Vezatin, a ubiquitous protein of adherens cell-cell junctions, is exclusively expressed in germ cells in mouse testis.

In the male reproductive organs of mammals, the formation of spermatozoa takes place during two successive phases: differentiation (in the testis) and maturation (in the epididymis). The first phase, spermiogenesis, relies on a unique adherens junction, the apical ectoplasmic specialization linking the epithelial Sertoli cells to immature differentiating spermatids. Vezatin is a transmembrane protein associated with adherens junctions and the actin cytoskeleton in most epithelial cells. We report here the expression profile of vezatin during spermatogenesis. Vezatin is exclusively expressed in

Effect of Cr(V) on reproductive organ morphology and sperm parameters: an experimental study in mice.

BACKGROUND: Cr(V) species are formed during the intracellular reduction of Cr(VI), a ubiquitous environmental pollutant. In this study, the acute toxicity of a physiologically stable Cr(V) compound, [CrV-BT](2-) (BT = bis(hydroxyethyl)aminotris(hydroxymethyl)methane) was investigated in the male reproductive system of sexually mature 60-day-old male ICR-CD1 mice. METHODS: Eight-week-old animals were subcutaneously injected daily with a dose of ca 8 micromol of Cr/mouse, during 5 days. The control group was injected with 0.5 mL of BT buffer. Testis and epididymis morphology was evaluated using

Seminal Plasma Exosomal miRNA Profiling Reveals hsa-miR-7-5p as a Key Regulator of Sperm Motility in Asthenozoospermia.

BACKGROUND: Asthenozoospermia is a significant contributing factor to male infertility. Accumulating evidence indicates that impaired sperm motility is closely linked to dysregulated microRNA expression during spermatogenesis. Seminal plasma exosomes are enriched with diverse microRNAs, which play pivotal roles in modulating sperm motility, the acrosome reaction, capacitation, and fertilization. OBJECTIVE: To investigate the association between exosomal microRNA profiles and sperm motility by comparing microRNA expression patterns in seminal plasma exosomes from asthenozoospermic patients and…

Integration analysis of transcriptome and metabolome revealed the potential mechanism of spermatogenesis in Tibetan sheep (Ovis aries) at extreme high altitude.

Testis has an indispensable function in male reproduction of domestic animals. Numerous genes and metabolites were related to testicular dev … velopment and spermatogenesis, and might affect the reproduction of Tibetan sheep by regulating the acrosome reaction, meiotic gene expression, and the production of sex hormones. Our results provide further understanding of the key genes and metabolites involved in testicular development and spermatogenesis in Tibetan sheep.

Different approaches for assessing sperm function.

Different approaches can be used to assess sperm function in different conditions, i.e. sperm storage, freezing-thawing or activation by induction of capacitation and acrosome reaction. In this review we will focus on the assays routinely performed in our laboratories, giving a literature support to critically analyse different approaches. In fact, researchers usually tend to look for the "one shot" parameter that could explain itself a specific process; it is our conviction that a multiparametric approach is still more valid, as some changes in sperm function are very complex and could be exp…

Identification, Characterization and Synthesis of Walterospermin, a Sperm Motility Activator from the Egyptian Black Snake Walterinnesia aegyptia Venom.

Animal venoms are small natural mixtures highly enriched in bioactive components. They are known to target at least two important pharmacological classes of cell surface receptors: ion channels and G protein coupled receptors. Since sperm cells express a wide variety of ion channels and membrane receptors, required for the control of cell motility and acrosome reaction, two functions that are defective in infertility issues, animal venoms should contain interesting compounds capable of modulating these two essential physiological functions. Herein, we screened for bioactive compounds from the …

Early defect in the expression of mouse sperm DNAJ 1, a member of the DNAJ/heat shock protein 40 chaperone protein family, in the spinal cord of the wobbler mouse, a murine model of motoneuronal degeneration.

