Melatonin Exerts Anticancer Effects in Human Tongue Squamous Cell Carcinoma Cells by Promoting Autophagy.

BACKGROUND/AIM: The global prevalence of head and neck squamous cell carcinoma (HNSCC) remains high, and its prognosis poor. We investigated the anticancer effects of melatonin in human tongue squamous cell carcinoma cells (SCC-25) and its mechanisms of action. MATERIALS AND METHODS: MTT assay was used to determine cell viability. To assess the effects of melatonin on SCC-25 cell metastasis, we conducted cell formation, wound healing, transwell migration and invasion assay. Western blot analysis was performed to measure the levels of autophage marker proteins. RESULTS: We found that melatoni

Current concepts in management of oral cancer--surgery.

Oral cancer is the sixth most common cancer worldwide, with a high prevalence in South Asia. Tobacco and alcohol consumption remain the most dominant etiologic factors, however HPV has been recently implicated in oral cancer. Surgery is the most well established mode of initial definitive treatment for a majority of oral cancers. The factors that affect choice of treatment are related to the tumor and the patient. Primary site, location, size, proximity to bone, and depth of infiltration are factors which influence a particular surgical approach. Tumors that approach or involve the mandible re

Photodynamic priming with Vitamin D and ALA-based PDT induces intratumoral immune cell recruitment and signaling pathway activation in cutaneous squamous cell carcinoma.

Photodynamic therapy (PDT) is an effective non-invasive treatment for epithelial pre-cancers, yet its efficacy in cutaneous squamous cell carcinoma (SCC) is limited by the immunosuppressive microenvironment of SCC. Vitamin D (VitD) has emerged as a potential neoadjuvant to enhance photodynamic priming in several cancers, but its immunologic effects in SCC remain poorly defined. Here, we investigated how VitD pretreatment modulates local and systemic immune responses to aminolevulinic acid-based PDT in two immunocompetent murine SCC models. VitD combined with PDT significantly amplified hallmar

Basaloid adenosquamous carcinoma mimicking adenoid cystic carcinoma (BAC MAC): A novel subtype of head and neck squamous cell carcinoma.

Head and neck squamous cell carcinoma (SCC) has many subtypes, including basaloid (BSCC) and adenosquamous carcinoma (ASC). We describe a surface-derived carcinoma with features of both BSCC and ASC, along with frequent myoepithelial differentiation. Five cases were retrieved, slides were reviewed and results tabulated. DNA- and RNA-sequencing were performed in 4 and 5 cases, respectively. Patients were 4 men and 1 woman, from 55 to 78 years (mean, 68.6). Sites of origin were 2 hypopharynx, 2 oral cavity and 1 oropharynx. All cases demonstrated basaloid morphology and duct formation. Squamous

DNA Methylation Signatures of Upper Aerodigestive Tract Squamous Cell Carcinoma and Sarcomatoid Squamous Cell Carcinoma Are Similar: A Potential Novel Diagnostic Tool for the Diagnosis of Sarcomatoid Squamous Cell Carcinoma.

PURPOSE: Upper aerodigestive tract squamous cell carcinoma (UATSCC) is often easy to recognize in its conventional form. However, a subset of tumors loses histologic evidence of squamous differentiation and, instead, become sarcomatoid (spindled). These cases may be difficult or impossible to appropriately classify on biopsy material, even with immunohistochemistry. DNA methylation has emerged as a tool to help in classifying undifferentiated malignancies. We sought to explore whether conventional and sarcomatoid UATSCC harbor the same epigenetic signature. METHODS: Cases of sarcomatoid UATSC

Multi-omics analysis reveals the crosstalk of epigenetic regulatory networks in cutaneous squamous cell carcinoma progression.

