Heterogeneity of T-acute lymphoblastic leukemia (T-ALL) cell lines: suggestion for classification by immunophenotype and T-cell receptor studies.

Hematopoietic cell lines are often used as representatives for a certain cell differentiation lineage and stage, particularly in immunological and hematological studies. Acute lymphoblastic leukemia (ALL) of T-cell type is a rather heterogeneous group of ALL at least by immunophenotyping. Our aim was to present a comprehensive characterization of frequently used T-cell leukemia cell lines and to suggest a correlation with the normal differentiation pattern. A total of 16 T-ALL cell lines were analyzed for their immunophenotype and for T-cell receptor (TCR) rearrangement and expression. The pan

Rearranged patterns of IgH and TcR gamma genes in patients with acute lymphoblastic leukemia.

The rearrangement of immunoglobulin heavy chain gene (IgH) and T cell receptor gamma gene (TcR gamma) was studied in 30 patients with acute lymphoblastic leukemia (ALL) by the polymerase chain reaction (PCR). 19 cases was found to have rearrangement of IgH gene, 12 of TcR gamma. Most of IgH rearrangement was characterized by one or two specific bands while some had more than two. Rearrangement of TcR gamma gene appeared as one specific band. A slight difference in number, size and lightness of bands was found among the patients. 4 different kinds of rearrangement were observed in the detection

Virus-cell interactions in a natural killer-like cell line from a patient with lymphoblastic lymphoma.

Lymphoproliferative disorders involving Epstein-Barr virus (EBV) infected natural killer (NK) cells are reported with increasing frequency, but the nature and role of EBV infection in these cells remains undefined. In this study, we have investigated virus-cell interactions in the EBV-positive YTN10 cell line, an NK-like cell line established from a patient with lymphoblastic lymphoma. Low level expression of the EBV receptor CD21 molecule was detected by FACS and reverse transcriptase polymerase chain reaction (RT-PCR) analysis. Immunoblotting and RT-PCR analysis identified a latency II patte

De novo Ph-negative acute T-lymphoblastic leukemia with BCR gene rearrangement.

We herein report a rare case of Ph chromosome-negative acute T-lymphoblastic leukemia (T-ALL) with a classical BCR rearrangement seen in chronic myelogenous leukemia (CML). There were no clinical or morphologic features to suggest a previous chronic phase of CML. The importance of a full analysis of the BCR gene in the case of adult acute lymphoblastic leukemia (ALL), especially in T-ALL, are discussed.

Expression of G alpha 16, a G-protein alpha subunit specific for hematopoiesis in acute leukemia.

G-proteins are essential in signal transduction pathways. A G-protein alpha subunit termed G alpha 16 was found to be exclusively expressed in hematopoietic cell lines. In cells derived from patients, G alpha 16 expression has been detected in progenitor- and pre-B ALL cells and also in peripheral blood stem cells (PBSC). In this study, we analyzed G alpha 16 expression using a RT-PCR technique by testing elutriated blood cells from normal donors, PBSC from breast cancer patients and bone marrow or peripheral blood cells from acute leukemia patients. Both of two ALL patients and 15/16 AML pati

Immunophenotypic, cytogenetic and molecular investigations in two cases of CALLA positive acute myeloid leukemia.

Clinicopathological and cytogenetic features of two patients with acute myelogenous leukemia (AML) whose blast cells coexpressed myeloid-associated antigens and CALLA are described. Leukemia cells revealed myelomonocytic (FAB-M4) and monocytic (FAB-M5) features, while the nonblast cell population exhibited trilineage myelodysplasia in both cases, a finding suggestive of multiple-cell-lineage involvement. Cytogenetically, a deletion of the long arm of chromosome 6 was found in one patient, and normal metaphases were detected in the other. Molecular studies disclosed a rearrangement of the IgH l

Optimal management of elderly/old Ph+ acute lymphoblastic leukemia patients.

Today, Philadelphia-positive acute lymphoblastic leukemia (Ph+ ALL) is a curable disease. In real life, too many adult ALL patients are not adequately worked up at diagnosis and treated, and this occurs, in particular, in elderly/old individuals. Here, we present representative case descriptions to discuss how Ph+ ALL patients diagnosed in their seventh, eight and ninth decade of life can, through a timely, accurate and personalized administration of tyrosine kinase inhibitor (TKI), without systemic chemotherapy, experience long-lived responses, minimal residual disease negativity, and a good

Risk of Hepatitis B Reactivation Among ALL Patients With Occult Hepatitis B Infection.

PURPOSE: Anticancer therapy increases the risk of hepatitis B virus (HBV) reactivation in patients with occult hepatitis B infection (OHBI), particularly in those who receive rituximab or stem-cell transplantation. However, the exact risk of HBV reactivation in patients with ALL/lymphoblastic lymphoma (LBL) receiving prolonged, intensive myelosuppressive chemotherapy is not known and thus the role of antiviral prophylaxis is not clearly established. PATIENTS AND METHODS: This prospective observational study was conducted at a single tertiary cancer center in India (October 2017 to August 2021

Structural rewiring of IL-7R dimerization by an oncogenic transmembrane mutation can be reversed by rational design.

