Predicting Modifiers of Genotype-Phenotype Correlations in Craniofacial Development.

Most human birth defects are phenotypically variable even when they share a common genetic basis. Our understanding of the mechanisms of this variation is limited, but they are thought to be due to complex gene-environment interactions. Loss of the transcription factor Gata3 associates with the highly variable human birth defects HDR syndrome and microsomia, and can lead to disruption of the neural crest-derived facial skeleton. We have demonstrated that zebrafish gata3 mutants model the variability seen in humans, with genetic background and candidate pathways modifying the resulting phenotyp

Neonatal brain injury influences structural connectivity and childhood functional outcomes.

Neonatal brain injury may impact brain development and lead to lifelong functional impairments. Hypoxic-ischemic encephalopathy (HIE) and congenital heart disease (CHD) are two common causes of neonatal brain injury differing in timing and mechanism. Maturation of whole-brain neural networks can be quantified during development using diffusion magnetic resonance imaging (dMRI) in combination with graph theory metrics. DMRI of 35 subjects with CHD and 62 subjects with HIE were compared to understand differences in the effects of HIE and CHD on the development of network topological parameters a

CDKL5 Deficiency Augments Inhibitory Input into the Dentate Gyrus That Can Be Reversed by Deep Brain Stimulation.

Cognitive impairment is a core feature of cyclin-dependent kinase-like 5 (CDKL5) deficiency, a neurodevelopmental disorder characterized by early epileptic seizures, intellectual disability, and autistic behaviors. Although loss of CDKL5 affects a number of molecular pathways, very little has been discovered about the physiological effects of these changes on the neural circuitry. We therefore studied synaptic plasticity and local circuit activity in the dentate gyrus of both Cdkl5-/y and Cdkl5+/- mutant mice. We found that CDKL5 haploinsufficiency in both male and female mice impairs hippocam

Transcriptome Profiling Identifies Ribosome Biogenesis as a Target of Alcohol Teratogenicity and Vulnerability during Early Embryogenesis.

Fetal alcohol spectrum disorder (FASD) is a leading cause of neurodevelopmental disability. Individuals with FASD may exhibit a characteristic facial appearance that has diagnostic utility. The mechanism by which alcohol disrupts craniofacial development is incompletely understood, as are the genetic factors that can modify individual alcohol vulnerability. Using an established avian model, we characterized the cranial transcriptome in response to alcohol to inform the mechanism underlying these cells' vulnerability. Gallus gallus embryos having 3-6 somites were exposed to 52 mM alcohol and th

Fasting plasma triglycerides predict the glycaemic response to treatment of type 2 diabetes by gastric electrical stimulation. A novel lipotoxicity paradigm.

BACKGROUND: Non-stimulatory, meal-mediated electrical stimulation of the stomach (TANTALUS-DIAMOND) improves glycaemic control and causes modest weight loss in patients with Type 2 diabetes who are inadequately controlled on oral anti-diabetic medications. The magnitude of the glycaemic response in clinical studies has been variable. A preliminary analysis of data from patients who had completed 6 months of treatment indicated that the glycaemic response to the electrical stimulation was inversely related to the baseline fasting plasma triglyceride level. METHOD: An analysis of 40 patients wh

Wireless, skin-attachable patch with 3D liquid metal electrodes for vagus nerve stimulation in depression.

Current nonsurgical vagus nerve stimulation (VNS) approaches often suffer from broad current spread, high electrode-skin impedance, and unintended stimulation of surrounding tissues, which can reduce efficacy and cause side effects. Here, we present a wirelessly operable, skin-attachable VNS patch with three-dimensional liquid-metal electrodes that mimic the low modulus of biological tissues, developed as a minimally invasive platform for investigating the biological mechanisms underlying VNS in a mouse model of depression. The soft, low-modulus three-dimensional liquid metal electrodes gently

Activity and State-Dependent Modulation of Salt Taste Behavior Via Pharyngeal Neurons in Drosophila melanogaster.

Sodium, which is present in NaCl (Sodium Chloride), is a vital nutrient required for numerous physiological processes. In animals, including Drosophila, low-salt concentrations induce attraction and high-salt concentrations trigger aversive behavior. Although the analysis of low and high salt concentrations in Drosophila has been described at the cellular level, the mechanisms governing high salt consumption and taste modulation remain unclear. Our study examined the neural basis of high NaCl consumption in adult Drosophila, focusing on how dietary adaptation influences salt acceptance. Our fi

Cephalic phase response in systemic glucose metabolism: How taste triggers glucose homeostasis.

BACKGROUND: Cephalic phase responses (CPRs) are anticipatory and conditioned responses elicited by the stimulation of food-related cues in the oral and nasal cavities. By initiating autonomic and endocrine reactions before absorption of nutrients, CPRs can blunt acute-postprandial hyperglycemia. Taste-related information is critical for triggering CPRs, but the neural mechanisms underlying taste-driven CPRs remain incompletely elucidated. HIGHLIGHT: Current evidence supports a model in which taste-driven CPRs arise from coordinated processing across peripheral taste receptors, the rostral com

Dual origins for neural cells during development of the Clytia planula larva.

Adult hydrozoan cnidarians undergo extensive tissue turnover, generating neural cell types that include nematocytes (stinging cells) and gland cells from interstitial stem cells (i-cells) expressing stemness proteins such as Piwi and Nanos. The contribution of i-cells during embryogenesis, however, has been unclear. We address neural cell origins during development of the Clytia hemisphaerica planula larva. In situ hybridisation of nematogenesis genes revealed that Piwi/Nanos1-expressing cells generate a substantial pool of nematoblasts (undifferentiated nematocytes) within the gastrula presum

Quantitative Neuropeptidomics Reveals Thermal Acclimation-Induced Remodeling of Peptidergic Signaling in the American Lobster Homarus americanus.

