Insights on the sex determination, vector capacity and ecological biology from a chromosomal level genome of vector mosquito, Armigeres subulbatus.

BACKGROUND: Mosquitoes with aggressive biting behavior are important disease vectors threatening public health. Armigeres subalbatus, as an emerging arbovirus and filarial disease vector, exhibits aggressive host-seeking behavior and unique breeding preference for contaminated water. However, the molecular mechanisms underlying these biological characteristics remain poorly understood. This study aimed to generate a high-quality genome assembly and characterize the genetic basis of vector competence and environmental adaptation in Ar. subalbatus. METHODS: We sequenced and assembled the Ar. su

The spinal cord facilitates cerebellar upper limb motor learning and control; inputs from neuromusculoskeletal simulation.

Complex interactions between brain regions and the spinal cord (SC) govern body motion, which is ultimately driven by muscle activation. Motor planning or learning are mainly conducted at higher brain regions, whilst the SC acts as a brain-muscle gateway and as a motor control centre providing fast reflexes and muscle activity regulation. Thus, higher brain areas need to cope with the SC as an inherent and evolutionary older part of the body dynamics. Here, we address the question of how SC dynamics affects motor learning within the cerebellum; in particular, does the SC facilitate cerebellar

Toward adaptive cyborg tissues.

Organoids and engineered tissues have emerged as powerful models of human biology, yet their development and function are often characterized through endpoint measurements that provide only static snapshots of dynamic biological processes. Recent advances in stretchable mesh electronics, developmental biointegration, and organoid engineering have enabled the creation of cyborg tissues with embedded sensing, stimulation, and computational capabilities. This review highlights recent progress in developmental bioelectronic integration, multifunctional tissue mapping, biological state reconstructi

Three-dimensional micro-scale strain mapping in living biological soft tissues.

UNLABELLED: Non-invasive characterization of the mechanical micro-environment surrounding cells in biological tissues at multiple length scales is important for the understanding of the role of mechanics in regulating the biosynthesis and phenotype of cells. However, there is a lack of imaging methods that allow for characterization of the cell micro-environment in three-dimensional (3D) space. The aims of this study were (i) to develop a multi-photon laser microscopy protocol capable of imprinting 3D grid lines onto living tissue at a high spatial resolution, and (ii) to develop image process

Intestinal adaptation following massive small bowel resection in the mouse.

BACKGROUND: Transgenic mice represent powerful tools for studying the role of genes and their expression under multiple conditions, and they may provide a unique model for studies of intestinal adaptation after massive small bowel resection (SBR). This study characterized a successful model for SBR and intestinal adaptation in the mouse. STUDY DESIGN: Sham operation (bowel transection with reanastomosis) or SBR was performed on male C57BL/6 mice. A solid or liquid diet, various sizes of monofilament suture for the anastomosis, and resection of 50 or 75 percent of the proximal small intestine

Deep learning predicts potential reassortments of avian H5N1 with human influenza viruses.

Frequent infection cases with avian H5N1 influenza A viruses (IAVs) are posing pandemic risks of human/avian-reassorted IAVs. We aimed to build an attentional deep learning model named HAIRANGE, for predicting potentially human-adapted reassortment of H5N1 and human IAVs. A biologically relevant and non-pretrained embedder named Codon2Vec in HAIRANGE performed competitively in benchmarking against other embedders, such as ESM2, DNABERT2 and others, indicating a high association of genomic context with viral hosts or serotypes, for IAV RNA polymerase-related genes. HAIRANGE accurately predicted

Circulating Epstein-Barr virus DNA as a precision biomarker in nasopharyngeal carcinoma: from risk stratification to real-time treatment adaptation and disease surveillance.

PURPOSE OF REVIEW: Nasopharyngeal carcinoma (NPC) is a virally driven malignancy with marked biological heterogeneity that is inadequately captured by anatomical staging alone. Circulating plasma Epstein-Barr virus (EBV) DNA has emerged as an archetypal biomarker with expanding roles across the disease continuum. This review summarizes the recent evidence that had expanded plasma EBV DNA utility beyond prognostication to real-time treatment adaptation and disease surveillance. RECENT FINDINGS: Contemporary studies had demonstrated that baseline EBV DNA refines risk stratification beyond TNM s

A reconfigurable and conformal liquid sensor for ambulatory cardiac monitoring.

The severe mismatch between solid bioelectronics and dynamic biological tissues has posed enduring challenges in the biomonitoring community. Here, we developed a reconfigurable liquid cardiac sensor capable of adapting to dynamic biological tissues, facilitating ambulatory cardiac monitoring unhindered by motion artifacts or interference from other biological activities. We employed an ultrahigh-resolution 3D scanning technique to capture tomographic images of the skin on the wrist. Then, we established a theoretical model to gain a deep understanding of the intricate interaction between our

Short-Term Combined Tat-Beclin1 and Endurance Training Improves Age-Related Decline in Physical Function in Male Mice.

UNLABELLED: Autophagy is a hallmark of aging, but autophagy-related proteins have not been exclusively targeted to attenuate the progressive decline in physical function associated with aging. Here, we combined Tat-Beclin1, an autophagy agonist, and endurance training to determine whether Tat-Beclin1 enhances exercise adaptation in old male mice. Tat-Beclin1 was administered intraperitoneally (TB group, 15 mg/kg, 2x/week) as a standalone therapy, or in combination with endurance training (TB+Exe group, 70% of maximal running speed 3x/week) for 1 month in 23-month-old male C57BL/6J mice. Early Hepatic Transcriptomic Responses to High-Fat Diet in Apolipoprotein A-IV Knockout Mice.

