PSMC5 Orchestrates an Immunosuppressive Niche and Metastasis in Colorectal Cancer via SMURF1-Mediated K11-Linked Ubiquitination of METTL14.

Colorectal cancer (CRC) metastasis requires coordination between tumor-intrinsic programs and the surrounding microenvironment, yet how proteasomal regulation intersects with the epitranscriptome in this process remains unclear. Here, analyses of TCGA, GEO, and an institutional cohort of 146 CRC patients identified PSMC5 upregulation as associated with metastatic progression and poor prognosis. Mechanistically, PSMC5 promoted SMURF1-dependent K11-linked ubiquitination of METTL14 at K263, leading to METTL14 destabilization, global m⁶A remodeling, and activation of EMT-associated malignant pheno

DNA methylation biomarkers associated with early gastric cardia carcinogenesis.

Gastric cardia cancer (GCC) is an aggressive malignancy with poor prognosis, underscoring the need for better characterization of molecular alterations during early gastric cardia carcinogenesis. This study aimed to identify and validate tissue-based DNA methylation markers associated with early precancerous and neoplastic lesions of the gastric cardia. We integrated genome-wide DNA methylation data (850 K array) from 69 gastric cardia samples, including normal mucosa (n = 22), intestinal metaplasia (IM, n = 32), intraepithelial neoplasia (IEN, n = 7), and GCC (n = 8). Differential methylation

Hypoxia Differentially Regulates Ferroptosis Sensitivity and Tumor Cell-Intrinsic Type I Interferon Signaling in Pancreatic Ductal Adenocarcinoma Cells.

Ferroptosis has emerged as a promising strategy to overcome resistance to conventional cancer therapies. Pancreatic ductal adenocarcinoma (PDAC) is characterized by hypoxia, therapy resistance, and an immunosuppressive microenvironment. Although hypoxia is likely to influence ferroptosis susceptibility and the associated inflammatory pathways that regulate antitumor immunity, their impact on ferroptosis sensitivity and innate immune responses remains poorly understood. In this study, we investigated the effects of hypoxia on the induction of ferroptosis and immune-related signaling in PDAC cel

CTPS1 modulates mitophagy to propel diffuse large B-cell lymphoma via reshaping CEPT1-mediated phospholipid metabolism.

Despite effective first-line regimens, some patients with diffuse large B-cell lymphoma (DLBCL) still experience relapse or resistance, emphasizing the urgent need for innovative treatment approaches. Cytidine triphosphate synthase 1 (CTPS1) is a key regulatory and rate-limiting enzyme for de novo nucleotide synthesis pathway. However, the role of CTPS1 in DLBCL and its potential therapeutic value remain unknown. We found that high levels of CTPS1 were associated with poor prognosis in patients with DLBCL. The single-cell RNA sequencing (scRNA-seq) revealed that phospholipid metabolism and mit

HNRNPU mutations redirect cell cycle control to E2F in MYC-driven lymphomas.

Heterogeneous nuclear ribonucleoprotein U (hnRNPU) is a ubiquitously expressed, pleiotropic DNA and RNA binding protein involved in RNA metabolism. Heterozygous HNRNPU nonsense mutations have been observed in a variety of B-cell lymphomas, but the functional consequence of these mutations remains largely unknown. Through a large meta-analysis of genome- and exome-wide sequencing data, we find that these mutations are more common among tumors with MYC rearrangements, including high-grade B-cell lymphoma with MYC and BCL2 and/or BCL6 rearrangements (12.1%), and Burkitt lymphoma (5.2%). Using iso

Integrative Bioinformatic Characterization of the HDAC6-Driven Cytoskeleton-Wnt Signaling Interface in Hepatocellular Carcinoma: Implications for Immune Modulation and Therapeutic Targeting.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide, characterized by marked molecular heterogeneity, late-stage diagnosis, and limited therapeutic options. Emerging evidence highlights the interplay between cytoskeletal dynamics, epigenetic regulation, and oncogenic signaling pathways in hepatocarcinogenesis. Histone deacetylase 6 (HDAC6), a key regulator of cytoplasmic protein acetylation, modulates α-tubulin stability, while CTNNB1 (β-catenin) serves as a central effector of the Wnt signaling pathway. However, the existence and functional relevance o

Spatially decoding genotype-associated epigenetic landscapes in human lymphoma FFPE tissues via epi-Patho-DBiT.

