Exosome-delivered pepDCBLD2 promotes glucose metabolism to induce chemoresistance in colon cancer.

BACKGROUND: Oxaliplatin is a standard treatment for colon cancer, but chemoresistance is a considerable challenge. Dysregulation of circRNAs is involved in cancer and translatomics has found that some circRNAs encode polypeptides. METHODS: Ribosome footprint profiling combined with circRNA sequencing was applied to screen circRNA with coding potential. Immunoprecipitation and mass spectrum were performed to explore the potential mechanism for regulating the downstream pathway. Ultracentrifugation and commercial kits were utilized to extract exosomes. Different kits were used to detect metabol

BRN2 in lineage plasticity: a prostate cancer-focused perspective.

Lineage plasticity is a major mechanism by which prostate cancer adapts to therapeutic pressure, particularly following sustained inhibition of androgen receptor signaling. A central mediator of this process is BRN2, a POU3F2 neural lineage transcription factor that is normally suppressed in AR-dependent luminal prostate epithelium but becomes aberrantly activated during progression to castration-resistant and neuroendocrine (NE) prostate cancer. This review examines how BRN2 connects developmental neurogenesis to therapy-induced tumor reprogramming. We integrate insights from neurodevelopment

Identification of STK35L1 as a potential prognostic biomarker in breast carcinoma, and its expression exhibits high correlation with EGFR activity.

Breast cancer (BC) is the second most prevalent malignancy after lung cancer, and the life expectancy is still very low due to therapeutic resistance and tumor relapse. It is crucial to identify novel biomarkers that can serve as potential therapeutic targets. In TNBC, aberrant activation of EGFR has also been implicated in the development of drug resistance. STK35L1 is a critical regulator of diverse cellular processes, including apoptosis and DNA damage. Notably, STK35L1 promotes drug resistance and regulates glycolysis and apoptosis through AKT signaling. The oncogenic role of STK35L1 is es

BCL11B predetermines a persister state in breast cancer that is reversed by TNFα.

The frequent development of chemoresistance in cancer presents a major clinical challenge, yet the underlying causes of heterogeneous drug responses remain largely elusive. Here, we systematically assessed the cellular differentiation status of human breast cancer cells using single-cell atlases and identified a distinct population of immature basal-like cancer cells marked by BCL11B. Notably, higher levels of BCL11B+ cancer cells are significantly associated with early relapse in patients with breast cancer who received chemotherapy. Functioning as a central regulator, B-cell lymphoma/leukemi

Integrated analysis of isorhamnetin-associated targets in chronic pancreatitis and pancreatic cancer: immune-infiltration associations and antitumor effects in pancreatic cancer cells.

OBJECTIVE: Chronic pancreatitis (CP) creates an inflammatory pancreatic microenvironment and is an established risk factor for pancreatic cancer (PC). However, immune-inflammatory links between CP and PC remain unclear. Isorhamnetin (ISO), a natural flavonoid with anti-inflammatory and antitumor activities, may influence pancreatic inflammatory signaling. This study investigated ISO-associated molecular networks shared by CP and PC, examined the relationships between prioritized targets and transcriptome-estimated immune-cell infiltration, and evaluated the effects of ISO in pancreatic cancer

Elevated miR-103b in exosomes derived from brain-metastatic triple-negative breast cancer cells remodels the brain pre-metastatic niche (PMN).

BACKGROUND: Triple-negative breast cancer (TNBC) is a highly aggressive breast cancer with high brain metastatic (BM) potential. Tumor-derived exosomes are implicated as key modulators during the formation of the pre-metastatic niche (PMN). However, the regulation of TNBC-BM-derived exosomes on brain PMN remains enigmatic. METHODS: The morphology and uptake of exosomes were identified using transmission electron microscopy and live-cell imaging, respectively. Differentially secreted miRNAs and mRNA were identified using high-throughput sequencing. The permeability of the blood‒brain barrier (

ncRNAs as Key Regulators in Gastric Cancer: From Molecular Subtyping to Therapeutic Targets.

Gastric cancer (GC) poses a major global health challenge, underscoring the need for advanced diagnostic and therapeutic approaches. Non-coding RNAs (ncRNAs), including microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs), have emerged as pivotal regulators in GC, with their dysregulated expression driving key processes such as tumorigenesis, metastasis, immune evasion, and chemoresistance. The functional diversity of ncRNAs across different GC molecular subtypes highlights their potential as biomarkers for improved subtype classification and patient stratification.

ESRP1 exerts anti-tumor role by promoting ferroptosis in diffuse-type gastric cancer.

