Chronobiological analysis of circadian patterns in transcription of seven key clock genes in six peripheral tissues in mice.

The molecular clock machinery in mammals consists of a number of clock genes (CGs) and their resultant proteins that form interlocking transcription-translation feedback loops. These loops generate and maintain the 24 h mRNA and protein oscillations and consequential biological and physiological rhythms. To understand whether peripheral oscillators share similarly-timed clock machinery, the temporal expression patterns of the seven recognized key CGs (mPer1, mPer2, mCry1, mCry2, mRev-erb alpha, mClock, and mBmal1) were examined simultaneously in six peripheral tissues in mice every 4 h for 24

Ceramidase is involved in metabolic reprogramming of fibroblasts under hypoxia conditions.

INTRODUCTION: Physiological metabolic reprogramming is a cells strategy to resist/counteract stress to preserve homeostasis. Hypoxia is an important stress stimulus that can promote tissue remodeling and fibrosis, in which alterations in sphingolipid metabolism may contribute to fibroblast responses. Here, we investigated the potential role of acid ceramidase (ASAH1), a key enzyme of the sphingolipid salvage pathway, in fibroblast adaptation to hypoxic stress. METHODS: Human fibroblasts were exposed to hypoxia for 24, 48, and 72 hours, and metabolic and redox changes were investigated followi

Alphitolic acid suppressed osteoclast differentiation via hindering Cox-2/NF-κB signal pathway and alleviated titanium particle-induced bone loss in vivo.

Total joint arthroplasty (TJA) effectively improves the quality of life for patients with end-stage joint disorders. However, aseptic loosening caused by titanium (Ti) wear debris-induced osteolysis remains the leading cause of implant failure and revision surgeries. In this study, we investigated the therapeutic potential of alphitolic acid (ALA), a pentacyclic triterpenoid with known anti-inflammatory properties, against Ti particle-induced osteolysis and explored its underlying mechanisms. Network pharmacology analysis combined with molecular docking predicted that ALA targets key regulator

RNA-binding protein hnRNPD promotes the proliferation of Wilms' tumor cells through activating the p38 MAPK signaling pathway.

Wilms' tumor (WT) represents a kidney carcinoma predominantly affecting children aged five years and younger. Heterogeneous nuclear ribonucleoprotein D (hnRNPD), an RNA-binding protein, has been implicated in oncogenic processes across various tumor types, whereas its expression pattern and biological function in WT remain largely unknown. hnRNPD expression within WT tissues was evaluated using publicly available databases, and co-expressed genes were identified through bioinformatic analysis. In vitro, hnRNPD expression in WT cell lines 17.94 and HFWT was modulated via gene silencing or overe

Endometrioid Versus Seromucinous Borderline Ovarian Tumors: Divergent Molecular Signatures and a Shared Role as Precursors to Endometrioid Carcinoma.

Endometrioid borderline tumors (EBTs) and seromucinous borderline tumors (SMBTs) are rare ovarian neoplasms with distinct histologic features. However, the molecular profiles of EBTs and SMBTs remain incompletely characterized. We performed histologic evaluation and DNA/RNA next-generation sequencing (NGS) using a 1425-gene pan-cancer panel on 11 EBTs and 10 SMBTs to define their mutational landscapes, conduct cross-comparisons between EBTs and SMBTs, and evaluate both against established profiles of endometrioid carcinoma and low-grade serous carcinoma. Histologically, EBTs showed adenofibrom

Helicobacter pylori activates histone lactylation to promote gastric cancer progression and immune evasion through the HAS2/c-MYC/PD-L1 axis.

BACKGROUND: Metabolic reprogramming and epigenetic alterations play important roles in driving tumor microenvironment (TME) remodeling and immune evasion in gastric cancer (GC). Histone lactylation links glycolytic metabolism to transcriptional regulation, but its role in Helicobacter pylori (H. pylori)-associated GC progression remains largely elusive. METHODS: Multiomics analysis (ChIP-seq and RNA-seq) with in vitro GC/CD8+ T-cell coculture systems and in vivo mouse xenograft models were employed in this study. Clinical GC tissue cohorts were analyzed to validate the clinical relevance of t

Hypoxia-driven TCF21/CCL11 axis in GCMSCs orchestrates macrophage polarization and lymphatic remodeling to promote gastric cancer lymph node metastasis.

Lymph node metastasis (LNM) is a primary critical factor for gastric cancer (GC) prognosis, fundamentally driven by the adaptive remodeling of the tumor microenvironment (TME). While gastric cancer-associated mesenchymal stem cells (GCMSCs) are recognized as pivotal orchestrators of the tumor niche, the paracrine mediators governing LNM remain poorly defined. In this study, integrative transcriptomic and cytokine profiling identified CCL11 as the most significantly upregulated chemokine in GCMSCs. Clinical analysis demonstrated that elevated CCL11 expression in tumor tissues correlated with ad

PMEPA1 modulates YAP1 nuclear translocation to disrupt EMT subtypes and promote metastasis in Biliary tract cancer.