BACKGROUND: Apolipoprotein A-IV (ApoA-IV) is involved in lipid metabolism and energy homeostasis, but its role in hepatic adaptation to dietary stress remains incompletely understood. This study aimed to determine whether ApoA-IV deficiency alters hepatic transcriptional responses to high-fat diet (HFD) and to examine how these molecular changes relate to biochemical and histological features of liver function. METHODS: Male ApoA-IV knockout (KO) and wild-type (WT) mice were fed either a normal diet (ND) or HFD for 12 weeks. Liver tissues were subjected to whole-transcriptome RNA sequencing,

Response and adaptation of the transcriptional heat shock response to pressure.

INTRODUCTION: The molecular mechanisms underlying pressure adaptation remain largely unexplored, despite their significance for understanding biological adaptation and improving sterilization methods in the food and beverage industry. The heat shock response leads to a global stabilization of the proteome. Prior research suggested that the heat shock regulon may exhibit a transcriptional response to high-pressure stress. METHODS: In this study, we investigated the pressure-dependent heat shock response in E. coli strains using plasmid-borne green fluorescent protein (GFP) promoter fusions and

MALDI mass spectrometry imaging and in situ microproteomics of Listeria monocytogenes biofilms.

UNLABELLED: MALDI-TOF Mass spectrometry Imaging (MSI) is a surface-sampling technology that can determine spatial information and relative abundance of analytes directly from biological samples. Human listeriosis cases are due to the ingestion of contaminated foods with the pathogenic bacteria Listeria monocytogenes. The reduction of water availability in food workshops by decreasing the air relative humidity (RH) is one strategy to improve the control of bacterial contamination. This study aims to develop and implement an MSI approach on L. monocytogenes biofilms and proof of concept using a

Beyond Resistance Genes: Pseudomonas aeruginosa as a Complex Adaptive System Driving Persistence, Evolution, and Antimicrobial Resistance.

Pseudomonas aeruginosa (P. aeruginosa) is one of the most adaptable bacterial pathogens and a major cause of difficult-to-treat infections worldwide. Although antimicrobial resistance (AMR) is commonly attributed to resistance genes and their associated mechanisms, this perspective does not fully explain the ability of P. aeruginosa to survive antimicrobial exposure, establish chronic infections, and persist across diverse environmental and host-associated habitats. In this review, we examine P. aeruginosa within a complex adaptive systems framework, integrating evidence from molecular microbi

A trade-off in evolution: the adaptive landscape of spiders without venom glands.

BACKGROUND: Venom glands play a key role in the predation and defense strategies of almost all spider groups. However, the spider family Uloboridae lacks venom glands and has evolved an adaptive strategy: they excessively wrap their prey directly with spider silk instead of paralyzing it first with toxins. This shift in survival strategy is very fascinating, but the genetic underpinnings behind it are poorly understood. RESULTS: Spanning multiple spider groups, we conducted multiomics analyses on Octonoba sinensis and described the adaptive evolution of the Uloboridae family at the genome lev

Nutrient availability regulates proline/alanine transporters in Trypanosoma brucei.

Trypanosoma brucei is a species of unicellular parasite that can cause severe diseases in livestock and humans, including African trypanosomiasis and Chagas disease. Adaptation to diverse environments and changes in nutritional conditions is essential for T. brucei to establish an infection when changing hosts or during invasion of different host tissues. One such adaptation is the ability of T. brucei to rapidly switch its energy metabolism from glucose metabolism in the mammalian blood to proline catabolism in the insect stages and vice versa. However, the mechanisms that support the parasit

Testing the drivers of the temperature-size covariance using artificial selection.

Body size often declines with increasing temperature. Although there is ample evidence for this effect to be adaptive, it remains unclear whether size shrinking at warmer temperatures is driven by specific properties of being smaller (e.g., surface to volume ratio) or by traits that are correlated with size (e.g., metabolism, growth). We used 290 generations (22 months) of artificial selection on a unicellular phytoplankton species to evolve a 13-fold difference in volume between small-selected and large-selected cells and tested their performance at 22°C (usual temperature), 18°C (-4), and 26

Parasite Sensing of Host Nutrients and Environmental Cues.

Parasites undergo complex life cycles that comprise a wide variety of cellular differentiation events in different host compartments and transmission across multiple hosts. As parasites depend on host resources, it is not surprising they have developed efficient mechanisms to sense alterations and adapt to the available resources in a wide range of environments. Here we provide an overview of the nutritional needs of different parasites throughout their diverse life stages and highlight recent insights into strategies that both hosts and parasites have developed to meet these nutritional requi

Influenza Virus-Host Co-evolution. A Predator-Prey Relationship?

Influenza virus continues to cause yearly seasonal epidemics worldwide and periodically pandemics. Although influenza virus infection and its epidemiology have been extensively studied, a new pandemic is likely. One of the reasons influenza virus causes epidemics is its ability to constantly antigenically transform through genetic diversification. However, host immune defense mechanisms also have the potential to evolve during short or longer periods of evolutionary time. In this mini-review, we describe the evolutionary procedures related with influenza viruses and their hosts, under the pris

Homeoviscous Adaptation and the Regulation of Membrane Lipids.

Biological membranes are complex and dynamic assemblies of lipids and proteins. Poikilothermic organisms including bacteria, fungi, reptiles, and fish do not control their body temperature and must adapt their membrane lipid composition in order to maintain membrane fluidity in the cold. This adaptive response was termed homeoviscous adaptation and has been frequently studied with a specific focus on the acyl chain composition of membrane lipids. Massspectrometry-based lipidomics can nowadays provide more comprehensive insights into the complexity of lipid remodeling during adaptive responses.
