We report the first case of a child with mild dystrophinopathy associated with Klinefelter's syndrome (karyotype 47, XXY). This 3.5-year-old boy was affected by some symptoms suggestive of Becker's muscular dystrophy. Dystrophin immunostaining and immunoblotting procedures confirmed the diagnosis, but polymerase chain reaction-directed gene analysis failed to reveal any macrodeletion. We report a patient who developed bronchiolitis obliterans organizing pneumonia (BOOP) after syngeneic BMT for ALL. The patient complained of persistent low-grade fever and non-productive cough after engraftment. Chest CT scan showed patchy infiltration bilaterally in the lower lung fields. This report presents two probable cases of Lyme arthritis triggered by athletic activity or trauma. One patient had ankle swelling after he began a jogging program, and a second had knee swelling that followed a twisting injury. The report describes the three stages of the illness and presents diagnostic lab tests and the therapies used. Polymerase chain reaction (PCR) testing of the synovial fluid can be useful in diagnosing active Lyme arthritis, but it may yield a false-negative result when there has been recent remission of the arthritis.

Between 1981 and 1994, the detection of specific antibodies against Toxoplasma gondii was performed simultaneously in serum and aqueous humor from 90 patients, and the Witmer-Desmonts coefficient measuring the relative concentration of antibodies produced locally was calculated to confirm the clinical diagnosis of ocular toxoplasmosis. A 61-year-old woman with chronic lymphocytic leukaemia (CLL) was found to have multiple pulmonary nodules on an annual chest radiograph 4 months after recovery from chickenpox. Human T-cell lymphotropic virus type I (HTLV-I) is characterized by a remarkable genetic stability and high proviral loads in the absence of malignant disease. This results from the effect of tax on cell cycling. The virus replicates essentially in concert with the cell that is, via mitosis, which can be shown by polymerase chain reaction amplification of the HTLV-I integration sites. This is true of all stages of HTLV-I infection and accompanies adult T-cell leukemia/lymphoma. The very low viremia results from its genetic organization.

Accurate carrier determination is an important aspect in providing prenatal diagnosis and genetic counselling to families with Duchenne/Becker muscular dystrophy patients. Using quantitative polymerase chain reaction, we have analyzed the carrier status of 31 mothers (5 familial and 23 sporadic) who have an affected son with known deletion in the dystrophin gene. Only four out of 23 mothers of sporadic cases turned out to be heterozygous for the deleted exons. The lower number of carrier mothers in sporadic cases suggests a higher frequency of new mutations in North Indian DMD@BMD patients.

Five hundred pregnant women were analyzed for the presence of alpha-thalassemia-1 of the Southeast Asian (SEA)-type by polymerase chain reaction (PCR) technique at the Maharaj Nakhon Chiang Mai University Hospital in Chiang Mai during the period from April to June 1995. Forty-four of them (8.8%) were recognized as carriers, corresponding to a frequency of 0.044. Homozygous alpha-thalassemia-1 of the SEA-type, the fatal condition of hemoglobin Bart's hydrops fetalis, has an expected frequency of 0.00194, or about 2 hydrops fetalis cases per 1,000 births in this population.

Our routine tests for tropical viruses document that several hundreds of Dengue fever cases are imported into Germany every year. In contrast, hemorrhagic fever cases are rarely diagnosed in Germany. Our investigations suggest that this low number is due to the different living conditions of the local population in the tropics compared with that of travellers from Europe or North America. Improved methods for detecting Dengue virus infections, e.g. three different antibody tests and the reverse transcriptase-polymerase chain reaction (RT-PCR) for detection of viral RNA, have been developed.

Blood lymphocytes from patients with eosinophilia are known to produce interleukin-5 (IL-5) with appropriate stimulation in vitro. OBJECTIVE: To assess whether the HLA-DR4 association found in rheumatoid arthritis (RA) is also seen in the large granular lymphocyte (LGL) syndrome.
METHODS: HLA-DR genotyping was performed using restriction fragment length polymorphism and polymerase chain reaction analysis.
