Suppressor system in murine interstitial nephritis. Analysis of tubular basement membrane (TBM)-specific suppressor T cells and their soluble factor in C57BL/6 mice using a syngeneic system.

We induced typical interstitial nephritis with high titers of anti-tubular basement membrane (TBM) autoantibody in genetically resistant C57BL/6 mice by treating them with sodium aurothiomalate (gold) and immunizing them with syngeneic TBM antigen. When gold was not used, the T-cell fraction of nylon wool adherent splenic cells showed prominent suppressive activity against the proliferative response of nonadherent cells to TBM antigen. However, this suppressive activity was remarkably decreased by the gold treatment. TBM antigen sensitized thymocytes, a thymocyte extract, and a spleen cell ext

Short lived, dividing cells mediate adoptive transfer of immunity to Trichinella spiralis in mice. II. In vivo characteristics of the cells.

The in vivo characteristics of mesenteric lymph node cells (MLNC) capable of mediating the adoptive transfer of immunity to Trichinella spiralis have been examined. Mediator cells were diverted into the peritoneal cavity of infected donor mice following the induction of a peritoneal exudate and its was shown that these were nylon-wool, non-adherent (T) cells. After density gradient separation of [125I]-UdR-labelled MLNC, the fractions that were most effective in transferring immunity were those containing a small proportion of cells but the largest proportion of incorporated activity. Improved Efficacy of Oral Immunotherapy Using Non-Digestible Oligosaccharides in a Murine Cow's Milk Allergy Model: A Potential Role for Foxp3+ Regulatory T Cells.

BACKGROUND: Oral immunotherapy (OIT) is a promising therapeutic approach to treat food allergic patients. However, there are some concerns regarding its safety and long-term efficacy. The use of non-digestible oligosaccharides might improve OIT efficacy since they are known to directly modulate intestinal epithelial and immune cells in addition to acting as prebiotics. AIM: To investigate whether a diet supplemented with plant-derived fructo-oligosaccharides (FOS) supports the efficacy of OIT in a murine cow's milk allergy model and to elucidate the potential mechanisms involved. METHODS: Af

Cell-surface nucleolin acts as a central mediator for carcinogenic, anti-carcinogenic, and disease-related ligands.

PURPOSE: Cell-surface nucleolin in human gastric cancer cell lines is a receptor for TNF-α-inducing protein (Tipα) of Helicobacter pylori. The binding complex of nucleolin and Tipα is internalized into the cells and then induces tumor progression of human gastric cancer. Surface nucleolin is also a receptor of human immunodeficiency virus-1, and the anti-HIV pseudopeptide (HB-19) showed anti-carcinogenic activity in vivo. Surface nucleolin has dual functions depending on the ligands: In order to understand the mechanisms of surface nucleolin, it is necessary to review surface nucleolin and its

Loss of the neuron-specific F-box protein FBXO41 models an ataxia-like phenotype in mice with neuronal migration defects and degeneration in the cerebellum.

The cerebellum is crucial for sensorimotor coordination. The cerebellar architecture not only requires proper development but also long-term integrity to ensure accurate functioning. Developmental defects such as impaired neuronal migration or neurodegeneration are thus detrimental to the cerebellum and can result in movement disorders including ataxias. In this study, we identify FBXO41 as a novel CNS-specific F-box protein that localizes to the centrosome and the cytoplasm of neurons and demonstrate that cytoplasmic FBXO41 promotes neuronal migration. Interestingly, deletion of the FBXO41 ge

Impairment of autophagosome-lysosome fusion in the buff mutant mice with the VPS33A(D251E) mutation.

The HOPS (homotypic fusion and protein sorting) complex functions in endocytic and autophagic pathways in both lower eukaryotes and mammalian cells through its involvement in fusion events between endosomes and lysosomes or autophagosomes and lysosomes. However, the differential molecular mechanisms underlying these fusion processes are largely unknown. Buff (bf) is a mouse mutant that carries an Asp251-to-Glu point mutation (D251E) in the VPS33A protein, a tethering protein and a core subunit of the HOPS complex. Bf mice showed impaired spontaneous locomotor activity, motor learning, and auto

Nucleotide excision repair functions in the removal of chromium-induced DNA damage in mammalian cells.

Some hexavalent chromium (Cr(VI))-containing compounds are human lung carcinogens. While ample information is available on the genetic lesions produced by Cr, surprisingly little is known regarding the cellular mechanisms involved in the removal of Cr-DNA adducts. Nucleotide excision repair (NER) is a highly versatile pathway that is responsive to a variety of DNA helix-distorting lesions. Binary Cr-DNA monoadducts do not produce a significant degree of helical distortion. However, these lesions are unstable due to the propensity of Cr(III) to form DNA adducts (DNA interstrand crosslinks, DNA-

Interactions of opioid and chemokine receptors: oligomerization of mu, kappa, and delta with CCR5 on immune cells.