Diffuse-type gastric cancer (DGC) accounts for approximately one-third of gastric cancer cases, but it is clinically more aggressive and associated with poorer survival prognosis compared to intestinal-type gastric cancer (IGC). Our study revealed significantly reduced ESRP1 expression in DGC patients relative to IGC counterparts, with its high expression positively correlating with DGC patient prognosis. Both in vitro and in vivo experimental models demonstrated that ESRP1 has the ability to hinder the malignant progression of DGC. Mechanistically, ESRP1 interacts with DHCR7, a ferroptosis-pr

CLIC1 regulates cellular oxidative stress and gastric cancer progress by enhancing ACOX1 ubiquitination.

Oxidative stress is a two-edged sword for tumors. However, how oxidative stress influences tumor progression remains unclear. We determined that chloride intracellular channel 1 (CLIC1) accelerates gastric cancer progression by enhancing acyl-CoA oxidase 1 (ACOX1) ubiquitination and increasing cellular oxidative stress. CLIC1 overexpression increased cellular oxidative stress. Furthermore, our results demonstrate that ACOX1 is an oxidative stress-related gene that is closely related to CLIC1. Functionally, downregulating ACOX1 promoted gastric cancer cell proliferation, migration, and invasion

The Clinical Prognostic Value of Lactylation-Regulated Proteins in Gastric Cancer.

Gastric cancer (GC) is a leading cause of cancer-related mortality globally. Histone lactylation, an emerging post-translational modification, holds promise as a therapeutic target and prognostic biomarker, though its expression patterns and clinical relevance in GC are underexplored. Pan-lactylation was quantified by IHC in tumor/adjacent normal tissues and validated via Western blot in GC/normal cell lines. Systematic profiling of lactylome and proteome of tumor-normal pairs (n = 3) identified 127 lactylation-regulated genes. Four prognostic models (stepCOX, LASSO, RSF, GBM) were built using

MAP4 kinase-regulated reduced CLSTN1 expression in medulloblastoma is associated with increased invasiveness.

De-regulated protein expression contributes to tumor growth and progression in medulloblastoma (MB), the most common malignant brain tumor in children. MB is associated with impaired differentiation of specific neural progenitors, suggesting that the deregulation of proteins involved in neural physiology could contribute to the transformed phenotype in MB. Calsynthenin 1 (CLSTN1) is a neuronal protein involved in cell-cell interaction, vesicle trafficking, and synaptic signaling. We previously identified CLSTN1 as a putative target of the pro-invasive kinase MAP4K4, which we found to reduce CL

The T-Box Transcription Factors TBX2 and TBX3 Are Molecular Targets of Piroctone Olamine in the Treatment of Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) has a poor 5-year survival rate of < 10% and its incidences are continuously rising worldwide. This highlights an urgent need for effective therapies to reduce its burden. Repurposing commercially available non-cancer drugs that inhibit key drivers of PDAC may facilitate the rapid identification of effective drugs. In PDAC, the expression of the TBX2 and TBX3 transcription factors correlates with metastasis and poor patient survival. This study showed that when TBX3 was depleted in 2D and 3D PDAC cell culture models, the cells underwent senescence and ha

Unveiling the impact of OVOL1 on prognosis, immune microenvironment, and proliferation in pancreatic cancer.

OVO-like protein 1 (OVOL1) has been implicated in the progression of various human cancers; however, the prognostic and immunological significance, as well as its biological role in pancreatic cancer, are unknown. In this study, by analyzing multiple patient cohorts, OVOL1 was found to be elevated in pancreatic cancer, which was associated with poor overall survival (OS), disease-specific survival (DSS), and progression-free interval (PFI). Two nomogram survival models for predicting 1-, 2-, and 3-year OS and DSS with excellent accuracy were constructed, respectively. Tumors with elevated OVOL

Cuproptosis-related lncRNAs and genes: Potential markers for glioblastoma prognosis and treatment.

Despite the availability of various treatment options, glioblastoma (GBM) remains an extremely aggressive form of glioma with a poor prognosis. In recent studies, regulatory cell death (RCD) has been identified as an effective mechanism to suppress glioma. Cuproptosis, caused by intracellular copper, is a novel RCD process that affects chemotherapy efficacy and glioma progression; however, the precise function of cuproptosis-related lncRNAs (CRLs) and cuproptosis-related genes (CRGs) in GBM remains uncertain. To determine whether CRLs and CRGs have prognostic significance, a GBM cohort in TCGA

Epigenetic mechanisms in non-small cell lung cancer therapy and chemoresistance.

Epigenetic mechanisms play a crucial role in cancer development and progression, encompassing DNA methylation, chromatin organization, histone modifications, and noncoding RNA regulation. Lung cancer is a major cause of cancer-related deaths worldwide, with non-small cell lung cancer (NSCLC) accounting for approximately 85 % of all cases. Despite advances in detection and treatment, the prognosis for NSCLC patients remains poor, largely attributable to the development of chemoresistance. Chemoresistance is a key driver of cancer recurrence and metastasis. Targeting epigenetic modifications eme