While formalin-fixed paraffin-embedded (FFPE) samples are invaluable for human non-Hodgkin B-cell lymphoma translational research, effective methods for spatial profiling of chromatin accessibility and histone modifications in these tissues remain limited. Here, we introduce epi-Patho-DBiT, a platform that combines reverse crosslinking of FFPE tissues with spatially resolved assays for transposase-accessible chromatin using sequencing (spatial-FFPE-ATAC) or cleavage under targets and tagmentation (spatial-FFPE-CUT&Tag). Using spatial-FFPE-ATAC, we map epigenetic landscapes in mucosa-associated

An oncogenic KRAS-driven secretome involving TNFα promotes niche preparation prior to pancreatic cancer onset.

BACKGROUND: Pancreatic ductal adenocarcinomas (PDACs) are highly lethal and aggressive with oncogenic KRAS being the main oncogenic driver of the disease. PDACs have been extensively profiled at advanced stages, and in advanced disease the tumor microenvironment is a major determinant that critically shapes patient outcomes. Since the molecular events occurring prior to invasive growth remain poorly understood, we aimed to investigate changes in the precancerous epithelium and its surrounding niche. METHODS: We acquired time-resolved, single-cell transcriptomic (scRNAseq), and accessible-chrom

Genotype-driven tumor ecosystems drive immune evasion and immunotherapy resistance in melanoma.

BACKGROUND: Genetic and transcriptional alterations in cancer cells shape their interactions with immune and stromal compartments, thereby influencing tumor progression, immune evasion, and response to immune checkpoint inhibitors (ICIs). Yet, how these interactions are spatially organized within melanoma and how they relate to clinical outcome remains incompletely understood. METHODS: We used spatial multi-omic profiling on melanoma tissues from patients, compared those with loss-of-function mutations in the Neurofibromin 1 (NF1) tumor suppressor gene to those with intact NF1, and integrated

GPR54 regulates non-small cell lung cancer development via dopa decarboxylase.

Non-small cell lung cancer (NSCLC), the most common type of lung cancer, is a leading cause of cancer death. G protein-coupled receptor 54 (GPR54) plays a role in cancer development by interacting with its endogenous ligand kisspeptin encoded by the KISS1 gene. However, the role of GPR54 in NSCLC development is not yet fully understood. Here, we demonstrate that GPR54 regulates NSCLC development via dopa decarboxylase (DDC). A mutant Kras-driven mouse lung cancer model revealed that adenoviral CMV-Cre-mediated Gpr54 deletion attenuated NSCLC development. Both Gpr54 deletion in mouse NSCLC tiss

Berberine Inhibited Tumor Growth Through Suppressing the WDR54/β-catenin Signal Pathway in Colorectal Cancer.

Berberine (BBR), a natural alkaloid, exhibits potent anti-tumor activity in colorectal cancer (CRC), but its mechanisms are not fully elucidated. Analysis of GEO datasets (GSE216908 and GSE184414) identified WDR54 as a key target of BBR treatment. In vitro, BBR treatment of CRC cell lines (HT-29 and SW480) markedly inhibited cell proliferation, DNA synthesis, and colony formation. Notably, BBR triggered G1-phase cell cycle arrest and promoted apoptosis, as validated by flow cytometry and molecular marker analysis. WDR54 overexpression partially rescued these anti-tumor effects, confirming its

Bestatin inhibits the development of cutaneous melanoma by inhibiting the expression of LTA4H.