Mutations within the transmembrane domains (TMDs) of single-pass transmembrane receptors often cause aberrant, ligand-independent receptor signaling associated with diverse malignancies, but their mechanism of action remains largely unknown. These TMD mutations are generally not targetable as they are buried in the membrane. Here, we determined the mechanism of a gain-of-function (GOF) TMD mutation of interleukin-7 receptor (IL-7R) associated with T cell acute lymphoblastic leukemia and addressed the possibility of directly targeting the TMD mutation by using rationally designed transmembrane

The association of urinary levels of heavy metals with childhood acute leukemia and its socio-environmental risk factors: A case-control study.

Childhood acute leukemia is the most common type of cancer in children worldwide, accounting for 30% of all childhood cancers. This study aimed to evaluate the possible association between urinary heavy metals (HMs) concentration and the risk of childhood acute lymphoblastic leukemia (C-ALL) in children. The effects of socio-environmental factors also were investigated. In this case-control study, urinary concentrations of selected HMS were determined in urine samples of 124 children with acute leukemia (cases) and 104 healthy controls using ICP-OES. The social, economic, and environmental cha

Heterogeneity in the Global Practice of Central Nervous System Staging in Pediatric Acute Lymphoblastic Leukemia.

BACKGROUND: Central nervous system (CNS) involvement in childhood acute lymphoblastic leukemia (ALL) is assessed by cell counting and cytomorphology from cerebrospinal fluid (CSF) and is used for treatment stratification worldwide. The ratio of "CNS2" patients in clinical trials ranges from 3% to 40%, with unclear prognostic significance. PROCEDURE: To assess real-world practice, a survey was distributed globally. Questions focused on the systemic treatment allowed before the staging lumbar puncture, sample handling, methodological details of different analytics, and staging definitions. RES

Comparison of obecabtagene autoleucel versus an external control arm in adult patients with relapsed/refractory B-cell acute lymphoblastic leukemia.

In the single-arm Phase Ib/II FELIX study (NCT04404660), obecabtagene autoleucel (obe-cel; CD19-directed autologous CAR T-cell therapy) demonstrated high overall remission rates (ORR) and a favorable safety profile in adults with relapsed/refractory (R/R) B-cell acute lymphoblastic leukemia (B-ALL). To contextualize results from FELIX, we compared the efficacy and safety of obe-cel with matched external control arms (ECA) derived from historical trials using propensity score matching. ECAs represented standard of care (SoC) non-CAR T-cell therapies: blinatumomab, inotuzumab ozogamicin, and con

Tracking Health Related Quality of Life From Diagnosis to Follow-Up Among Australian Children With Acute Lymphoblastic Leukaemia.

BACKGROUND: Acute lymphoblastic leukaemia (ALL) is one of the most treatable forms of paediatric cancer; however, there is a substantial burden of treatment-related toxicities (TRTs). In addition, the long-term changes in children's health-related quality of life (HRQoL) due to toxic treatments are not well understood. AIMS: To describe parent-reported HRQoL of children with ALL over a 5-year period from diagnosis and identify risk factors associated with parent reporting of low HRQoL in children. METHODS: Through a national longitudinal prospective study, we invited parents of children diag

Integrative Genetic and Transcriptomic Subtyping Improves Prognosis Prediction in B-Lineage Acute Lymphoblastic Leukemia.

Whole-transcriptomic sequencing (WTS) has remarkably advanced our understanding of B-lineage acute lymphoblastic leukemia (B-ALL), allowing for detailed gene expression profiling and discovery of novel therapeutically relevant subtypes. The aim of this study was to evaluate the diagnostic and prognostic relevance of combining WTS with traditional genetic methods in risk-stratifying B-ALL. In a cohort of 394 patients (301 children and 93 adults), conventional techniques such as fluorescence in situ hybridization, cytogenetics, and reverse-transcription PCR identified sentinel chromosomal abnorm

Age-adjusted dosing of fludarabine for lymphodepletion in CAR T-cell therapy: a clinical trial simulation study.

Fludarabine (FLU) is used for lymphodepletion and improves the persistence and expansion of chimeric antigen receptor (CAR) T cells in vivo. Higher FLU systemic exposure is associated with lower relapse risk and improved leukemia-free survival in pediatric patients with acute lymphoblastic leukemia treated with CD19 CAR T-cell therapy. FLU pharmacokinetics (PKs) is age dependent, with increased clearance in younger children. Here, we used modeling and simulation, including clinical trial simulations, to define age-adjusted FLU dosage regimens that may maintain effective FLU exposures and impro

L-asparaginase: a review of microbial production and protein engineering with translational applications for oncology and food safety.

L-Asparaginase (LA; EC 3.5.1.1) is a clinically and industrially relevant enzyme that hydrolyzes L-asparagine to L-aspartate and ammonia. LA has demonstrated efficacy as a chemotherapeutic agent by depleting L-asparagine, an amino acid essential for leukemic cell survival. FDA-approved formulations derived from Escherichia coli and Erwinia chrysanthemi face challenges, including a short half-life and adverse side effects, necessitating continuous efforts to develop more stable and specific variants. Concurrently, LA's role in acrylamide mitigation during food processing has garnered attention,
