Anti-Inflammatory and Analgesic Properties of Polygonum minus: Mechanistic Insights and Therapeutic Potential.

Inflammation is a complex protective biological response triggered by harmful stimuli such as pathogens, chemical agents, or physical injury, and is closely associated with the development of pain and various chronic diseases. It involves a coordinated cascade of cellular and molecular events, including activation of pattern recognition receptors, release of pro-inflammatory cytokines, production of reactive oxygen species, and recruitment of immune cells. Acute inflammation is essential for host defense and tissue repair. However, persistent or dysregulated inflammation may lead to chronic in

Early blood pressure lowering and cerebral oedema in thrombolysis-treated stroke: secondary analysis of the ENCHANTED trial.

BACKGROUND AND PURPOSE: Controversy persists over the balance of benefits and harms of early intensive blood pressure (BP) lowering in thrombolysis-treated acute ischaemic stroke (AIS) patients. The BP-control arm of the Enhanced Control of Hypertension and Thrombolysis Stroke Study (ENCHANTED) (n=2196) showed that compared with guideline-recommended management (systolic BP (SBP)<180 mm Hg), intensive BP lowering did not improve functional outcome despite reducing intracranial haemorrhage. We aimed to evaluate the relationship between the BP parameters and cerebral oedema in ENCHANTED BP-contr

Lactiplantibacillus plantarum MTCC 25433 and Quercetin Blend Ameliorate Benzo[a]pyrene-Induced Gut Dysregulation in a Murine Model.

Benzo[a]pyrene (B[a]P), a carcinogenic food contaminant, is implicated in colonic abnormalities, including colorectal cancer. This study evaluated the synergistic potential of a novel synbiotic blend of Lactiplantibacillus plantarum MTCC 25433 (9 log CFU/d) and quercetin (25 mg/kg/d) against B[a]P-induced gut dysregulation in Swiss albino mice. Oral B[a]P exposure (50 mg/kg, twice weekly for 28 days) disrupted the intestinal homeostasis, enhancing lipid peroxidation (+ 44.1%), proinflammatory cytokines levels (TNF-α: + 69.6%, IL-6: + 52.9%), inflammatory transcription factors (Nf-κB: 1.92-fold

Comparison between a chimeric anti-methamphetamine monoclonal antibody and humanized antibodies on pharmacological effects of methamphetamine.

RATIONALE: Individuals with methamphetamine use disorder (MUD) can experience significant suffering due to the harmful effects of methamphetamine (METH) on physical and mental health. Although there are no approved medications for MUD, immunotherapies, including monoclonal antibodies (mAb), could serve as treatments for this debilitating condition. Our candidate anti-METH mAb called "devextinetug" features a mouse-derived variable binding region called "7F9" and a humanized constant domain. Our earlier work has shown that devextinetug is effective in altering METH's pharmacological effects. Ho

Endotracheal Tube Sizes During Surgical Procedures in Adult Patients. A Systematic Review With Meta-Analyses and Trial Sequential Analyses.

BACKGROUND: Despite the long-standing practice of endotracheal intubation, limited attention has been given to perioperative complications and benefits of choosing different sizes of endotracheal tubes. This review examines the existing literature to evaluate the potential benefits and harms of selecting different tube sizes for adult surgical patients undergoing general anesthesia for elective procedures. METHODS: This review followed the Preferred Reporting Items for Systematic Reviews and Meta-analysis (PRISMA) guidelines. The primary outcome is the proportion of participants with postoper

Family-based cognitive behavioral therapy versus family-based psychoeducation and relaxation training for obsessive-compulsive disorder in children and adolescents: a randomized clinical trial (TECTO).

Few randomized clinical trials (RCTs) have compared cognitive behavioral therapy (CBT) versus active control interventions for pediatric obsessive-compulsive disorder (OCD), and the range of investigated outcomes has been limited. We investigated benefits and harms of family-based CBT with exposure and response prevention (FCBT) versus family-based psychoeducation and relaxation training (FPRT) in pediatric OCD. This single-center RCT was investigator-initiated, independently funded, including participants with OCD aged 8-17 years with a Children's Yale-Brown Obsessive-Compulsive Scale (CY-BOC

Patterns of heated tobacco product use and biochemically verified smoking status among customers of dedicated heated tobacco product stores.

To understand the population impact of heated tobacco products (HTPs), examining the profile of their users is crucial. This study aimed to assess characteristics, patterns of HTP use, and biochemically verified smoking status of customers visiting specialized HTP stores in Greece. Two specialized stores selling IQOS HTPs were randomly selected in Athens. Researchers recruited consecutive adult customers purchasing products for personal use during store visits. Current smoking status was determined by measuring exhaled carbon monoxide (eCO), with ≥ 7 ppm classifying participants as current smo

The eyes have it: Alcohol-induced eye movement impairment and perceived impairment in older adults with and without alcohol use disorder.

BACKGROUND: While alcohol has been shown to impair eye movements in young adults, little is known about alcohol-induced oculomotor impairment in older adults with longer histories of alcohol use. Here, we examined whether older adults with chronic alcohol use disorder (AUD) exhibit more acute tolerance than age-matched light drinkers (LD), evidenced by less alcohol-induced oculomotor impairment and perceived impairment. METHOD: Two random-order, double-blinded laboratory sessions with administration of alcohol (0.8 g/kg) or placebo. Participants (n = 117; 55 AUD, 62 LD) were 40-65 years of ag

Deprescribing Benzodiazepine Receptor Agonists in Older Adults and People With Cognitive Impairment: A Systematic Review.

