Helicobacter pylori Genetic Polymorphisms in Gastric Disease Development.

Infecting half of the world's population, Helicobacter pylori is a medically important bacterium that induces a variety of gastric diseases, including gastritis, peptic ulcer disease, and gastric cancer. Sequencing of almost 1000 H. pylori isolates has revealed a diverse genome that contains abundant polymorphic genetic elements; many of these lie in factors likely to be associated with virulence. To ascertain the effect of these varying genetic elements, numerous epidemiological studies have investigated the contribution of the various polymorphisms to gastric disease development; particular

Genetic investigations on causes of male infertility in Western Saudi Arabia.

In recent years, genetic studies have yielded great progress in elucidating causes of male infertility. This investigation aims to identify frequent genetic abnormalities, that is, sex chromosome aneuploidies and Y-chromosome microdeletions among infertile men in Western Saudi Arabia. From a population of infertile patients, 88 male patients with either azoospermia or severe oligozoospermia (sperm concentration <5 million/ml) were selected. In addition to a thorough clinical workup, karyotypes and Y-chromosomal microdeletions were investigated. Among those 88 infertile patients, we detected si

The Association Between Schizophrenia Risk Variants and Creativity in Healthy Han Chinese Subjects.

Although previous evidence has suggested that there is a genetic link between schizophrenia and creativity, the specific genetic variants that underlie the link are still largely unknown. To further explore the potential genetic link between schizophrenia and creativity, in a sample of 580 healthy Han Chinese subjects, this study aimed to (1) validate the role of Neuregulin 1 (NRG1) rs6994992 (one schizophrenia risk variant that has been previously linked to creativity in the European population) in the relationship between schizophrenia and creativity and (2) explore the associations between

A genetic variant of NLRP1 gene is associated with asbestos body burden in patients with malignant pleural mesothelioma.

The presence of asbestos bodies (ABs) in lung parenchyma is considered a histopathologic hallmark of past exposure to asbestos fibers, of which there was a population of longer fibers. The mechanisms underlying AB formation are complex, involving inflammatory responses and iron (Fe) metabolism. Thus, the responsiveness to AB formation is variable, with some individuals appearing to be poor AB formers. The aim of this study was to disclose the possible role of genetic variants of genes encoding inflammasome and iron metabolism proteins in the ability to form ABs in a population of 81 individual

The routes for transformation of follicular lymphoma.

PURPOSE OF REVIEW: Aggressive transformation, a frequent event in the natural history of follicular lymphoma, is associated with increased lymphoma-related mortality and yet the underlying biology remains poorly defined. This review outlines recent advances in our understanding of the genetic basis and evolutionary process leading to transformation. RECENT FINDINGS: Both the antecedent indolent and transformed follicular lymphoma (tFL) arise through branched divergent evolution with tumors emerging from a founder precursor population, the common progenitor cell. Although the majority of tFLs

Characterization of familial breast cancer in Saudi Arabia.

BACKGROUND: The contribution of genetic factors to the development of breast cancer in the admixed and consanguineous population of the western region of Saudi Arabia is thought to be significant as the disease is early onset. The current protocols of continuous clinical follow-up of relatives of such patients are costly and cause a burden on the usually over-stretched medical resources. Discovering the significant contribution of BRCA1/2 mutations to breast cancer susceptibility allowed for the design of genetic tests that allows the medical practitioner to focus the care for those who need i

Canine models of copper toxicosis for understanding mammalian copper metabolism.

Hereditary forms of copper toxicosis exist in man and dogs. In man, Wilson's disease is the best studied disorder of copper overload, resulting from mutations in the gene coding for the copper transporter ATP7B. Forms of copper toxicosis for which no causal gene is known yet are recognized as well, often in young children. Although advances have been made in unraveling the genetic background of disorders of copper metabolism in man, many questions regarding disease mechanisms and copper homeostasis remain unanswered. Genetic studies in the Bedlington terrier, a dog breed affected with copper t

Genetic variability of Anisakis simplex s.s. parasitizing European hake (Merluccius merluccius) in the Little Sole Bank area in the Northeast Atlantic.

In this study, we researched the presence of anisakids in specimens of Merluccius merluccius caught in the area of Little Sole Bank, in the Northeast Atlantic, and found that 100% of the European hake examined were infected and showed high average values of abundance (976.88) and intensity (976.88). The larvae were identified in morphological terms as morphotype type I and in molecular terms as Anisakis simplex s.s via polymerase chain reaction (PCR) restriction fragment length polymorphism of the rDNA. The genetic variability of the A. simplex s.s population in the North Atlantic is notable,

Genetic variation in apolipoprotein D affects the risk of Alzheimer disease in African-Americans.

Apolipoprotein D (APOD, gene; apoD, protein) is involved in neuroregenerative and neurodegenerative processes, and is upregulated in late-onset Alzheimer disease (AD) patients compared to nondemented controls. No genetic association studies have yet been carried out to investigate the role of APOD in AD. We have reported recently several sequence variants in the APOD gene, which are present exclusively among African blacks. In the present study we examined the role of four APOD genetic variants (Intron 1, codons 36, 108, 158) in modifying the risk of AD in 70 subjects with AD and 163 nondement

How familial is familial tremor? The genetic epidemiology of essential tremor.

Essential tremor (ET) is commonly assumed to be partly genetic. This is due to a seemingly excess aggregation within certain families. There are many families containing more than one member with ET, and some large kindreds with multiple affected individuals. Despite this, the proportion of ET cases with affected family members is not known. Estimates vary from 17 to 100%. Additionally, none of the published studies employed control subjects. Therefore, it is unknown to what extent ET aggregates within families beyond that expected by chance, and the importance of genetic susceptibility in the

Genetic dissection of the eggplant rhizosphere microbiome enables microbiome-assisted breeding.