Prevention of protein misfolding is ensured by chaperone proteins, including the heat shock proteins (HSP) of the DNAJ/HSP40 family. Detection of abnormal protein aggregates in various neurodegenerative diseases has led to the proposal that altered chaperone activity contributes to neurodegeneration. Msj-1, a DNAJ/HSP40 protein located around the spermatozoa acrosome, was recently found to be down-regulated in the testis of wobbler mutant mice. Wobbler is an unidentified recessive mutation which triggers progressive motoneuron degeneration with abnormal intracellular protein accumulations, and

The percentage of spermatozoa displaying the chlortetracycline Pattern AR (acrosome reaction) was also statistically related to fertility (r = 0.05, P = 0.04), but the biological importance of this relationship is questionable given the small variation among ejaculates (range: 0 to 2%). No other sperm parameter was significantly related to …

Chronic stress disturbs neuroendocrine control of reproduction and fertility in male rats.

Currently, stress is considered one of the risk factors for infertility in male humans, altering sperm function. Sperm production and maturation depends on the hypothalamic-pituitary-testis axis control. Therefore, the objective of the current study was to evaluate the effects of chronic stress on the neuroendocrine control of male reproduction, the oxidative status in the epididymis, and male fertility. Adult male rats were assigned to control or chronic stress groups. Chronically stressed males were exposed to cold-water immersion (CWI) for 50 consecutive days. After euthanasia, the hypothal

Motility enhancement of human spermatozoa using electrical stimulation in the nano-Ampere range with enzymatic biofuel cells.

Sperm motility is a crucial factor for normal fertilisation that is partly supported by mitochondrial activity. Enzymatic biofuel cells (EBFCs) generate electric currents by an electron grade from anodic to cathodic electrodes in a culture media. We demonstrate that electrical stimulation by EBFC at the nano-Ampere range enhances sperm motility that can potentially allow the development of a new therapeutic tool for male infertility, including poor motility. EBFC was set up with three different electrical currents (112 nA/cm2 and 250 nA/cm2) at two different times (1 h, 2 h). Each sample was e

Lipid Biochemical and Biophysical Changes in Rat Spermatozoa During Isolation and Functional Activation In Vitro.

In spermatozoa isolated from rat epididymis, lipids are differentially modified after in vitro induction of capacitation (Cap) and the acrosomal reaction (AR). This study uses Laurdan fluorescence generalized polarization values (GPv) to evaluate the effect of lipid changes occurring after isolation and functional activation on sperm membrane biophysical properties. In gametes isolated in the presence of a divalent cation chelator, no lipid changes occurred and the GPv were the lowest recorded, indicating maximal membrane lipid mobility. In sperm isolated as rapidly and gently as possible in t

Evaluation of Lasting Effects of Heat Stress on Sperm Profile and Oxidative Status of Ram Semen and Epididymal Sperm.

Higher temperatures lead to an increase of testicular metabolism that results in spermatic damage. Oxidative stress is the main factor responsible for testicular damage caused by heat stress. The aim of this study was to evaluate lasting effects of heat stress on ejaculated sperm and immediate or long-term effects of heat stress on epididymal sperm. We observed decrease in motility and mass motility of ejaculated sperm, as well as an increase in the percentages of sperm showing major and minor defects, damaged plasma and acrosome membranes, and a decrease in the percentage of sperm with high m

DDI-4 induces a temperature-sensitive exocytotic remodeling during C. elegans spermiogenesis via nsun-2-dependent sphingosine signaling.

Exocytotic sperm remodeling is essential for fertilization, yet whether conserved molecular mechanisms underlie this process across species remains unclear. In Caenorhabditis elegans, membranous organelle fusion (MOF), an exocytotic event cytologically analogous to the mammalian acrosome reaction (ASR), occurs during spermiogenesis and likely contributes to the acquisition of fertilization competence. Here, we identify DDI-4, a benzylamine analog as a small-molecule inducer of both MOF and ASR, revealing a shared sensitivity between these evolutionarily distant systems. DDI-4-induced MOF was s…

Novel mutations in FSIP2 cause male infertility through multiple morphological abnormalities of the sperm flagella.

Infertility is a global concern, and oligoasthenoteratozoospermia (OAT) is the most severe form of male infertility, characterized by reduced sperm count, decreased motility, and increased abnormal morphology. Multiple morphological abnormalities of the sperm flagella (MMAF) characterize the most severe type of OAT and are usually caused by loss-of-function mutations in the genes essential for vital aspects of sperm biology, including concentration, motility, and morphology. The fibrous sheath interacting protein 2 (FSIP2) plays an essential role in sperm flagellar structure and function by re