BACKGROUND: Cutaneous squamous cell carcinoma (cSCC) is the second most common form of non-melanoma skin cancer (NMSC), with a steadily increasing global incidence, especially in populations with prolonged ultraviolet (UV) exposure. Advanced or metastatic cSCC carries a poor prognosis, while the application of targeted therapies and immunotherapies remains in the exploratory stage, due to limited understanding of the underlying mechanisms of cSCC occurrence and progression. Epigenetic dysregulation plays important roles in the progression of cSCC. However, how multi-dimensional regulatory netw

The Prognostic Role of Pre-Treatment Neutrophil-to-Lymphocyte Ratio in an Asian Cohort of Patients with Oropharyngeal Squamous Cell Carcinoma.

PURPOSE: The neutrophil-to-lymphocyte ratio is a simple biomarker that reflects the balance between the systemic inflammatory and immunity status. Here we investigate the prognostic role of pre-treatment neutrophil-to-lymphocyte ratio (NLR) in an Asian cohort of oropharyngeal squamous cell carcinoma (OPSCC) patients. METHODS: A retrospective review of OPSCC patients from a tertiary institution was conducted. The NLR was calculated from the haematological specimen taken within a month before treatment. Survival rates were estimated via the Kaplan-Meier method, and Cox proportional hazards regr

Integrin α11 is overexpressed by tumour stroma of head and neck squamous cell carcinoma and correlates positively with alpha smooth muscle actin expression.

BACKGROUND: Cancer-associated fibroblasts (CAFs) were shown to be important for tumour progression in head and neck squamous cell carcinomas (HNSCCs). Their heterogeneity and lack of specific markers is increasingly recognized. Integrin α11 was recently shown to be expressed by CAFs and might serve as a specific CAF marker. AIM: To investigate integrin α11 expression and its correlation with the expression of a well-known marker of CAF, alpha smooth muscle actin (α-SMA), in HNSCC. METHODS: Fresh frozen (FF) and formalin-fixed paraffin-embedded (FFPE) samples from healthy volunteers (n = 24),

Reconstruction of full-thickness buccal defects with submental island flap.

Our goal was to introduce the application of submental island flap in reconstructing through-and-through cheek defects. From January 2009 to January 2013, 7 patients (5 men and 2 women) with full-thickness buccal defects due to tumor resection received submental flap reconstruction at the Affiliated Tumor Hospital of Zhengzhou University; surgical procedure and success rate as well as functional results were described. Distal partial necrosis occurred in 1 flap, but all flaps survived. All patients were capable of maintaining a regular oral diet, and no patients complained of an inability to e

Tunicamycin-induced endoplasmic reticulum stress inhibits chemoresistance of FaDu hypopharyngeal carcinoma cells in 3D collagen I cultures and in vivo.

The prognosis in patients with advanced head and neck squamous cell carcinoma (HNSCC) is widely affected by the resistance to chemotherapy. As a culture scaffold, collagen I was showed to promote CSC (cancer stem cell) properties of cancer cells which could be used as in vitro models to study the chemoresistance in HNSCC. Endoplasmic reticulum (ER) stress is a cellular stress condition which could affect tumor progression and promote the anti-tumor effects of certain drugs. However, the impact of ER stress on collagen I induced CSC properties and chemoresistance of HNSCC cells has not been add

LncOCMRL1 promotes oral squamous cell carcinoma growth and metastasis via the RRM2/EMT pathway.

BACKGROUND: Long noncoding RNAs (lncRNAs) are widely involved in cancer development and progression, but the functions of most lncRNAs have not yet been elucidated. Metastasis is the main factor restricting the therapeutic outcomes of various cancer types, including oral squamous cell carcinoma (OSCC). Therefore, exploring the key lncRNAs that regulate OSCC metastasis and elucidating their molecular mechanisms will facilitate the development of new strategies for effective OSCC therapy. METHODS: We analyzed the lncRNA expression profiles of tumor tissues from OSCC patients with and without ce

LINC00629, a KLF10-responsive lncRNA, promotes the anticancer effects of apigenin by decreasing Mcl1 stability in oral squamous cell carcinoma.