Rhizosphere microbiome critically influences plant growth and health, yet the genetic mechanisms underlying host regulation of microbiome composition remain unclear. Here, we analyzed whole-genome genotype and 16S rhizosphere microbiome data from 432 globally sourced eggplant accessions (Solanum melongena L.). Eggplant population structure corresponded to geographic origins, with rhizosphere microbiomes varying significantly among subpopulations. Host genetics explains 9%-39% of the variation in individual microbial taxa abundance, with core taxa more affected by host genetic variation. Microb

The landscape of variants in pre-mRNA-processing factor genes in an Irish cohort.

PURPOSE: Variants in pre-messenger RNA (mRNA) processing genes account for ∼15% to 20% of autosomal dominant retinitis pigmentosa, causing severe vision loss. We provide an in-depth characterization of pre-mRNA-processing variants in the population, with a focus on genotype-phenotype correlations. METHODS: Participants underwent next-generation sequencing, variant filtering and interpretation. Impact(s) on protein structure/function were predicted through AlphaFold, ColabFold, among others. Clinical information was collated and segregation analysis undertaken. Variants were classified accordi

Proteomic Profiling of Plasma to Uncover Novel Intervention Targets and Prognostic Biomarkers for Chronic Liver Diseases.

AIMS: The burden of chronic liver disease (CLD) is increasing. This study aims to identify protein markers for CLD and its progression, and develop a protein-based risk prediction model. MATERIALS AND METHODS: We used proteome-wide Mendelian randomization (MR), Bayesian colocalization and summary-data-based MR with proteomic data from deCODE Genetics to identify CLD-related proteins. Multivariable MR was used to assess independent protein effects. Protein-protein interaction, druggability and mediation analyses were conducted to prioritise therapeutic targets. We further constructed a protein

Structure and multipartite genome architecture of the mitochondrial genome in the endangered medicinal plant Fritillaria taipaiensis P. Y. Li.

Fritillaria taipaiensis: a medicinal plant of the Liliaceae family endemic to the Qinling Mountains and southern regions of China, is renowned for its therapeutic properties in relieving cough and moistening the lungs. Despite growing interest in the organellar genomes of related species, the mitochondrial genome (mitogenome) of F. taipaiensis has remained uncharacterized. In this study, we successfully assembled and annotated the complete mitogenome of F. taipaiensis using a combination of Illumina and Nanopore technologies. The mitogenome consisted of 23 circular chromosomes with a total len

Contribution of intestinal mucosa to the disease mechanisms of Spondyloarthritis: Unlocking new insights through bioengineered tissue models.

Spondyloarthritis (SpA) represents a spectrum of chronic inflammatory rheumatic disorders with similar articular and extra-articular features and a complex pathogenesis stemming from the interplay between genetics, environmental triggers, and immune system dysregulation. The significant prevalence of SpA and its onset in the working age population place a considerable burden on healthcare systems. Furthermore, many patients fail to achieve optimal treatment outcomes and present physical and psychological distress, underscoring the need for improved therapeutic approaches. The emerging concept

Food-chain spirochetes: a unified hypothesis for Parkinson's disease and dementia risk.

Parkinson's disease (PD) and dementia have risen markedly in many regions. While aging, genetics, toxicants, and protein misfolding explain key aspects of pathology, they do not fully account for synchronized temporal trends or pronounced regional heterogeneity in recent increases. This hypothesis and theory article proposes that persistent microorganisms in industrial food systems-particularly biofilm-forming spirochetes in high-throughput poultry processing-may represent an under-recognized upstream contributor via the gut-brain axis. Analysis of Global Burden of Disease and FAOSTAT data rev

Positive and relaxed selection on mitochondrial DNA in parasitic versus predatory mites.

Mitochondrial genomes (mtDNA) have distinct evolutionary trajectories owing to their inheritance, ploidy and underlying mutation rates, making them prone to accumulate slightly deleterious mutations. Positive selection on mtDNA has been suggested to be important during adaptation to 'high-energy' lifestyles and environments. Disentangling positive versus relaxed selection in molecular mtDNA evolution studies is therefore important, although common metrics such as elevated dN/dS ratios (the ratio of non-synonymous to synonymous substitution rates) can be interpreted as signs of both relaxed pur

Beyond the skin: endocrine, psychological and nutritional aspects in women with hidradenitis suppurativa.

Hidradenitis suppurativa (HS), also known as acne inversa, is a chronic inflammatory skin disorder that primarily affects body folds and the genital area, with a higher prevalence in women across Europe. The pathogenesis of HS involves a complex interplay of intrinsic and extrinsic factors, including genetics, immunity, hormones, and environmental influences. HS is frequently associated with a variety of comorbidities, such as metabolic, endocrine, and gastrointestinal conditions, as well as mental health disorders. Although the symptoms of HS are generally similar in both men and women, femal

Novel Drug Targets for Neurodegenerative Diseases of Elderly People to Develop Effective Therapeutics: A Comprehensive Analysis.

Neurodegenerative diseases (NDs) represent an increasingly important burden of disease, particularly in the aging population. The etiology of NDs like Alzheimer's disease (AD), Parkinson's disease (PD), and Huntington's disease (HD) is associated with progressive neuronal degeneration and a paucity of effective therapies. Accumulating evidence suggests that common and intersecting genetic and pathological pathways play a critical role in disease onset and progression, revealing new opportunities for target discovery. Here, we review promising therapeutic targets based on the convergence of gen
