Sedentary behavior as a Gut-Brain metabolic state.

Sedentary behavior (SB) is prevalent in daily life, with a growing number of studies reporting that higher SB exposure is associated with poorer cardiometabolic and brain health. Thus, in line with a broader paradigm shift, SB should be considered as a postural and low-contractile metabolic state, shaped by both total exposure and bout patterning, that is related to, but distinct from, physical activity (PA), rather than simply the absence of the latter. Regarding the gut microbiome, current evidence on the role of SB remains limited and indirect, as human studies have not yet experimentally m…

Peripheral pathway rerouting shapes whole-brain functional network.

Pathway rerouting is critical for severe peripheral nerve injury (PNI), which induces widespread neuroplastic alterations within the central nervous system, yet whether different rerouting approaches differentially affect whole-brain functional networks remains unknown. The present study investigated alterations in brain network organization following different peripheral pathway rerouting after PNI. Adult Sprague-Dawley rats underwent complete left brachial plexus transection and were assigned to nerve repair, end-to-end nerve transfer, end-to-side nerve transfer, or untreated PNI groups. Res…

Decoding brain anatomy from neuronal neighborhoods with MYCEL.

Extracellular recordings carry information about neuronal location, and pooling per-unit predictions across neighboring electrodes improves its readout, but whether this local pooling should be learned is untested. We present MYCEL (Message-passing Yields Cellular Embeddings of Location), a graph neural network whose nodes are spike-sorted units carrying waveform and spike-timing features selected on cell-type ground truth, and whose edges encode recording geometry. On the Allen Brain Observatory Visual Coding Neuropixels dataset, learned message passing outperforms fixed aggregation, with the…

Brain Mechanisms of Lexicality in Bilingual Emotion Processing.

Understanding how native speakers and second language (L2) learners process language differently is fundamental to theories of language acquisition, comprehension, and use. A central debate in this literature concerns the inconsistent findings in emotional prosody processing between native speakers and L2 learners, partly attributable to stimulus lexicality: some studies use real words, whereas others use pseudowords. To resolve this discrepancy, we measured fMRI responses in 15 native speakers and 15 L2 learners while they listened to Mandarin real-word and pseudoword stimuli. Both groups eng

The brain fatigue syndrome: a cross-diagnostic phenomenon.

Fatigue arising from diseases or conditions that affect the brain is well recognized. This form of fatigue, brain fatigue, is not comparable to ordinary tiredness that resolves with rest; rather, it reflects a profound and often long-lasting exhaustion. Many individuals struggle in silence, lacking the energy to advocate for their needs. As a result, they may become socially isolated and unable to participate fully in everyday life, despite intact motivation and desire to engage. This fatigue has a substantial impact on work ability, social functioning, and daily activities, making it a seriou…

Trait-relevant movies amplify neuroticism-associated brain responses.

High neuroticism is a risk factor for mental disorders. Understanding whether individuals share a common foundation in brain function underlying neuroticism is therefore an essential goal of neuroscience. We applied Inter-Subject Representational Similarity Analysis on data from 174 Human Connectome Project participants to investigate if their similarity in neuroticism is reflected in their similarity of fMRI-recorded brain activity during movie watching. To test behavioral personality theories that consider trait expression as dependent on situational context, we examined whether brain-trait …

Circadian biology in brain cancer diagnosis and treatment.

Glioblastoma (GBM), the most common and lethal primary brain cancer, has a median survival of 15 months. Despite repeated efforts over 20 years to introduce life-extending therapies, little has changed in the standard of care. Recent studies have revealed that GBM cells express daily rhythms in gene expression and sensitivity to chemotherapy. In this review, we highlight the role of circadian biology in GBM growth, and its potential applications to GBM diagnosis and treatment. The hallmarks of cancer often show daily variation. We emphasize opportunities to advance our understanding of gliobla…

Generative modeling of complex-valued brain MRI data.

OBJECTIVE: Standard Magnetic Resonance Imaging (MRI) reconstruction pipelines discard phase information captured during acquisition, despite its sensitivity to tissue properties that are not represented by magnitude alone. The aim of this study is to develop a generative framework that jointly models magnitude and phase information in complex-valued brain MRI data and enables the conditional generation of MRI samples representing normal and abnormal brain tissue. APPROACH: The proposed generative framework combines a conditional variational autoencoder, which compresses complex-valued MRI sca…

Deep whole genome sequencing of epileptogenic brain lesions.

