Chronic fluoride toxicity: dental fluorosis.

Dental fluorosis occurs as a result of excess fluoride ingestion during tooth formation. Enamel fluorosis and primary dentin fluorosis can only occur when teeth are forming, and therefore fluoride exposure (as it relates to dental fluorosis) occurs during childhood. In the permanent dentition, this would begin with the lower incisors, which complete mineralization at approximately 2-3 years of age, and end after mineralization of the third molars. The white opaque appearance of fluorosed enamel is caused by a hypomineralized enamel subsurface. With more severe dental fluorosis, pitting and a l

Maxillary segmental distraction in children with unilateral clefts of lip, palate, and alveolus.

OBJECTIVES: Alveolar clefts are commonly closed by a bone grafting procedure. In cases of wide clefts the deficiency of soft tissue in the cleft area may lead to wound dehiscence and loss of the bony graft. Segmental maxillary bony transfer has been mentioned to be useful in such cases. Standard distraction devices allow unidirectional movement of the transported segment. Ideally the distraction should strictly follow the dental arch. The aim of this study was to analyze distraction devices that were adapted to the individual clinical situation of the patients. The goal was to achieve a distra

Homozygous defects in LMNA, encoding lamin A/C nuclear-envelope proteins, cause autosomal recessive axonal neuropathy in human (Charcot-Marie-Tooth disorder type 2) and mouse.

The Charcot-Marie-Tooth (CMT) disorders comprise a group of clinically and genetically heterogeneous hereditary motor and sensory neuropathies, which are mainly characterized by muscle weakness and wasting, foot deformities, and electrophysiological, as well as histological, changes. A subtype, CMT2, is defined by a slight or absent reduction of nerve-conduction velocities together with the loss of large myelinated fibers and axonal degeneration. CMT2 phenotypes are also characterized by a large genetic heterogeneity, although only two genes---NF-L and KIF1Bbeta---have been identified to date.

Ovariectomy hinders the early stage of bone-implant integration: histomorphometric, biomechanical, and molecular analyses.

Postmenopausal osteoporosis is a contributing factor to alveolar bone atrophy associated with tooth loss in the elderly. The use of dental titanium implants has been increasingly adapted to treat these edentulous patients. This study examines whether female gonadal hormone deficiency interferes with the critical integration process between bone and implants. Two types of experimental titanium implants with acid-treated surfaces were placed in the femurs of ovariectomized (ovx) and sham-operated control rats: T-cell implants with a hollow chamber for histomorphometric and steady-state mRNA expr…

Subgingival debridement of root surfaces with a micro-brush: macroscopic and ultrastructural assessment.

AIM: The aim of this study was to assess the use of a micro-brush to remove plaque deposits from subgingival, periodontally involved root surfaces in vivo. METHODS: 30 periodontally involved teeth requiring extraction for periodontal or prosthetic reasons in 26 adult patients were utilised. For inclusion, teeth had to display at least 30% bone loss radiographically. Following the establishment of local anaesthesia, grooves were cut on the proximal root surface adjacent to the gingival margin at the line angles. For each tooth, 1 proximal root surface was rubbed with the micro-brush for 2 min

The prevalence and intraoral distribution of periodontal attachment loss in a birth cohort of 26-year-olds.

BACKGROUND: Most research efforts in periodontal epidemiology have focused on middle-aged or older people, giving a picture of disease occurrence at a relatively late stage in the natural history of the condition. There is a paucity of comprehensive descriptive data from younger age groups. Understanding the epidemiology and clinical presentation of the condition earlier in the disease course may enable more appropriate interventions. METHODS: The aim of this study was to describe the occurrence of gingival recession, probing depth, periodontal attachment loss (AL), and gingivitis among parti

Neuropeptide Y expression in the trigeminal ganglion and mandibular division of the trigeminal nerve after inferior alveolar nerve axotomy in young rats.

Neuropeptide Y (NPY) is a 36-amino-acid peptide residing in sympathetic nerve terminals, originating from the superior cervical ganglion in oral tissues. NPY exerts vasoconstrictor action together with noradrenalin and has been found to inhibit the release of neurotransmitters from primary afferent fibers. During regeneration of the axotomized inferior alveolar nerve (IAN), NPY-immunoreactive (IR) nerve fibers have been shown in the odontoblast layer and dentin, an area normally innervated by afferent nerve fibers. The dynamic shift in neuropeptide expression in the trigeminal ganglion and in

Edentulousness and Frailty: A Study of Directionality and Pathways.