Apigenin, a naturally occurring flavonoid, is known to exhibit antitumor activity in many cancers. However, the regulatory mechanism of apigenin and the long noncoding RNAs (lncRNAs) altered upon apigenin treatment in oral squamous cell carcinoma (OSCC) remain unclear. In this study, we found that LINC00629 was significantly upregulated in response to apigenin treatment. Upregulated LINC00629 enhanced the growth-suppressive and proapoptotic effects of apigenin on OSCC cells by interacting with Mcl1 and facilitating its degradation. Subsequently, our data indicated that KLF10, an important tran

Cysteine proteases in oral potentially malignant disorders: Molecular drivers, diagnostic signatures, and therapeutic opportunities.

Cysteine proteases are crucial enzymes regulating protein turnover, apoptosis, signalling, and tissue remodelling, and have emerged as important regulators of the oral epithelial microenvironment with increasing evidence implicating them in the progression of oral potentially malignant disorders (OPMDs) and their potential contribution to malignant transformation toward oral squamous cell carcinoma (OSCC). Dysregulated expression of these enzymes influences key molecular pathways, notably NF-κB, MAPK, protease-activated receptor (PAR) signalling, and apoptotic cascades, although the extent to

Natural Compounds for the Treatment of Cutaneous Squamous Cell Carcinoma: A Systematic Review.

Cutaneous squamous cell carcinoma (cSCC) is one of the most common non-melanoma skin cancers worldwide. Although surgery and adjuvant therapies are often effective, the treatment of high-risk or advanced lesions remains challenging due to recurrence, resistance, toxicity, and limited long-term control. Natural compounds have, therefore, gained interest as multi-target agents for cancer prevention and treatment. This systematic review aimed to evaluate the antitumoral activity of natural compounds against cSCC. A systematic literature search was conducted following PRISMA 2020 guidelines. Sixty

COPD-Associated PANoptosis Genes Predict Prognosis and Chemotherapy Sensitivity in Lung Squamous Cell Carcinoma.

Chronic obstructive pulmonary disease (COPD) is a progressive inflammatory disease that increases the risk of lung squamous cell carcinoma (LUSC). PANoptosis integrates pyroptosis, apoptosis, and necroptosis, but the relationship between COPD-associated PANoptosis genes and LUSC prognosis remains unclear. In this study, we first derived COPD-associated differentially expressed genes from GSE57148 by comparing COPD lung tissues with normal lung tissues and intersected these genes with a curated PANoptosis-associated gene list. The resulting gene set was then applied to TCGA-LUSC, GSE30219, and

Immunotherapy for locally advanced head and neck squamous cell carcinoma: A review.

The standard of care (SoC) for locally-advanced (LA) head and neck squamous cell carcinoma (HNSCC) has remained relatively unchanged for decades, usually consisting of multimodal treatment that can include surgery, radiotherapy, and systemic therapy. Survival rates are low, so there is a key unmet need to improve clinical outcomes. Immune checkpoint inhibitors (ICIs), including anti-programmed cell death protein-1 (PD-1) agents, redefined the SoC for recurrent/metastatic HNSCC, and based on recent positive outcomes in Phase III studies, the SoC for resectable LA-HNSCC has evolved to include IC

Robotic sleeve lobectomy with bronchoplasty for primary squamous cell carcinoma of the left upper lobe.

Sleeve lobectomy with bronchoplasty is a complex but lung-preserving operation, often requiring open thoracotomy. Robotic-assisted thoracoscopic surgery offers enhanced visualization and instrument dexterity, enabling such procedures to be performed with minimal invasiveness. We report a case of a 75-year-old man with squamous cell carcinoma located in the lingual segment of the left upper lobe, which invaded to the entrance of left upper lobe bronchus. The tumour was staged as cT2aN0M0. Due to its central location, robotic left upper sleeve lobectomy with bronchoplasty was selected. A five-po
