecture of gray and white matter remains challenging to reconstruct. The present study explores a strategy for reconstructing this zonal structure via the integration of oriented fibers with hydrogels. The SFs were rationally arranged to conform to the morphological features of the white matter. The incorporation of the hydrogel, in turn, filled the gray matter regions and immobilized the spatial distribution of the fibers. This gray/white matter zoned scaffold (GWZS) effectively mimics the inherent characteristics of native spinal cord gray a

Lactobacillus amylovorus KU4 inhibits adipocyte senescence in aged mice through necdin regulation of p53 activity.

Previously, we reported that Lactobacillus amylovorus KU4 (LKU4) ameliorates diet-induced metabolic disorders by regulating adipose tissue (AT) physiology. Since metabolic disorders and age-related pathological conditions mutually exacerbate each other, this study hypothesizes that LKU4 may protect against adipose senescence during aging. Thus, this study demonstrates that LKU4 administration suppresses age-related metabolic dysfunction and aging phenotypes in AT of 24-month-old mice. Furthermore, LKU4 suppressed the expression of senescence marker genes, including p53, in the AT of these mice

Huntington's disease: an update of therapeutic strategies.

Huntington's disease (HD) is an autosomal dominant triplet repeat genetic disease, which results in progressive neuronal degeneration in the neostriatum and neocortex, and associated functional impairments in motor, cognitive, and psychiatric domains. Although the genetic mutation caused by abnormal CAG expansion within the htt gene on chromosome 4p16.3 is identified, the mechanism by which this leads to neuronal cell death and the question of why striatal neurones are targeted both remain unknown. Patients manifest a typical phenotype of sporadic, rapid, involuntary control of limb movement,

Differential localization of alpha-, beta- and gamma-synucleins in the rat CNS.

Alpha-synuclein is a presynaptic protein that normally participates in the homeostasis of synaptic vesicles. Missense mutations in its gene cause the protein to participate actively in the development of heritable forms of Parkinson's disease. Moreover, its metabolism is perturbed in all cases of Parkinson's disease where alpha-synuclein accumulates in a filamentous form in the Lewy body nerve cell lesion. Lewy bodies also develop in other common neurodegenerative disorders, like dementia with Lewy bodies and Lewy body variant of Alzheimer's disease. In the present study, we have studied the d

Prominent perikaryal expression of alpha- and beta-synuclein in neurons of dorsal root ganglion and in medullary neurons.

Synucleins (syns) are a family of small, highly conserved proteins expressed predominantly in neurons. Although the normal function of syns is unknown, alpha-syn plays a pivotal role in several neurodegenerative diseases. The expression patterns of syns have been described in several studies, but much of this information was obtained before the cloning of all four members of this family of proteins and previous studies were limited to the analysis of single species. Here, we used antibodies specific for alpha-, beta-, and gamma-syn to study the patterns of expression in human, mouse, and rat n

Survival-fixation of the cochlea: a technique for following time-dependent degeneration and repair in noise-exposed chinchillas.

To minimize problems with data interpretation due to interanimal variation in susceptibility to noise, we developed a survival-fixation paradigm which involves fixing one cochlea of an experimental chinchilla at one post-exposure time and fixing the second cochlea as much as 14-24 days later. This paradigm is analytically effective because there is a high correlation in the magnitude and pattern of damage in the left and right cochleas of binaurally exposed animals. Thus, each experimental animal provides two snapshots in the degeneration and repair continua in which it can be certain that bot
