Striatal dopamine release and biphasic pattern of locomotor and motor activity under gas narcosis.

Inert gas narcosis is a neurological syndrome appearing when humans or animals are exposed to hyperbaric inert gases (nitrogen, argon) composed by motor and cognitive impairments. Inert gas narcosis induces a decrease of the dopamine release at the striatum level, structure involved in the regulation of the extrapyramidal motricity. We have investigated, in freely moving rats exposed to different narcotic conditions, the relationship between the locomotor and motor activity and the striatal dopamine release, using respectively a computerized device that enables a quantitative analysis of this

Dissociable processing of temporal structure in repetitive eye-hand movements in Parkinson's disease.

Movement takes place in multiple dimensions, including the temporal dimension, which itself may be made up of dissociable aspects. From this perspective, the present research tests three hypotheses: first, that the generation of regular, repetitive movement frequency is neurophysiologically dissociable from the generation of appropriate phase relations, or latencies, of such movements with respect to external stimuli; second, that the frontostriatal system is critically involved in the control of latency and not frequency and third that while the control of latency is closely linked to the eff

Response perseveration in adolescent boys with stable and unstable histories of physical aggression: the role of underlying processes.

BACKGROUND: It was unclear whether response perseveration and underlying processes, often related to antisocial externalizing disorders, were also related to histories of physical aggression. METHOD: Boys of age 13 years were selected on the basis of childhood histories of physical aggression: stable, unstable, and non-aggressive. Performance on a Card Playing Task provided a perseveration index. RESULTS: Physical aggression, regardless of history, predicted perseveration in adolescence. However, qualitative differences revealed that Neuroticism increased the risk for perseveration only in t

Follow-up of children with language delay and features of autism from preschool years to middle childhood.

Eighteen children (13 males, five females) who had severe developmental language delay/disorder and some features of autism (although insufficient in severity and combination to meet ICD-10 diagnostic criteria for childhood autism) at preschool age (Time 1; mean age 4 years 4 months) were followed up 4 years later (Time 2; mean age 8 years 7 months). At the initial assessment the diagnostic dilemma was how much the social communication impairments and behavioural problems were secondary to the language problem and how much they constituted a genuine case of a pervasive developmental disorder.

Current directions in research on autism.

One of the most active areas of current research in the field of developmental disorders is autism. Since the NIH State of the Science conference, held in 1995 (Bristol et al. [1996] J. Autism Dev. Disorders 26:121-154), funding opportunities for comprehensive research programs addressing genetic, neurobiological, and behavioral aspects of this complex disorder have grown exponentially. Although we are far from having a complete understanding of the causes and deficits that define autism, significant progress has been made over the past few years. In this review, we summarize recent developmen

Comparison of two intracranial self-stimulation (ICSS) paradigms in C57BL/6 mice: head-dipping and place-learning.

A variety of intracranial self-stimulation (ICSS) paradigms have been utilized for investigations of reward. Among them, nose-poking and spatial-preference paradigms are known to be relatively more resistant to the effects of drug-induced motor-deficits in rat studies, although these two ICSS paradigms have not been directly compared in previous studies. In the present study, head-dipping and place-learning (forms of nose-poking and spatial-preference tasks, respectively) paradigms with lateral hypothalamus stimulation were systematically analyzed using C57BL/6 mice in the presence and absence

Effect of muscarinic receptor agonists on animal models of psychosis.

The purpose of this study was to determine the effects of muscarinic receptor agonists on models of psychosis in rats. A battery of three tests, catalepsy (haloperidol-induced) augmentation, amphetamine-induced stereotypy antagonism and conditioned avoidance response were used. Both muscarinic receptor agonists physostigmine and pilocarpine produced similar results. They dose-dependently augmented the haloperidol-induced catalepsy, inhibited amphetamine-induced stereotypy behavior and inhibited the conditioned avoidance response. Results demonstrate that muscarinic receptor agonists have actio

High-frequency auditory feedback is not required for adult song maintenance in Bengalese finches.

Male Bengalese finches do not normally change their vocal patterns in adulthood; song is stereotyped and stable over time. Adult song maintenance requires auditory feedback. If adults are deafened, song will degrade within 1 week. We tested whether feedback of all sound frequencies is required for song maintenance. The avian basilar papilla is tonotopically organized; hair cells in the basal region encode high frequencies, and low frequencies are encoded in progressively apical regions. We restricted the spectral range of feedback available to a bird by killing either auditory hair cells encod

Behavioral interactions produced by co-administration of 7-OH-DPAT with cocaine or apomorphine in the rat.

Previous research from our laboratory suggests that low doses (<0.1 mg/kg) of the dopamine (DA) D3-preferring agonist 7-hydroxy-N,N-di-n-propyl-2-aminotetralin (7-OH-DPAT) attenuate conditioned place preference (CPP) produced by the indirect DA agonist d-amphetamine, but enhance d-amphetamine-induced stereotypic behaviors. This study further examined the effects of 7-OH-DPAT on behaviors produced by the indirect DA agonist, cocaine, and the non-selective direct DA agonist, apomorphine. To examine whether 7-OH-DPAT would alter cocaine and apomorphine dose-response curves for motor behaviors and

Loss of tolerance to amphetamine-induced hypophagia in rats: homeostatic readjustment vs. instrumental learning.

