A Molecularly Defined Midbrain Circuit Mechanism for Sound-Driven Defensive Behavior and Anxiety.

The innate defense behaviors in response to threatening sounds play a fundamental role in survival. However, the neural circuits that detect threat-relevant auditory cues and transform them into defensive behaviors remain poorly defined. Here, we identified Cerebellin-2-expressing (Cbln2+) neurons work as threatening sound detector in the inferior colliculus shell region (ICx) that were essential for the induction of sound-driven defensive responses. Cbln2+ ICx neurons specifically responded to threatening sound in a graded manner, and their activation induces escape behavior, autonomic arousa

The neural signature of high myopia: structural and functional brain alterations and their cognitive-emotional associations.

Beyond refractive error, myopia is increasingly recognized as a systemic condition with neurological implications, associated with visual dysfunction and structural retinal-choroidal alterations. This review synthesizes neuroimaging evidence demonstrating widespread neuroanatomical and functional brain changes in myopia, including cortical thinning, white matter disorganization, and disrupted functional connectivity, which may be associated with changes in cognitive-emotional systems rather than just the visual system. Mechanistically, these neural signatures reflect experience-dependent neura

A circuit from the locus coeruleus to the anterior cingulate cortex modulates offspring interactions in mice.

Social sensitivity to other individuals in distress is crucial for survival. The anterior cingulate cortex (ACC) is a structure involved in making behavioral choices and is influenced by observed pain or distress. Nevertheless, our understanding of the neural circuitry underlying this sensitivity is incomplete. Here, we reveal unexpected sex-dependent activation of ACC when parental mice respond to distressed pups by returning them to the nest ("pup retrieval"). We observe sex differences in the interactions between excitatory and inhibitory ACC neurons during parental care, and inactivation o

Postsynaptic neuronal activity promotes regeneration of retinal axons.

The wiring of visual circuits requires that retinal neurons functionally connect to specific brain targets, a process that involves activity-dependent signaling between retinal axons and their postsynaptic targets. Vision loss in various ophthalmological and neurological diseases is caused by damage to the connections from the eye to the brain. How postsynaptic brain targets influence retinal ganglion cell (RGC) axon regeneration and functional reconnection with the brain targets remains poorly understood. Here, we established a paradigm in which the enhancement of neural activity in the dista

A Review on Peripheral Tinnitus, Causes, and Treatments from the Perspective of Autophagy.

Tinnitus is the perception of phantom noise without any external auditory sources. The degeneration of the function or activity of the peripheral or central auditory nervous systems is one of the causes of tinnitus. This damage has numerous causes, such as loud noise, aging, and ototoxicity. All these sources excite the cells of the auditory pathway, producing reactive oxygen species that leads to the death of sensory neural hair cells. This causes involuntary movement of the tectorial membrane, resulting in the buzzing noise characteristic of tinnitus. Autophagy is an evolutionarily conserved

Celiac Vagus Nerve Stimulation Recapitulates Angiotensin II-Induced Splenic Noradrenergic Activation, Driving Egress of CD8 Effector Cells.

Angiotensin II (AngII) is a peptide hormone that affects the cardiovascular system, not only through typical effects on the vasculature, kidneys, and heart, but also through less understood roles mediated by the brain and the immune system. Here, we address the hard-wired neural connections within the autonomic nervous system that modulate splenic immunity. Chronic AngII infusion triggers burst firing of the vagus nerve celiac efferent, an effect correlated with noradrenergic activation in the spleen and T cell egress. Bioelectronic stimulation of the celiac vagus nerve, in the absence of othe

Abnormal Neural Progenitor Cells Differentiated from Induced Pluripotent Stem Cells Partially Mimicked Development of TSC2 Neurological Abnormalities.

Tuberous sclerosis complex (TSC) is a disease featuring devastating and therapeutically challenging neurological abnormalities. However, there is a lack of specific neural progenitor cell models for TSC. Here, the pathology of TSC was studied using primitive neural stem cells (pNSCs) from a patient presenting a c.1444-2A>C mutation in TSC2. We found that TSC2 pNSCs had higher proliferative activity and increased PAX6 expression compared with those of control pNSCs. Neurons differentiated from TSC2 pNSCs showed enlargement of the soma, perturbed neurite outgrowth, and abnormal connections among

Long-Lasting Sound-Evoked Afterdischarge in the Auditory Midbrain.

Different forms of plasticity are known to play a critical role in the processing of information about sound. Here, we report a novel neural plastic response in the inferior colliculus, an auditory center in the midbrain of the auditory pathway. A vigorous, long-lasting sound-evoked afterdischarge (LSA) is seen in a subpopulation of both glutamatergic and GABAergic neurons in the central nucleus of the inferior colliculus of normal hearing mice. These neurons were identified with single unit recordings and optogenetics in vivo. The LSA can continue for up to several minutes after the offset of

Tbx1, a DiGeorge syndrome candidate gene, is regulated by sonic hedgehog during pharyngeal arch development.

