Experimental Naegleria fowleri meningoencephalitis in sheep: light and electron microscopic studies.

A sheep infected intranasally with Naegleria fowleri of human origin died 7 days after inoculation. The olfactory lobes were distinctly soft … n vesicles, electron-dense granules, mitochondria, numerous free ribosomes, scant rough endoplasmic reticulum, and occasionally a phagocytized erythrocyte. Trophozoites were grouped close to arterioles, except when phagocytized by a neutrophil or endothelial cell.

Differentiation of melanomas occurring in platyfish-swordtail hybrids of different ages: an ultrastructural study.

The degree of differentiation of two types of hereditary melanomas of interspecific hybrids between the spotted variety of the platyfish (Xiphophorus maculatus) and the swordtail (Xiphophorus helleri) was examined electron microscopically. Melanomas of the fry consisted mainly of pigmented cells containing intermediately matured melanosomes and few other cytoplasmic structures. Melanomas of the adult consisted of lightly pigmented cells having a small number of immature melanosomes; numerous polysomes; well-developed, rough-surfaced endoplasmic reticulum (RER) and Golgi systems; and irregularl…

Ultrastructural observations on epithelioid cell granulomas induced by zirconium in the guinea-pig.

An ultrastructural study has been made of the allergic-type nodular granulomas that can be induced in guinea-pigs sensitised to sodium zirconium lactate. Lesions taken at the peak of granuloma development 7 days after skin testing contained epithelioid cells which could be recognised particularly by their content of rough endoplasmic reticulum, fimbriated cell membrane and typical nuclear appearance. Multinucleate giant cells were also seen. These cells were characterised by their large number of lysosome-like dense bodies and phagocytic structures. Both epithelioid cells and multinucleate gia…

Fine structural comparison of Ewing's sarcoma with neuroblastoma.

Two cases of Ewing's sarcoma and two neuroblastoma, rosette forming and round cell type, were studied electron microscopically and their fine structures were compared. The neoplastic cells of Ewing's sarcoma were characterized by aggregated glycogen particles in the cytoplasm. They had pseudopod-like cytoplasmic processes having tight junctions, which never contained microtubules or mitochondria. Ewing's sarcoma cells exhibited several stages of cell maturation and some mature cells possessed a large amount of smooth and rough endoplasmic reticulum, large Golgi complexes and numerous phagosome…

Ultrastructural evidence for the existence of two types of neurosecretory cells in the abdominal ganglia of the chelicerate arthropod, Limulus polyphemus.

Evidence suggesting the existence of two types of neurosecretory cells in each abdominal ganglion of Limulus polyphemus has been obtained by … onspicuous cytoplasmic vesiculation and possess more prominent parallel arrays of rough endoplasmic reticulum. Granules similar to those found in both cell types are present in the neuropile and certain nerves, but the specific pathways of the axons of these cells have not yet been determined.

MOSPD3 mediates the ER recruitment of ATG2A to promote autophagy.

ATG2A transfers glycerophospholipids from the endoplasmic reticulum (ER) to the expanding phagophore during autophagy, but how it is anchored to the ER in mammalian cells has been unclear. Here, we identify MOSPD3, an atypical member of the VAP family, as the ER adaptor for ATG2A. Endogenous MOSPD3 occupies ER subdomains adjacent to nascent autophagic structures, and it captures ATG2A through a direct interaction between its major sperm protein (MSP) domain and an FFNT (two phenylalanines in a neutral tract) motif near the ATG2A N terminus. Disrupting either side of this interface abolishes ER…

A cargo receptor entrapment complex is a therapeutic node for genetically and clinically distinct proteinopathies.

Severe proteinopathies-such as retinitis pigmentosa, a form of inherited blindness-are driven by genetic mutations that overwhelm the quality control of the post-endoplasmic reticulum (post-ER) secretory pathway, causing toxic protein accumulation. Here, we identify a therapeutic node defined by a hetero-oligomeric cargo receptor complex consisting of TMED7, 2, 9, and 10. This "entrapment complex" anchors structurally and functionally diverse mutant clients within the early secretory pathway via TMED7 binding to the integral Golgi protein GRASP55. Disruption of the entrapment complex results i…

Hung-up reflex: the history of a clinical sign and differential diagnosis.