Associations between tooth loss and frailty have been reported, but longitudinal evidence on directionality and pathways of this association is lacking. We examined the direction and pathways of the association between edentulousness and frailty among older adults in England using a nationally representative sample of 6,800 adults aged ≥50 y who participated in the English Longitudinal Study of Ageing (ELSA) at wave 3. Edentulousness and frailty (determined using the Frailty Index) were assessed at waves 3 (2006/7), 5 (2010/11), and 7 (2014/15). The following mediators were assessed at waves 4…

The nuclear-encoded plastid ribosomal protein bL31c of Arabidopsis thaliana functions in chloroplast biogenesis and leaf development.

Plastid ribosomes are composed of large (50S) and small (30S) subunits, each made up of ribosomal RNAs (rRNA) and plastid ribosomal proteins (PRPs). While numerous PRPs have been characterized, the functions of several PRPs remain elusive. We followed a reverse genetics approach in Arabidopsis to functionally characterize nuclear genes encoding PRPs for which no viable mutant alleles had been described. We report here the characterization of 50S subunit PRP bL31c. Its gene, RPL31, was previously identified as essential for embryo development (EMB2184), but viable alleles were unavailable. RPL3

Overexpression of the IL-23/IL-17A Axis and Their Receptors (IL-23R and IL-17RA) in Gingival Tissue of Patients With Periodontitis.

BACKGROUND: Periodontitis is a chronic immunoinflammatory disease involving various components that affect the tissues surrounding the tooth. The host immune response to the presence of periodontopathogenic microorganisms activates several cytokine systems involved in alveolar bone resorption in periodontitis; among them is the receptor activator of nuclear factor-κB (RANK)/receptor activator of nuclear factor-κB ligand (RANKL) system, which depends on the interleukin (IL)-23/IL-17 axis. There is a tendency toward increased IL-23, IL-17, and IL-17 receptor (IL-17RA) and a discrepancy in IL-23

A Case of Peripheral Odontogenic Myxofibroma Arising in the Palatal Gingiva of the Maxillary Second Premolar Region: A Case Report.

Odontogenic myxofibroma (OMF) is a rare benign mesenchymal odontogenic tumor characterized by myxoid stroma with a prominent fibrous component. Although it usually arises intraosseously within the jaws, the peripheral variant, peripheral odontogenic myxofibroma (POMF), which occurs in extraosseous soft tissues, is uncommon and may be clinically misdiagnosed as a reactive gingival lesion. We report a case of POMF in a 68-year-old man who was referred for evaluation of a painless, slowly enlarging swelling of the palatal gingiva in the left maxillary second premolar region, which had initially b

Myeloid dendritic cells and periodontal disease association: integrated study of single-cell sequencing and Mendelian randomization analysis.

BACKGROUND: Periodontal disease is a widespread inflammatory condition that compromises the supporting structures of the teeth, potentially resulting in tooth loss if left untreated. Despite advancements in therapeutic interventions and an enhanced understanding of its pathophysiology, emerging techniques such as single-cell RNA sequencing (scRNA-seq) and Mendelian randomization (MR) present new opportunities for precision medicine in the management of periodontal disease. METHODS: Data derived from the GSE152042 dataset underwent rigorous quality control, normalization, and dimensionality re…

Prevalence of periodontitis in adolescents: A systematic review protocol.

Periodontitis is considered an inflammatory disease that affects the tooth's supporting structures, resulting in its progressive destruction, which can lead to several problems, such as loss of teeth, bone reabsorption, and even exacerbation of chronic systemic diseases. Periodontitis is a common periodontal condition in the adult dentate population, affecting approximately 6 in 10 individuals. Among those affected, approximately 25% develop the most severe form of the disease. The prevalence of periodontitis in adolescents is not well established. Therefore, this study protocol of a systemati

Gingipains protect Porphyromonas gingivalis from macrophage-mediated phagocytic clearance.

Porphyromonas gingivalis (Pg) is a keystone pathogen in periodontitis, a highly prevalent disease manifested by chronic inflammation of the periodontium, alveolar bone resorption and tooth loss. During periodontitis pathobionts such as Pg can enter the bloodstream and growing evidence correlates periodontitis with increased risk of cardiovascular and neurodegenerative diseases. However, the mechanism by which immune cells respond to Pg challenge in vivo remains elusive. Pg produce aggressive proteolytic virulence factors termed gingipains which not only provide nutrients necessary for bacteria…

The inhibitory effects of 7ND protein on osteoclast differentiation in apical periodontitis.

BACKGROUND: Apical periodontitis (AP) is a highly prevalent inflammatory condition that affects the tissue surrounding the apex of a tooth root. 7ND protein, a mutant form of monocyte chemoattractant protein-1 (MCP-1), functions as a dominant negative inhibitor of MCP-1. Previous studies have shown that 7ND protein can suppress osteoclast differentiation in peripheral blood mononuclear cells, suggesting its potential to prevent inflammatory bone destruction. However, whether 7ND protein can inhibit AP-induced osteolysis remains unknown. METHODS: To investigate the effects of 7ND protein on os