Appropriate interactions between the epithelium and adjacent neural crest-derived mesenchyme are necessary for normal pharyngeal arch development. Disruption of pharyngeal arch development in humans underlies many of the craniofacial defects observed in the 22q11.2 deletion syndrome (del22q11), but the genes responsible remain unknown. Tbx1 is a T-box transcription factor that lies in the 22q11.2 locus. Tbx1 transcripts were found to be localized to the pharyngeal endoderm and the mesodermal core of the pharyngeal arches, but were not present in the neural crest-derived mesenchyme of the phary

Mechanism of periocular acupuncture in alleviating dry eye neuropathic pain via regulation of the "periocular acupoint-trigeminal ganglion-ventral posteromedial thalamic nucleus" pathway.

OBJECTIVE: Dry eye disease (DED) is a common ocular surface disorder characterized by tear film instability and trigeminal nerve-mediated corneal hyperesthesia. Periocular acupuncture has been demonstrated to relieve DED symptoms; however, the underlying mechanisms remain unclear. Thus, in this study, we aimed to investigate the therapeutic effect of periocular acupuncture on DED and its peripheral-central regulatory mechanisms. MATERIALS AND METHODS: A guinea pig model of dry eye neuropathic pain was established through subcutaneous injection of scopolamine hydrobromide combined with topical

Disturbed Balance of Inhibitory Signaling Links Hearing Loss and Cognition.

Neuronal hyperexcitability in the central auditory pathway linked to reduced inhibitory activity is associated with numerous forms of hearing loss, including noise damage, age-dependent hearing loss, and deafness, as well as tinnitus or auditory processing deficits in autism spectrum disorder (ASD). In most cases, the reduced central inhibitory activity and the accompanying hyperexcitability are interpreted as an active compensatory response to the absence of synaptic activity, linked to increased central neural gain control (increased output activity relative to reduced input). We here sugges

Gut microbiota-mediated neuroinflammation following stroke: mechanisms and intervention strategies.

BACKGROUND: Stroke is a complex and highly heterogeneous acute cerebrovascular disease characterized by high incidence, disability rates, and mortality. The occurrence of a stroke is the result of a series of chain reactions. Neuroinflammation, as a core component of the pathological process of stroke, serves as the dynamic hub of this chain of reactions. It acts both as a driving force in disease progression and a key regulator of functional recovery. Increasing evidence indicates that the gut microbiota has a dynamic coupling relationship with the neuroinflammatory pathological processes inv

Castration-resistant prostate cancer as an adaptive tumor ecosystem: coupling tumor evolution with microenvironmental reprogramming.

Castration-resistant prostate cancer (CRPC) represents a lethal stage of prostate cancer that emerges following sustained androgen deprivation therapy (ADT) and remains a major clinical challenge in the era of androgen receptor (AR)-targeted therapies. Although AR reactivation and lineage plasticity are recognized as central mechanisms of resistance, these tumor-intrinsic processes alone cannot fully explain the profound heterogeneity and therapeutic failure observed during CRPC evolution. Here, integrating recent advances in single-cell and spatial profiling, we propose a unified framework in

Role and modulation of various spinal pathways for human upper limb control in different gravity conditions.

Humans can perform movements in various physical environments and positions (corresponding to different experienced gravity), requiring the interaction of the musculoskeletal system, the neural system and the external environment. The neural system is itself comprised of several interactive components, from the brain mainly conducting motor planning, to the spinal cord (SC) implementing its own motor control centres through sensory reflexes. Nevertheless, it remains unclear whether similar movements in various environmental dynamics necessitate adapting modulation at the brain level, correctin

Increased coactivation during clinical tests of spasticity is associated with increased coactivation during reactive standing balance control in cerebral palsy.

Joint hyper-resistance is a common symptom in cerebral palsy (CP). It is assessed by rotating the joint of a relaxed patient. Joint rotations also occur when perturbing functional movements. Therefore, joint hyper-resistance might contribute to reactive balance impairments in CP. Our aim was to investigate relationships between altered muscle responses to isolated joint rotations and perturbations of standing balance in children with CP. Twenty children with CP and 20 typically developing children participated in the study. During an instrumented spasticity assessment, the ankle was rotated as

Prediction on nature of cancer by fuzzy graphoidal covering number using artificial neural network.

Predicting the chances of various types of cancers for different organs in the human body is a typical decision-making process in medicine and health. The signaling pathways have played a vital role in increasing or decreasing the possibility of the deadliest disease, cancer. To combine the pathways concept and ambiguity in the prediction techniques of such diseases, we have used the proposed research on fuzzy graphoidal covers of fuzzy graphs in this paper. Determining a path with uncertainty and shortest length is a challenging topic of graph theory, and a collection of such shortest paths m

An analysis of differential gene expression in peripheral nerve and muscle utilizing RNA sequencing after polyethylene glycol nerve fusion in a rat sciatic nerve injury model.

Application of polyethylene glycol (PEG) to a peripheral nerve injury at the time of primary neurorrhaphy is thought to prevent Wallerian degeneration via direct axolemma fusion. The molecular mechanisms of nerve fusion and recovery are unclear. Our study tested the hypothesis that PEG alters gene expression in neural and muscular environments as part of its restorative properties. Lewis rats underwent unilateral sciatic nerve transection with immediate primary repair. Subjects were randomly assigned to receive either PEG treatment or standard repair at the time of neurorrhaphy. Samples of sci