OBJECTIVE: Malformations of cortical development and low-grade epilepsy-associated tumors often cause drug-resistant lesional focal epilepsy (LFE) amenable to surgical treatment. Genetic testing of resected brain tissue can provide diagnostic and mechanistic information and may eventually support personalized treatment approaches. However, systematic >300× whole genome sequencing (GS) of bulk epileptogenic lesions, particularly after nondiagnostic deep exome sequencing and array-based genotyping, has not been evaluated. METHODS: Here, we performed deep GS (>300× coverage) on 33 epileptogenic …

Cortical microarchitecture supports preadolescent functional brain network connectivity.

A century ago, Brodmann suggested that local cortical microarchitecture dictates function, yet modern structure- function mapping overwhelmi … vity estimates from functional MRI, in 6,320 children ages 9-11 years from the U.S.-wide Adolescent Brain Cognitive Development (ABCD) Study, we identified several principles of structure-function coupling. First, we found that functional connectivity is associated with one dominant and global pattern of cellular density differences explaining nearly 80 …

Cortex-grounded diffusion models for brain image generation.

Synthetic neuroimaging data can mitigate critical limitations of real-world datasets, including the scarcity of rare phenotypes, domain shifts across scanners, and insufficient longitudinal coverage. However, existing generative models largely rely on weak conditioning signals, such as labels or text, which lack anatomical grounding and often produce biologically implausible outputs. To this end, we introduce Cor2Vox, a cortex-grounded generative framework for brain magnetic resonance image (MRI) synthesis that ties image generation to continuous structural priors of the cerebral cortex. It le…

Substance abuse and inflammation: consequences beyond the brain.

Substance use disorders are traditionally conceptualized as disorders of brain reward circuitry; however, accumulating evidence demonstrates that addictive substances exert profound and persistent effects on central and peripheral immune and inflammatory systems. Opioids, psychostimulants, alcohol, nicotine, cannabinoids, and psychedelics modulate immune signaling across multiple organs, including the brain, gut, liver, lungs, skin, and systemic circulation. In parallel, drugs of abuse remodel neuroimmune communication both centrally and peripherally, altering interactions between neurons, gli…

Transcranial acoustoelectric brain imaging with high spatiotemporal resolution.

Non-invasive neuroimaging has long faced the inherent trade-off between spatial and temporal resolution. Acoustoelectric brain imaging (ABI) holds promise to bridge this gap by combining the spatial precision of focused ultrasound (millimeter-level) with the temporal resolution of electroencephalography (EEG) signals (millisecond-level). However, its transcranial application remains fundamentally challenged by skull-induced wavefront aberration and attenuation. Here, we developed a full ABI system featuring a custom 128-element ultrasound phased array. Our system integrates developed algorithm…

Development of a Brain-Penetrant Nurr1 Agonist Tool.

The ligand-sensing transcription factor nuclear receptor-related 1 (Nurr1), is thought to mediate neuroprotective activity and is implicated in various neurodegenerative diseases. Nurr1 ligands have been developed as tools to capture the receptor's potential in vitro and in vivo, but a dedicated brain-targeting agonist has been lacking. Here, we employed the scaffold of the dihydroorotate dehydrogenase (DHODH) inhibitor and strong Nurr1 activator vidofludimus to develop a descendant with a high brain-to-blood ratio. Scaffold-hopping from cyclopentene to thiophene, enhanced fluorination, and re…

Neuroscience-Inspired Deep Learning Brain-Machine Interface Decoder.

Brain-machine interfaces (BMIs) aim to decode motor intentions from neural activity to enable direct control of external devices. However, most existing decoders rely on monolithic architectures that fail to capture the distinct neural representations of different joint movement directions, limiting their generalizability. In this work, we propose a Single-Direction CNN-LSTM decoder inspired by motor cortex encoding mechanisms, which separately models extension and flexion dynamics through parallel CNN-LSTM branches. Each branch extracts spatial-temporal features from neural spike data and pre…

Radiomics in fetal brain MRI: a narrative review.

Fetal MRI has emerged as a crucial supplement to prenatal ultrasonography in the evaluation of the developing brain and in identifying congenital defects and minor developmental malformations. While fetal brain MRI interpretation has always depended on visual examination of signal properties and morphology, images can provide quantitative information that could be missed or hidden from the human eye. Radiomics allows for characterizing tissue characteristics and heterogeneity by extracting quantitative information from imaging data. In this narrative review, after summarizing the technical fou…