OBJECTIVE: In this study, we used Mendelian randomization analysis to explore the causal relationships between drug targets and cutaneous melanoma (CM) and subsequently screened for new drug targets for CM. In addition, we verified whether targeted drugs could inhibit the development of CM in CM cells by suppressing the expression of target genes. METHODS: The eQTL data of 2,664 drug target genes (exposures) were sourced from the eQTLGen database. The GWAS summary data for CM (outcome) were obtained from the UK Biobank and FinnGen databases. The inverse-variance weighted (IVW) model was used

NRF2-mediated ferroptosis suppression defines a cancer-specific vulnerability in tumors.

NRF2 is a master regulator of redox and metabolic homeostasis that protects normal tissues from stress but is frequently hijacked by cancers to sustain survival and therapy resistance. Although NRF2 is dispensable for normal tissue function, its role in maintaining cancer cells within the native tumor microenvironment has remained undefined. Here, we uncover an essential and previously unrecognized tumor-specific dependency on NRF2. Using an inducible KrasFSF.G12D/+;Nrf2Fl/Fl;Rosa26CreERT2/CreERT2 (KNR) mouse lung cancer model, we demonstrate that NRF2 deletion alone, without pharmacologic int

Autophagy-associated exosomal microRNAs in triple-negative breast cancer: a discovery-phase study.

BACKGROUND: Triple-negative breast cancer (TNBC) is characterised by aggressive pathobiology and frequent chemoresistance, yet the mechanisms underpinning treatment failure remain incompletely understood. Chemotherapy-induced autophagy has been implicated in small extracellular vesicle (sEV) biogenesis and cargo remodelling; whether the exosomal microRNA (exomiR) landscape of TNBC is altered under autophagy-inducing chemotherapy has not been established. METHODS AND RESULTS: sEVs were isolated from MDA-MB-231 and MDA-MB-468 cells under doxorubicin-induced autophagy-inducing conditions (10 nM,

Comprehensive study of the tumor immune microenvironment and the prognostic significance of reticulocalbin-1: a pan-cancer analysis.

Reticulocalbin-1 (RCN1), an endoplasmic reticulum (ER)-related gene, demonstrates critical regulatory functions in cellular growth. RCN1 has been previously revealed to be closely associated with initiation, metastasis and chemosensitivity in some tumors. However, the prognostic significance of RCN1 and its influence on the cancer immune microenvironment remains unclear. By leveraging multi-omics data from TCGA and GEO, this study delineated the expression profile of RCN1 across various human malignancies and normal tissues. Analysis of RCN1 expression in correlation with tumor immune microenv

Spindle Assembly Checkpoint Competency Determines Sensitivity to KIF18A Inhibition in Small Cell Lung Cancer.

UNLABELLED: Small cell lung cancer (SCLC) is characterized by pervasive chromosomal instability (CIN) and remains largely refractory to targeted therapies. KIF18A, a motor protein that regulates chromosome alignment during mitosis, has emerged as a selective dependency in CIN-high tumors. Whether this dependency extends to SCLC, a prototypical CIN-high cancer, has not been established, and biomarkers predicting response to KIF18A inhibition, currently in clinical trials, are lacking. We integrated analyses of patient tumor datasets, neuroendocrine (NE) and non-NE SCLC cell lines, and functiona

Programmable ZBTB7A RNA N6-methyladenosine site-specific demethylation suppresses colorectal cancer liver metastasis.

Colorectal cancer liver metastasis (CRLM) is the primary cause of CRC-related mortality, with inevitable chemoresistance to targeted therapies and immunotherapy. N6-methyladenosine (m6A), as a crucial epigenetic regulator of gene expression and cellular physiology, involved in the pathogenesis of CRLM. Precise manipulation of m6A modifications could offer a non-pharmacological precision treatment for many diseases. However, the precise editing of m6A modification in regulating CRLM progression remains elusive. Here we integrated multi-omics and identified zinc finger and BTB domain-containing