According to the homeostatic model, the loss of tolerance to amphetamine-induced hypophagia requires a period of unrestricted feeding in the drug-free state, which transforms the compensatory response mediating tolerance ("hyperhunger") into a functional disturbance to homeostasis. In the absence of such a disturbance, tolerance should be retained. To test this prediction, rats tolerant to amphetamine's hypophagic effect were given a 4-week tolerance retention period during which milk intakes were restricted and deprivation levels held relatively constant. During this period the rats were assi

Eating disorders and the cultural forces behind the drive for thinness: are African American women really protected?

Responding to mainstream ideals of female beauty, many women and girls view thinness as a requirement for feeling acceptable to themselves and to others. The drive to be thin can lead to problematic eating patterns, such as self-starvation, binge-eating, and purging, symptoms of the eating disorders, anorexia and bulimia nervosa. Most current literature on eating disorders and the drive for thinness focuses only on White middle-class women and girls. African American females have been largely excluded from studies, due to the assumption that the Black community's acceptance of women with fulle

Long-term effects of repeated maternal separation on three different latent inhibition paradigms.

In the present study we investigated the effect of repeated maternal separation on postnatal days 12, 14, 16, and 18 for 6 h/day on Wistar rats on three latent inhibition (LI) paradigms: two-way active avoidance, conditioned emotional response (CER), and conditioned taste aversion (CTA). In addition, hyperactivity induced by d-amphetamine and stereotypies induced by apomorphine were evaluated. In all three LI experiments, the control animals showed only marginal LI, whereas the maternally separated animals showed enhanced LI (only males in CTA). In two-way active avoidance within the nonpreexp

Activity profiles of two hyperforin-containing hypericum extracts in behavioral models.

The behavioral activity profile of a therapeutically used alcoholic hypericum extract containing hyperforin (4.5%) in rodent models was compared with that of an experimental CO2 extract devoid of hypericines but highly enriched in hyperforin (38.8%). The antidepressant activities of 50, 150 and 300 mg/ kg/day of the alcoholic extract were similar to those of 5, 15 and 30 mg/kg/day respectively of the CO2 extract. The ethanol extract in the same dose range potentiated dopaminergic behavioral responses, whereas these effects were either absent or less pronounced in the CO2 extract treated groups

Effects of intraaccumbens microinjections of quinopirole on head turning and circling movement in the rat.

This study was designed to evaluate whether nucleus accumbens dopamine D2 receptors are involved in the initiation of the movement, as distinguished from its execution. For this purpose, the effects of the quinpirole-induced increase of nucleus accumbens dopamine D2 receptor activity were observed on specific parameters of the circling behavior and of its first stage, the head-turning (HT) movement. The experiments were performed on rats with unilateral 6-hydroxydopamine (6-OHDA) lesion of the pars compacta of the substantia nigra and d-amphetamine i.p. (3 mg/kg). Bilateral intraaccumbens micr

Development of self-sustaining limbic status epilepticus by continuous ventral hippocampal stimulation followed by low dose pilocarpine in rats.

Sequential treatment of rats with low doses of lithium and pilocarpine, a high dose of pilocarpine, or continuous hippocampal stimulation [CHS] (9 epochs, 10 min each) is reported to result in status epilepticus (SE). We report a novel method to establish SE based on continuous ventral hippocampal stimulation (5 epochs) followed by low dose pilocarpine (40 mg/kg) challenge. Motor limbic seizures occured in all the control rats. The latency to spike activity was 15 +/- 1 min after pilocarpine administration. Ventral hippocampal [VHc] and cortical EEG recordings were used to monitor the protecti

Under continuous dizocilpine infusion an N-methyl-D-aspartate receptor independent form of cocaine sensitization develops in rats.

N-methyl-D-aspartate (NMDA) receptor blockade is thought to prevent the development of cocaine-induced sensitization. Moreover, when cocaine is administered daily along with dizocilpine infusion to previously sensitized rats, the extinction of sensitization occurs. We report here two sets of experiments: (1) Rats were infused with dizocilpine through a subcutaneous mini-pump (0.1 mg/kg/day) during the induction of cocaine sensitization and, after 2 or 7 days of wash-out, were challenged with: cocaine, dizocilpine plus cocaine or 3-((+/-)2-carboxypiperazin-4-yl)-propyl-L-phosphonic acid (CPP) p

Stigma associated with onchocercal skin disease among those affected near the Ofiki and Oyan Rivers in western Nigeria.

Skin diseases have been a major source of social stigma, whether they be infectious or not. The potential stigamtizing effect of skin disease associated with onchocerciasis is currently receiving attention because half of the 17 million victims of onchocerciasis in Africa live where the non-blinding form of the disease is prevalent. Some reports are available that onchocercal skin disease (OSD) is associated with social stigma including problems in finding a marriage partner. Previous studies have also implied positive effects of ivermectin treatment on OSD. Therefore a multi-country trial of

The phenomenology of ritualized and repeated behaviors in nonclinical populations in the United States.

The question of whether ritualized and repeated behaviors might be part of a person's "normal" repertoire, providing them with feelings of self-efficacy, is explored in a phenomenological study in which nonclinical subjects who engage in ritualized and repeated behaviors describe their behaviors, and reflect on their feelings before, during, and after engaging in these rituals. Results seem to support the hypothesis that engaging in these behaviors is perceived as reducing feelings of anxiety, fear, and discomfort, and increasing feelings of control and security. Subjects also report that they