Biliary tract cancer (BTC) is an aggressive tumor with poor prognosis and limited treatment options. Epithelial-mesenchymal transition (EMT) significantly contributes to BTC metastasis, yet the critical regulators and underlying molecular mechanisms of EMT remain poorly unclear. In our study, we conducted an integrative analysis of single-cell RNA sequencing (scRNA-seq) data from 47 BTC samples, identifying two EMT-enriched epithelial subpopulations and a BTC specific EMT gene set. Using the EMT gene set, we classified BTC patients and noticed that those with elevated EMT scores had worse prog

Characterization of andrographolide-loaded exosome-like nanoparticles from Nauclea officinalis and their modulation of inflammatory and lipid metabolic pathologies in WRL68 cells.

Andrographolide (AG), a natural diterpenoid compound derived from Andrographis paniculata, exhibits potential against non-alcoholic steatohepatitis (NASH). However, its therapeutic utility is limited by poor solubility, short half-life, and low bioavailability. This study aimed to enhance AG's efficacy by encapsulating it into exosome-like nanoparticles (ELNs) isolated from Nauclea officinalis (N. officinalis, Rubiaceae family) and evaluating its anti-NASH activity in WRL68 cells. This study isolated Nauclea officinalis-derived ELNs (N-ELNs) via ultracentrifugation. Loganin, andrographolide, a

Dynamic effects of sleep deprivation on emotional behavior, circadian rhythm genes, and inflammatory infiltration in the medial prefrontal cortex.

BACKGROUND: Sleep, a core circadian rhythm, maintains physiological homeostasis. Its dysfunction links to neuropsychiatric disorders. Clinically, poor sleep impairs positive emotions and enhances negative emotion susceptibility, but the mechanism remains unclear, potentially involving circadian clock genes and neuroinflammatory pathways. METHODS: Divide the male C57BL/6J mice into the following five groups: Non-sleep deprivation (SD) control (CON), sleep recovery 14-day after SD 7-day (SD7R14), sleep recovery 21-day after SD 7-day (SD7R21), sleep recovery 14-day after SD 14-day (SD14R14), and

N-Acylethanolamine Acid Amidase Inhibition Reduces SARS-CoV-2 Infection in Human Precision Cut-Lung Slices and Downregulates NF-KBB Signalling.

SARS-CoV-2, like other positive-sense RNA viruses, manipulates host lipid metabolism to facilitate its replication by enhancing lipogenesis and lipid droplet formation. This infection disrupts bioactive lipid levels associated with the inflammatory response by increasing nuclear factor-κB (NF-κB) transcription. Recent findings have shown that NF-κB activation is essential for sustaining SARS-CoV-2 replication. Therefore, we proposed that counteracting NF-κB-driven pro-inflammatory lipid production could be accomplished by enhancing an anti-inflammatory, lipolytic pathway. Our goal was to incre

Divergent mechanistic reliance on the phosphatidylinositol 4-kinase (PI4K)/oxysterol-binding protein (OSBP) axis during Zika virus versus dengue virus and West Nile virus replication organelle biogenesis.

Positive-strand RNA viruses remodel host endoplasmic reticulum (ER) membranes into replication organelles (ROs) that support genome replication. Members of the genus Orthoflavivirus exploit host lipid pathways, particularly the phosphatidylinositol 4-kinase (PI4K)-phosphatidylinositol 4-phosphate (PI4P)-oxysterol-binding protein (OSBP) axis, yet they display marked mechanistic divergence. Specifically, it raised a central and unresolved question: is local PI4P enrichment an indispensable determining factor for viral vesicle biogenesis, or is it merely a contributing factor to a more fundamenta

Panax notoginseng saponins alleviates metabolic dysfunction-associated steatotic liver disease in mice by influencing Nrf2/MT2.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a metabolic liver injury driven largely by oxidative stress. The nuclear factor erythroid 2-related factor 2 (Nrf2) pathway plays a central role in antioxidant defense, and metallothionein-2 (MT2) is a potential downstream target. Panax notoginseng saponins (PNS) exhibit anti-inflammatory and antioxidant properties, but their effects on MASLD via Nrf2/MT2 signaling remain unclear. This study aimed to investigate whether PNS exerts protective effects against MASLD through Nrf2/MT2 signaling pathway. METHODS: Apolip

Ex vivo pretreatment of donor organ with siRNA nanoparticles attenuates cold ischemia-reperfusion injury in cardiac transplantation.

Heart transplantation is an optimal therapeutic regimen for terminal-stage cardiac failure. However, cold ischemia-reperfusion injury (CIRI) remains an unavoidable and outstanding challenge, which is a significant obstacle to early graft dysfunction and long-term survival. Blockage of complement, apoptosis, and inflammation by small interfering RNA is considered a strategy for attenuating CIRI and protecting cardiac function. However, their delivery to the donor organ is still a serious challenge due to the polyanionic nature and high molecular weight properties. Here, we have designed a novel
