Can a Single Night of Sleep Deprivation Unmask Human Sleep and Neurophysiobehavioral Impairments During the Post-Acute Phase of Mild Traumatic Brain Injury?

The purpose of this study was twofold: (a) to determine whether, and the extent to which, the challenge of a single night of total sleep deprivation (TSD) unmasks lingering brain health-related deficits in individuals who within the past 3 to 12 months had been diagnosed (yet medically cleared) with a mild traumatic brain injury (mTBI+), and (b) to determine whether mTBI+ results in any neurophysiobehavioral deficits in the ability to recover from TSD. Seven previously concussed (mTBI+) adults (24.5 ± 5.3 years old) and six non-concussed control (mTBI-) adults underwent 24 h TSD preceded by 8 …

Associations between objectively measured sleep characteristics and six-minute walk distance in older adults: the Disparities in Sleep and Cognitive Outcomes (DISCO) study.

STUDY OBJECTIVES: A decline in physical functioning in older adults has been associated with a higher risk of chronic diseases. Subjective sleep disturbances have been associated with declines in physical functioning with aging. We examined the association between objectively measured sleep characteristics and 6-minute walk distance, a marker of functional capacity, in a sample of older adults. METHODS: This analysis included 199 participants who were enrolled in the Disparities in Sleep and Cognitive Outcomes (DISCO) study, a longitudinal cohort of older adults aged ≥55 years. Exposures incl…

Predicting Sleep and Sleep Stage in Children Using Actigraphy and Heartrate via a Long Short-Term Memory Deep Learning Algorithm: A Performance Evaluation.

Children's ambulatory sleep is commonly measured via actigraphy. However, traditional actigraphy measured sleep (e.g., Sadeh algorithm) struggles to predict wake (i.e., specificity, values typically < 70) and cannot predict sleep stages. Long short-term memory (LSTM) is a machine learning algorithm that may address these deficiencies. This study evaluated the agreement of LSTM sleep estimates from actigraphy and heartrate (HR) data with polysomnography (PSG). Children (N = 238, 5-12 years, 52.8% male, 50% Black 31.9% White) participated in an overnight laboratory polysomnography. Participants …

Role of Adenosine A1 Receptor in Sleep Deprivation-Induced Neuroinflammation: Insights on Rapid Eye Movement Sleep and Fear Extinction Memory Recall in Rats.

INTRODUCTION: Sleep deprivation (SD), stemming from a myriad of aetiologies, is a prevalent health condition frequently overlooked. It typically impairs memory consolidation and synaptic plasticity, potentially through neuroinflammatory mechanisms and adenosinergic signalling. It is still unclear whether the adenosine A1 receptor (A1R) modulates SD-induced neurological deficits in the hippocampus. OBJECTIVES: This study aims to evaluate the effects of SD on fear extinction memory recall and emotional behaviour in male Sprague Dawley rats; to investigate the role of A1R antagonism by the admin…

Standardized Extract of Valeriana officinalis Improves Overall Sleep Quality in Human Subjects with Sleep Complaints: A Randomized, Double-Blind, Placebo-Controlled, Clinical Study.

INTRODUCTION: Sleep deficit or poor sleep leads to ill-health, whereas sleep deprivation for longer periods of time increases the risk of developing adverse conditions associated with poor quality of life, and high socioeconomic impact. The treatments for sleep disturbances include melatonin and over-the-counter medicines like diphenhydramine and doxylamine, all of which have negative side effects. Valerian (Valeriana officinalis L.) is a traditional herb and the most preferred alternate sleep solution to manage sleep complaints. METHODS: Eighty adult subjects with sleep complaints were rando…

Abnormally abrupt transitions from sleep-to-wake in Huntington's disease sheep (Ovis aries) are revealed by automated analysis of sleep/wake transition dynamics.

Sleep disturbance is a common and disruptive symptom of neurodegenerative diseases such as Alzheimer's and Huntington's disease (HD). In HD patients, sleep fragmentation appears at an early stage of disease, although features of the earliest sleep abnormalities in presymptomatic HD are not fully established. Here we used novel automated analysis of quantitative electroencephalography to study transitions between wake and non-rapid eye movement sleep in a sheep model of presymptomatic HD. We found that while the number of transitions between sleep and wake were similar in normal and HD sheep, t…

Effect of mouth closure on upper airway obstruction in patients with obstructive sleep apnoea exhibiting mouth breathing: a drug-induced sleep endoscopy study.

PURPOSE: Snoring and obstructive sleep apnoea (OSA) exhibit multifactorial aetiologies; mouth breathing increases airway obstruction and upper respiratory tract resistance. Of the many published studies, few have evaluated sleeping subjects. We explored how mouth breathing affected the upper respiratory tract anatomy and OSA during sleep. METHODS: Eighteen patients with OSA, confirmed via full-night polysomnography, were enrolled in this study. We performed drug-induced sleep endoscopy (DISE) and defined obstruction sites before and after mouth closure using commercial mouth strips. We evalua…

Does sleep predict next-day napping or does napping influence same-day nocturnal sleep? Results of a population-based ecological momentary assessment study.

STUDY OBJECTIVES: The temporal relationship between nocturnal sleep and daytime napping has only been assessed in small non-representative samples, and suggests that nocturnal sleep and napping are interdependent, although mixed results exist. In this study, we investigated the temporal relationship between nocturnal sleep and napping (and vice versa). METHODS: A population-based sample of middle-aged adults (N = 683, mean age 60.7 [SD 9.5]) completed seven days of ecological momentary assessment reporting sleep and nap characteristics. Multilevel random-effects models were used to assess the…
