Ancestry-informative regulatory variants at KCNB1 modulate adipogenesis and body mass index.

INTRODUCTION: Obesity and related metabolic disorders represent a major global health burden, yet their genetic determinants remain incompletely characterized, particularly in non-European populations. We aimed to identify body mass index (BMI)-associated loci in an admixed Brazilian population and to functionally characterize ancestry-enriched variants contributing to obesity risk. METHODS: We conducted a genome-wide association study (GWAS) of BMI in 1,079 admixed Brazilian individuals. Significant and suggestive loci were evaluated using integrative analysis, including epigenomic annotatio…

The causal relationship between polycystic ovary syndrome and structural changes in brain: a Mendelian randomization study.

The impact of sex hormones on the brain has been widely recognized. Polycystic ovary syndrome (PCOS) is frequently accompanied by abnormal hormone. Furthermore, psychiatric disorders are among the most common comorbidities associated with PCOS. However, whether PCOS induces alterations in brain structure remains unclear. Mendelian randomization (MR) was conducted to determine this causal association and pinpoint specific brain regions prone to PCOS. Genome wide association study (GWAS) data from the FinnGen database, for PCOS phenotypes from 39,004 PCOS patients and 731,830 controls were inclu

Transcriptome-wide association studies implicate RCC1 and PHACTR4 in prostate cancer survival.

While prostate cancer (PrCa) is highly heritable, the genes associated with PrCa survival after diagnosis remain poorly understood. We aimed to identify genes associated with PrCa-specific survival through transcriptome-wide association studies (TWAS) using genetic predictors of gene expression in the prostate. We used the Transcriptome-Integrated Genetic Association Resource (TIGAR) to train expression prediction models separately using normal prostate, primary tumor, and metastatic tumor tissues. We performed TWAS using these models in data from the Malmö Diet and Cancer (MDC) study (discove

Multiple metabolic factors and kidney dysfunction: Causal evidence and translational insights from a mendelian randomization study.

Chronic kidney disease is a major global public health burden. This study investigated the causal effects of systolic blood pressure (SBP), apolipoprotein B (ApoB), and serum urate on renal function and albuminuria, and explored potential mechanistic implications. Two-sample Mendelian randomization (MR) analyses were conducted using large-scale genome-wide association study summary statistics from European populations. Univariable MR estimated total causal effects on estimated glomerular filtration rate (eGFR) and albuminuria. Multivariable MR assessed independent effects of correlated exposur…

Leucine, autoimmune diseases, and circulating mediators shape the causal landscape of lymphoid malignancies.

INTRODUCTION: While genetic abnormalities underlying lymphoid malignancies (LM) are well-characterized, the environmental and metabolic determinants driving LM remain largely unexplored. Autoimmune diseases are known risks, yet the specific circulating mediators and potential protective dietary factors linking these conditions to LM require systematic elucidation. OBJECTIVES: This study aimed to comprehensively screen for causal risk factors of LM, identify the metabolic mediators bridging autoimmune diseases and LM, and validate the therapeutic potential of leucine as a protective factor. Multivariate genetic analysis reveals three distinct pathological dimensions in musculoskeletal disorders.

The substantial overlap in symptoms and pathology across musculoskeletal disorders underscores the need to investigate their shared genetic mechanism. Using multivariate genetic modeling, we identify three distinct pathological factors, including a degenerative musculoskeletal disorder factor, a bone mass factor, and an autoimmune disorder factor, which collectively explain 48.27% of the total genetic variance in musculoskeletal disorders. Genome-wide association analyses of these factors identify 795 genomic risk loci, 136 of which are novel, along with 273 candidate genes, 20 of which intera

The causal effect of juvenile idiopathic arthritis on IgA nephropathy: A Mendelian randomization study.

IgA nephropathy (IgAN) has been documented in patients with various comorbidities. Previous observational studies showed an association between juvenile idiopathic arthritis (JIA) and IgAN. The aim of this study was to determine whether there is a causal effect of JIA on the risk of IgAN using a 2-sample Mendelian randomization (MR) approach. A 2-sample MR analysis was conducted to elucidate the potential causal relationship between JIA and IgAN. Summary-level data from genome-wide association studies in the European population were utilized, including IgAN (5556 cases and 21,178 controls) and

Shared genetic architecture and therapeutic targets across paediatric immune-mediated diseases.

OBJECTIVES: Paediatric-onset immune-mediated inflammatory diseases (IMIDs), including juvenile idiopathic arthritis and related rheumatic diseases, remain genetically undercharacterised. We aimed to define shared and category-specific genetic architecture across paediatric IMIDs, compare signals with adult IMIDs, and identify therapeutic opportunities. METHODS: We analysed 24 paediatric IMIDs classified as autoimmune, polygenic-autoinflammatory, mixed-pattern, or allergic. Genome-wide association analyses included 18,086 cases and 131,019 controls of European ancestry. We estimated single nuc

The causal association between plasma amino acid levels and abdominal aortic aneurysm: insights from Mendelian randomization and an experimental validation.

Abdominal aortic aneurysm (AAA) is a severe vascular condition with a high mortality rate upon rupture. Emerging evidence suggests that metabolic factors, particularly plasma amino acids, may influence AAA risk, though specific causal roles remain unclear. This study investigates the causal relationship between plasma amino acids and AAA using Mendelian randomization (MR) and in vivo validation. A bidirectional two-sample MR analysis was conducted using genome-wide association study (GWAS) data. Genetic variants associated with amino acid levels and AAA served as instrumental variables, with i…

Association Between Polymorphisms of 4 Common Genes and High Myopia Risk: A Comprehensive Analysis.

BACKGROUND Genome-wide association studies have been suggested single-nucleotide polymorphisms (SNPs) can influence susceptibility to high myopia (HM). To investigate the associations of multiple SNPs of 4 common genes and HM, we collected all related articles about these 4 common SNPs and risk of HM. MATERIAL AND METHODS PubMed and Wanfang databases were searched for articles published until Dec 10, 2025 using the keywords 'GJD2' or 'ZC3H11B' or 'MMP1' or 'MMP9', 'polymorphism' and 'myopia' or 'shortsightedness'. Odds ratios and 95% confidence intervals were used to examine the association be

Angiographic Burden of Coronary Atherosclerosis Partially Mediates the Association Between ASCVD Risk Factors and Outcomes.

BACKGROUND: Coronary artery disease (CAD) is a major contributor to cardiovascular morbidity (including myocardial infarction and heart failure) and mortality. Although the burden of CAD (number and degree of coronary artery stenosis) has been observationally linked to these outcomes, the causal contribution and independence from traditional cardiovascular risk factors have been poorly defined. METHODS: We developed a polygenic risk score for angiographic CAD burden using data from the VA Million Veteran Program (n=41 507) and validated this score using data from the Penn Medicine Biobank (n=

Deep learning aging marker from retinal images unveils sex-specific clinical and genetic signatures.

Retinal fundus images offer a non-invasive window into systemic aging. Here, we fine-tune a foundation model (RETFound) to predict chronological age from color fundus images in 71,343 participants from the UK Biobank, achieving a mean absolute error of 2.85 years. The resulting retinal age gap, i.e. the difference between predicted and chronological age, is associated with cardiometabolic traits, inflammation, cognitive performance, all-cause mortality, dementia, cancer, and incident cardiovascular disease. Genome-wide analyses identify genes related to longevity, metabolism, neurodegeneration