BACKGROUND: Benzodiazepine receptor agonists (BZRAs) (benzodiazepines and z-drugs) may provide limited benefits in the management of insomnia when used short-term. However, they increase the risk of harms including falls, fractures, hospitalizations, and cognitive impairment. Deprescribing (cessation) of BZRAs may be beneficial for older adults and people living with cognitive impairment as they are at heightened risk of adverse events. This review investigated the effects of patient-directed, non-pharmacological interventions to support deprescribing of BZRAs used for insomnia in older adults

The earliest impact of restricted protein intake during pregnancy on heart development in male mouse offspring.

INTRODUCTION: Maternal protein restriction in animal models poses significant risks that lead to detrimental outcomes such as decreased birth weights, reduced nephron counts, neurological disorders, and increased arterial hypertension. Previous studies have shown that gestational protein deficiency is linked to reductions in cardiac mass, an increased presence of cardiac fibroblasts, and the development of fibrosis in both the left and right ventricles of adult rodents. This leads to the hypothesis that nutritional deprivation negatively impacts cellular proliferation mechanisms, ultimately re

Expression of the aryl hydrocarbon receptor in Osterix-lineage cells regulates adult skeletal homeostasis in a compartment-specific manner.

Kynurenine (KYN), a tryptophan metabolite that increases with age, impairs osteoblast function. The aryl hydrocarbon receptor (AhR) has been proposed to mediate KYN's actions in bone. To test whether deletion of AhR in osteoblasts is beneficial for bone, we established an adult-onset AhR conditional knockout (CKO) model using Osx-Cre and examined the effects of AhR CKO at 4.5 and 6 mo of age (representing ~6 and 12 wk of CKO). While BMSC-derived osteoblasts from WT mice demonstrated reduced matrix formation from KYN treatment, AhR CKO osteoblasts were unaffected by KYN. Kynurenine's harmful ef

Cryoballoon versus radiofrequency ablation for persistent atrial fibrillation: an updated systematic review and meta‑analysis.

BACKGROUND: Compared with paroxysmal atrial fibrillation, persistent atrial fibrillation (AF) is more harmful and difficult to treat, and the efficacy of different catheter ablation for both also varies greatly. So this study aimed to systematically evaluate the efficacy and safety of cryoballoon (CB) and radiofrequency (RF) ablation in the treatment of persistent AF. METHODS: We searched the PubMed, Embase, and Cochrane library databases for studies comparing the efficacy and safety between CB and RF ablation for persistent AF. All included studies met our inclusion criteria. RESULTS: A tot

Effects of Supplemented Coriander, Ajwain, and Dill Seed Essential Oils on Growth Performance, Carcass Characteristics, Gut Health, Meat Quality, and Immune Status in Broilers.

Numerous studies have assessed the benefits and optimal dosage of supplementation with essential oils (EOs), including extracts from plants of the Apiaceae family, as an alternative to antibiotic growth promoters (AGPs) in broilers. However, little consideration has been given to the actual chemical composition of the evaluated EOs when drawing critical conclusions, even though EO composition can vary with different extraction conditions and plant characteristics. Therefore, the present study was conducted to evaluate the effects of EOs from seeds of plants of the Apiaceae family: coriander (C

Canagliflozin Mitigates Acute Kidney Injury Secondary to Resuscitative Endovascular Balloon Occlusion of the Aorta in a Porcine Model of Hemorrhagic Shock.

OBJECTIVE: To investigate the potential of acute canagliflozin administration to mitigate acute kidney injury (AKI) and attenuate deleterious pro-inflammatory cytokine release in a clinically relevant swine model of severe renal ischemia reperfusion injury (IRI) induced by hemorrhage and aortic occlusion. BACKGROUND: Long-term canagliflozin use attenuates renal function decline and reduces AKI in diabetes mellitus and heart failure patients. While several reports indicate prophylactic SGLT2 inhibition prevents AKI in IRI, the efficacy of acute administration on IRI and inflammation is not kno

Reducing financial toxicity in bladder cancer care.

PURPOSE OF REVIEW: Financial toxicity is a significant concern for many individuals with bladder cancer, which is, overall, the most expensive malignancy, per patient. Financial toxicity, defined as the harmful effects of treatment costs on an individual's quality of life, is associated with worse outcomes and decreased quality of life. Awareness of the objective and subjective factors that contribute to financial toxicity, and ways to mitigate their effects on patients, is essential to reduce the burden of bladder cancer care. This commentary aims to discuss the elements contributing to finan

Differences in Intraoperative Fluoroscopic Radiation Exposure During Ankle Fracture Open Reduction and Internal Fixation Between Orthopaedic Surgery and Podiatry.

Surgical ankle fractures pose a unique situation because both podiatrists and orthopaedic surgeons manage these injuries. Intraoperative fluoroscopy is routinely used; however, excessive radiation can be harmful to both the patient and the surgical team. The primary goal of this study was to determine whether there is a difference in the amount of intraoperative radiation exposure during ankle fracture open reduction and internal fixation (ORIF) when performed by orthopaedic surgeons versus podiatrists. This is a retrospective review of patients who underwent ankle fracture ORIF at an urban le

HLA-A*24 Increases the Risk of HTLV-1-Associated Myelopathy despite Reducing HTLV-1 Proviral Load.

Increased human T-cell leukemia virus type 1 (HTLV-1) proviral load (PVL) is a significant risk factor for HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). There is controversy surrounding whether HTLV-1-specific cytotoxic T lymphocytes (CTLs) are beneficial or harmful to HAM/TSP patients. Recently, HTLV-1 Tax 301-309 has been identified as an immunodominant epitope restricted to HLA-A*2402. We investigated whether HLA-A*24 reduces HTLV-1 PVL and the risk of HAM/TSP using blood samples from 152 HAM/TSP patients and 155 asymptomatic HTLV-1 carriers. The allele frequency of H
