We performed sep … metabolic dysregulation may increase POAG risk prior to T2D diagnosis. The T2D risk variant in the melatonin receptor 1B gene (MTNR1B) predicted risk of POAG independently of T2D status, indicating possible pleiotropic physiological functions of melatonin, but instrumental variable effect estimates were significant even excluding MTNR1B variants. To our kn …

Pharmacological benefits of agomelatine and vanillin in experimental model of Huntington's disease.

Huntington's disease (HD), a devastating neurodegenerative disorder, is characterized by progressive motor dysfunction, emotional disturbances, dementia, weight loss, depression. Melatonin receptors are widely expressed in the central nervous system. Vanilloids are also valuable as pharmacological tools for investigating neurobiology. This study investigates the utility of agomelatine, a dual agonist of MT₁ and MT₂ melatonin receptor as well as vanillin, a selective agonist of TRPV₁ (vanilloid receptor) in 3-nitropropionic acid (3-NPA) induced experimental HD in rats. Locomotor activity (Actop…

Effects of pinealectomy on wool growth and wool follicle density in merino sheep.

There is evidence to indicate that pinealectomy may enhance wool growth in the sheep. The aim of this study was to determine the effect of pinealectomy on wool growth and wool follicle density in Merino sheep. Castrated Merino rams (4 months old) were either pinealectomized (P), sham-pinealectomized (S) or not treated (C). Wool growth on mid-side patches was measured every 4 weeks and follicle density was monitored in skin biopsies collected before treatment and at regular intervals for 60 weeks. Venous blood samples were taken on each of these occasions for prolactin analysis. Melatonin conce…

Pinealectomy in Rats.

The pinealectomy technique consists of the surgical removal of the superficial pineal gland. This procedure allows the ablation of circulating indoles produced by this gland. Withdrawal of systemic melatonin, a pineal hormone, affects animal circadian rhythms and induces several physiological changes that are the subject of many investigations. In this chapter, we describe the pinealectomy protocol adapted to rats. We describe the animal placement on the stereotaxic fixation system, and the procedure for the pineal gland removal and animal recovery from surgery.

Maternal pinealectomy increases depressive-like responses in Siberian hamster offspring.

This study investigated the effect of maternal pinealectomy and postnatal pinealectomy on affective responses. Siberian hamsters were born to either pinealectomized or sham-operated dams and then underwent pinealectomy or a sham operation. Maternal pinealectomy increased depressive-like responses of offspring in the forced swim test. Maternal pinealectomy increased rearing behaviour and postnatal pinealectomy increased locomotor behaviour in the open field test. These results suggest that prenatal melatonin organizes adult affective responses.

APP SUMOylation prevents BACE1 cleavage of APP and increases BACE1 degradation to promote the nonamyloidogenic pathway.

Amyloid precursor protein (APP) plays a key role in the pathogenesis of Alzheimer’s disease (AD). APP undergoes different posttranslational … SUMO1ΔGG mutant vector transduction to APP/PS1 mice produces an opposite effect for these measures. Melatonin increases Ubc9 expression and enhances APP SUMOylation. In addition, blockade of APP phosphorylation at Thr-668 facilitates APP SUMOylation. These results together suggest that APP SUMOylation promotes the nonamyloidogenic processing of APP and func …

Comparison of milk production and endocrine profiles of dairy cows exposed to either white light-emitting diode or induction lighting.

White light-emitting diodes (WLEDs) are characterized by their high blue light intensity, whereas induction lighting (IL) emits lower levels … cids concentration tended to be higher for the WLED than for the IL (P = 0.09). In addition, plasma melatonin and cortisol concentrations were higher (P < 0.01) for the WLED group than for the IL group. These findings suggest that differences in light wavelength between WLED and IL affect melatonin and cortisol secretion and may also impact lipid metabolism …

Sleep and stress in the acute phase of concussion in youth.

This study sought to address the complex interplay between both biological and psychological perceptions of stress and sleep in the acute stages following a mild traumatic brain injury. A secondary goal was to identify potential targets for intervention. Eleven acutely injured youth (mean age 12 years) were studied at home with overnight actigraphy, salivary cortisol and melatonin assays, and subjective ratings of stress and fatigue (injured group). Nine matched control youth also were assessed (control group). Results suggested longer sleep latencies (time to fall asleep) and higher levels of…

[Agomelatine: the first "melatoninergic" antidepressant].

The intimate relationship between depressive disorders and biological rhythm disturbances has long been known. One major component in diurnal rhythm regulation in mammals, like humans, is the melatonin axis and its close connections with serotonergic and noradrenergic structures. In major depression various functional anomalies have been shown in all of these systems. Agomelatine is a novel antidepressant with direct agonist activity at the melatonin MT1 and MT2 receptors, and a selective antagonist action at the serotonin 5HT2C receptor. It has no measurable affinity to any other known recept…

Morphometry of the pineal gland in overweight individuals.

While obesity is an ever increasing problem in today's world, numerous facts suggest that its side-effects continue to be underestimated. Recent studies link obesity with pineal gland hormone and melatonin. The aim of this study was to determine differences in morphological attributes of the pineal gland in normal and overweight people in terms of pineal width and volume. A lower pineal width and volume in overweight individuals stated in the study may suggest a different excretory activity of the pineal gland.

Does a centralized clock for ageing exist?

It is proposed that a centralized clock controlling ageing is located in the pineal gland with the calcification process occurring there providing a highly accurate bio-inorganic timing mechanism and the secreted melatonin carrying a signal to all cells in the organism. An explicit programme of data gathering and experiments suitable for the falsification of the proposal, which is consistent with presently known anatomical and physiological facts, is presented. The underlying motivation comes from evolutionary biology and the invariance, that is the allometry, of life expectancy curves.

Chronotype-based radiotherapy in localised prostate cancer: protocol for a prospective cohort integrating MCTQ and salivary DLMO.

We present a prospective observational cohort protocol evaluating whether chronotype-based radiotherapy timing reduces treatment-related toxicities in localised prostate cancer. Individual circadian phase is characterised using the Munich ChronoType Questionnaire and salivary dim-light melatonin onset. The primary endpoint is late genitourinary toxicity grade ≥2 at 12 months, with secondary endpoints including gastrointestinal toxicity and patient-reported outcomes. The protocol is conducted at IPO Porto and designed for multi-site adoption.

Circadian disruption as a driver and target in neurodegenerative diseases: from molecular mechanisms to chronotherapeutic strategies.

The Circadian System is a complex network of coordinated clocks that regulates the organism’s internal clock in synchronisation with the outside world. These rhythms are controlled by genetically controlled positive and negative transcriptional-translational feedback loops (TTFL) that generate 24-hour oscillations in the protein level and mRNA of core circadian components. Circadian disruption is recognised as a significant contributor to the molecular pathogenesis of neurodegenerative illnesses, as disease-specific alterations in clock gene expression and melatoninergic signalling have been i…