Activation of opioid receptors by morphine was previously shown to specifically induce the expression of chemokine receptor CCR5, promoting simian AIDS virus entry and replication in immune cells. The present study was undertaken to determine whether these two structurally and functionally distinct G-protein-coupled receptors are in close proximity and form an oligomeric complex in the cell membrane so that the activation of one triggers the activity of the other. Both human CEM x174 and monkey lymphocytes were used in this study and gave similar results. Immunoprecipitation experiments showed

A comprehensive study on the isolation and characterization of the HeLa S3 nuclear matrix.

Different agents have been employed to extract the histones and other soluble components from isolated HeLa S3 nuclei during nuclear matrix isolation. We report that 0.2M (NH4)2SO4 is a milder extracting agent than NaCl and LIS (lithium 3,5-diiodosalicylate), on the basis of the apparent preservation of the elaborate fibrogranular network and the residual nucleolus that resemble the in situ structures in whole cells and nuclei, minimal aggregation, and sufficient solubilization of DNA and histones. The importance of intermolecular disulfide bonds, RNA and 37 degrees C stabilization on the stru

Sodium Butyrate Attenuates Doxorubicin Sardiotoxicity Through GPR41/GPR43-Sox4-PI3K/Akt Signaling.

Doxorubicin cardiotoxicity limits anthracycline chemotherapy, and mechanism-directed pharmacological protection remains limited. We investigated the cardioprotective action of sodium butyrate and its receptor-associated signaling mechanisms in doxorubicin-treated mice and H9c2 rat cardiomyoblasts. Targeted short-chain fatty acid profiling showed time- and compartment-dependent changes in endogenous butyrate after doxorubicin exposure. Oral sodium butyrate (300 mg/kg) improved left ventricular ejection fraction and fractional shortening, reduced circulating cardiac troponin I, creatine kinase-M

ALFAssay: A feed‑forward neural network for quantitative fragmentomics‑based ctDNA profiling in breast cancer.

The analysis of cell‑free DNA (cfDNA) is transforming cancer diagnostics, yet quantifying the fraction of circulating tumour DNA (ctDNA) from shallow whole‑genome sequencing (sWGS) remains challenging in tumours with low copy‑number aberration burden. We introduce ALFAssay, a feed‑forward neural network that estimates ctDNA fraction from fragmentation profiles of cfDNA in breast cancer. Using cfDNA from 896 plasma samples spanning early and metastatic HR + /HER2- and triple‑negative breast cancer, and healthy controls, (id: NCT03616886, NCT02028364, NCT03065621) we extract 204 bin‑level fragme

Fractional CO2 laser treatment of post-surgical nasal scars: a retrospective analysis on 8 patients.

Surgical scars of the nasal region following basal cell carcinoma (BCC) excision often present aesthetic and functional challenges, particularly the "trap-door" deformity related to flap reconstruction. Fractional CO₂ laser represents a promising therapeutic option to improve scar quality through collagen remodeling and dermal reorganization. This retrospective study analysed eight patients (six males and two females, mean age 71 ± 10 years) with nasal post-surgical scars after BCC excision and flap reconstruction. All patients underwent two sessions of fractional CO₂ laser in Deep Pulse or Hi

A Korean native halophyte extract attenuates the virulence of methicillin-resistant Staphylococcus aureus by inhibiting biofilm formation.

Since the COVID-19 outbreak, global attention toward infectious diseases has intensified, and many experts anticipate that the next pandemic may stem from multidrug-resistant (MDR) bacteria, often termed "Superbugs". Among them, methicillin-resistant Staphylococcus aureus (MRSA) represents one of the most widespread MDR bacterium responsible for severe nosocomial infections. As the emergence of new resistant strains accelerates due to overuse and misuse of antibiotics, development of anti-pathogenic therapeutics has gained significant interest. In this study, we explored the anti-biofilm activ

Immuno-acoustic trapping for extracellular vesicle subpopulations.

Extracellular vesicles (EVs) in biofluids are heterogeneous in origin, surface protein expression, and biomolecular cargo. Isolation of specific EV subpopulations may enable studies of biomarkers originating from specific cell phenotypes and disease states. Acoustic trapping is a rapid, automatable EV isolation technique requiring minimal sample preprocessing; however, it lacks the specificity needed for some clinical studies. Combining acoustic trapping with immunoaffinity bead-based isolation could provide access to more specific EV content and broaden the applications of acoustic trapping.