Global warming and rising ocean temperatures pose substantial challenges to marine ecosystems and crustacean populations. As an ectothermic species, the American lobster ( Homarus americanus ) relies on physiological and neurochemical mechanisms to maintain homeostasis under varying environmental conditions. To elucidate the role of neuropeptides in neuronal plasticity and systemic adaptation to temperature fluctuations, we employed a quantitative mass spectrometry-based approach to probe key neuropeptides involving thermal adaptation in four lobster neural tissues at three temperatures: 4 °C

The Role of the Nervous System in Lung Disease.

PURPOSE OF REVIEW: Peripheral nerves regulate pulmonary physiology in a nerve-type- and compartment-specific manner through neurotransmitters, neurotrophins and axon-guidance cues. This review examines how sensory and autonomic pathways influence pulmonary homeostasis and disease, and evaluates the experimental models available to investigate these interactions. RECENT FINDINGS: Dysregulated pulmonary innervation has been implicated in acute lung injury, airway hyperresponsiveness, pulmonary fibrosis and lung cancer, where neural signals may modulate immune activation, stromal remodelling and

An energy-boosting hydrogel couples magnetoelectrical stimulation and metabolic support for enhanced spinal cord regeneration.

Spinal cord injury (SCI) disrupts both neural and vascular networks, leading to a dual deficiency of bioelectric and biochemical energy that severely impedes neuroregeneration. To address this challenge, we developed an energy-boosting hydrogel that integrates biosafe electrical stimulation with a sustained-release biochemical energy system, thereby alleviating energy deficits to promote neural repair. Biodegradable potassium sodium niobate/ferrosoferric oxide (KNN/FO) nanoparticles were synthesized to enable in situ magnetoelectric conversion, and their efficacy in promoting regeneration thro

Sensory-secretory neuroimmune circuits of the ocular surface.

The ocular surface is a sensory-rich mucosal barrier in which trigeminal input, autonomic output, tear composition, epithelial stress, glandular secretion, goblet-cell function and local immunity are closely coupled. This review uses the sensory-secretory mucosal immune unit as a tissue-level map across four measurable output domains: secretory function, neural state, immune activity and barrier integrity. The term is not proposed as a new anatomical compartment or as a renamed lacrimal functional unit or eye-associated lymphoid tissue. Direct ocular evidence is distinguished from ocular-adjac

Hungry for Knowledge: Octopamine Signaling Regulates Hunger-Enhanced Olfactory Learning.

Hunger, a state of nutrient deficiency, significantly enhances cognitive behaviors by heightening sensitivity to food-related stimuli. However, the mechanisms by which hunger influences olfactory learning remain unclear. In this study, it is shown that aversive or appetitive memory is formed faster in hungry C. elegans. These findings reveal that hunger-induced octopamine release from the interneuron RIC enhances both aversive and appetitive olfactory learning. By analyzing neural circuits downstream of RIC, two distinct pathways involved in memory formation are identified. For aversive learni

HIV-associated depression: a translational framework targeting neuroimmune inflammation and psychosocial stress modulation.

People living with HIV (PLWH) are at increased risk for depression, anxiety, and other comorbid psychiatric disorders. HIV-associated depression involves complex neurobiological disturbances, including chronic neuroinflammation. This includes microglial activation, elevated levels of pro-inflammatory cytokines and mediators, and altered brain metabolites. Additionally, there is dysregulation of monoaminergic neurotransmission, particularly impaired serotonergic signaling. Prolonged hyperactivation of the hypothalamic-pituitary-adrenal axis, indicated by abnormally high cortisol levels, is also

Kappa and Mu Opioid Receptors in Chronic Cough: Current Evidence and Future Treatment.

Chronic cough is a significant burden on patient quality of life and is associated with poor health outcomes. Chronic cough may be a result of neural hypersensitivity due to changes in both the peripheral and the central nervous systems, although the exact mechanisms underlying its pathogenesis are not completely understood. Opioid receptors, specifically kappa and mu, are potential therapeutic targets in the management of chronic cough because they play a pivotal role in both the peripheral and the central neural pathways implicated in the act of coughing. Morphine, a mu opioid receptor agoni…

Otx2 mRNA expression is downregulated following traumatic brain injury in zebra finches.

Traumatic brain injury (TBI) induces a wide range of neurodegenerative symptoms, yet effective treatment strategies remain limited. Emerging evidence suggests that post-TBI recovery recapitulates aspects of early brain development, highlighting the potential for developmental molecular mechanisms to inform therapeutic interventions. The transcription factor Otx2 is critical for early brain and sensory organ development, as well as the maintenance of retinal and neural function in adulthood. Notably, the transfer of Otx2 homeoprotein into parvalbumin-expressing (PV+) GABAergic interneurons is e

The gut-brain axis in the context of colorectal cancer.

The gut-brain axis (GBA) plays a pivotal role in the pathophysiology of colorectal cancer (CRC), influencing tumor progression, immune responses, and therapeutic outcomes. Bidirectional communication between the gut and the brain, mediated through microbial metabolites, neurotransmitters, and neural signaling pathways, shapes the tumor microenvironment and systemic inflammation. CRC cells not only modify the gut microbiota but also release neuroactive molecules that influence neural circuits, further impacting tumor growth. The autonomic nervous system regulates gut motility, immune responses,