The hung-up reflex (HUR) is a tonic reflex response characterized by delayed muscular relaxation following the elicitation of a tendon reflex. First described by Woltman in patients with myxedema and hypothyroidism, it is best demonstrated in the Achilles tendon. Currently, two primary techniques are used for its measurement: photomotography and the anemometer. The pathogenic mechanism underlying this phenomenon is closely linked to the physiological process of cellular membrane recovery, particularly the reabsorption of calcium by the sarcoplasmic reticulum. This sign has been documented in v…

Glucose-regulated protein 94 (Grp94/gp96) in viral pathogenesis: Insights into its role and therapeutic potentials.

Glucose-regulated protein 94 (Grp94/gp96) is endoplasmic reticulum (ER) resident form of the 90 kDa heat shock protein 90 (Hsp90) that is responsible for folding, maturation and stabilization of more than 400 client proteins. Grp94 has been implicated for various diseases including metastatic cancer, primary open-angle glaucoma, and infectious diseases. In fact, Grp94 plays critical roles in different stages of viral infection cycle. It chaperones receptor proteins and viral glycoproteins that are necessary for viral entry and replication. Beyond its role in protein homeostasis, Grp94 modulate…

TRx0237 induces apoptosis and enhances anti-PD-1 immunotherapeutic efficacy in anaplastic thyroid Cancer.

Anaplastic thyroid cancer (ATC) is a highly malignant and lethal tumor with poor prognosis, but there is a lack of effective treatment strategies. In our study, we screened a drug library and identified that TRx0237, a tau protein inhibitor, showed inhibitory effect on ATC cells. Further research demonstrated that the inhibitory effect of TRx0237 was mainly through the induction of apoptosis via reactive oxygen species (ROS)-mediated endoplasmic reticulum stress pathway. Meanwhile, the pro-apoptosis effect and mechanism of TRx0237 on ATC were verified in xenograft and ATC patient-derived organ…

Simpson-Golabi-Behmel Syndrome Associated With a Missense Variant at the Signal Peptide Cleavage Site of GPC3.

Translocation of newly synthesized proteins into the lumen of the endoplasmic reticulum (ER) is mediated by signal peptide recognition and cleavage. Here we report an individual with Simpson-Golabi-Behmel syndrome (SGBS) bearing a GPC3 missense variant at the signal peptide cleavage site of the glypican-3 protein. The c.71C>T; p.(Ala24Val) alteration in the key -1 position of the cleavage greatly reduces cleavage of the signal peptide, resulting in failure of the protein to efficiently exit the ER, and impaired glycosylation. Functional characterization of two other engineered variants at this…

[Research progress on the correlation between intestinal microecology and sepsis].

Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection, with increasing mortality as septic shock and organ failure progress. Mechanisms such as vascular endothelial dysfunction, microcirculatory disorders, coagulation abnormalities, immune suppression, mitochondrial damage, cell pyroptosis, ferroptosis, endoplasmic reticulum stress, and autophagy play crucial roles in organ dysfunction and death caused by sepsis. Concurrently, the imbalance of the gut microbiota also plays an undeniable role in the development of sepsis, with recent studies demonstr…

Molecular basis of the TRAP complex function in ER protein biogenesis.

The translocon-associated protein (TRAP) complex resides in the endoplasmic reticulum (ER) membrane and interacts with the Sec translocon and the ribosome to facilitate biogenesis of secretory and membrane proteins. TRAP plays a key role in the secretion of many hormones, including insulin. Here we reveal the molecular architecture of the mammalian TRAP complex and how it engages the translating ribosome associated with Sec61 translocon on the ER membrane. The TRAP complex is anchored to the ribosome via a long tether and its position is further stabilized by a finger-like loop. This positions…
