Differences in mRNA expression of adipocyte-derived factors in response to fasting, refeeding and leptin.

The present study examines whether and to what extent the profiles of adipose-derived factors are altered in epididymal and subcutaneous adipose tissues of long-term fasted/refed and of fasted rats treated by recombinant leptin. Fasting was characterized by three successive metabolic phases. Minor differences in the time-course and magnitude of response were detected between the two adipose sites. Leptin, adiponectin, resistin, adiponutrin, and insulin-like growth factor-1 (IGF-1) gene expressions differentially decreased according to the fasting duration. mRNA levels reached a minimum in late

Defective prelamin A processing and muscular and adipocyte alterations in Zmpste24 metalloproteinase-deficient mice.

The mouse ortholog of human FACE-1, Zmpste24, is a multispanning membrane protein widely distributed in mammalian tissues and structurally related to Afc1p/ste24p, a yeast metalloproteinase involved in the maturation of fungal pheromones. Disruption of the gene Zmpste24 caused severe growth retardation and premature death in homozygous-null mice. Histopathological analysis of the mutant mice revealed several abnormalities, including dilated cardiomyopathy, muscular dystrophy and lipodystrophy. These alterations are similar to those developed by mice deficient in A-type lamin, a major component

Regulation of activating transcription factor-2 in early stage of the adipocyte differentiation program.

p38beta mitogen-activated protein kinase activity is required for the differentiation of 3T3-L1 fibroblasts into adipocytes. Activating transcription factor-2 (ATF-2) is efficiently phosphorylated and activated by p38beta kinase. These findings led us to examine a regulatory role of ATF-2 in adipocyte differentiation. The induction of ATF-2 protein precedes the expression of the transcription factors, peroxisome proliferator-activated receptor (PPAR) gamma and CCAAT/enhancer-binding protein (C/EBP) alpha. Consistent with early activation of p38beta kinase, the phosphorylation of ATF-2 was also…

A cysteine-rich adipose tissue-specific secretory factor inhibits adipocyte differentiation.

A 12.5-kDa cysteine-rich adipose tissue-specific secretory factor (ADSF/resistin) is a novel secreted protein rich in serine and cysteine re … . The mRNA level was markedly increased during 3T3-L1 and primary preadipocyte differentiation into adipocytes. Its expression in adipose tissue is under tight nutritional and hormonal regulation; the mRNA level was very low during fasting and increased 25-fold when fasted mice were refed a high carbohydrate diet. It was also very low in adipose tissue of st …

Antiadipogenic properties of retinol in primary cultured differentiating human adipocyte precursor cells.

The aim of this study was to investigate the effect of retinol on the human adipose conversion process using primary cultured human adipocyt … ogical analysis. These results strongly suggest that retinol, by inhibiting the ability of human preadipocytes to convert into mature adipocytes, could be of potential interest in the prevention of human adipose tissue development in general and of cellulitis in particular.

The inhibitory guanine nucleotide-binding protein Gi2alpha induces and potentiates adipocyte differentiation.

The present study further elucidates the involvement of the alpha-subunit of the GTP-binding protein Gi2 in the differentiation of murine 3T3-L1 cells. Control and vector-transfected cells attained a fully differentiated adipocyte phenotype showing ample lipid droplets. Cells expressing wild type (WT)-Gi2alpha or the constitutively active R179E-Gi2alpha, however, became enlarged, less confluent, and produced large amounts of lipids. Differentiation consistently increased the triglyceride (TAG) content in control cells. In both WT-Gi2alpha and R179E-Gi2alpha clones, a marked increase in TAG cou

Regulation by noradrenaline of the mitochondrial and microsomal forms of glycerol phosphate acyltransferase in rat adipocytes.

Incubation of rat adipocytes with 1 microM-noradrenaline caused a decrease in both the N-ethylmaleimide-sensitive (microsomal) and N-ethylmaleimide-insensitive (mitochondrial) glycerol phosphate acyltransferase activities measured in homogenates from freeze-stopped cells. The effects of noradrenaline on glycerol phosphate acyltransferase activity were apparent over a wide range of concentrations of glycerol phosphate and palmitoyl-CoA. The effect of noradrenaline was reversed within cells by the subsequent addition of insulin or propranolol. Inclusion of albumin in homogenization buffers aboli…

Halogenated bisphenol A alternatives inhibit aldo-keto reductase 1C1 and induce metabolic alterations in human adipocytes.

Bisphenol A (BPA) is a widely used plastic monomer with well-established endocrine-disrupting properties and emerging evidence for metabolism-disrupting effects. Following regulatory restrictions, structurally modified alternatives, including halogenated derivatives, have been introduced, but information on their metabolic effects remains scarce. Hence, we analyzed proteomic and metabolomic responses in SGBS human adipocytes after exposure to BPA and five analogues (TBBPA, TCBPA, BPS, TBBPS, and TCBPS), including environmentally relevant concentrations of 10 nM. Intracellular target proteins w…

Cold-induced serum amyloid A3 amplifies cAMP-PKA signaling to drive adaptive thermogenesis in brown adipocytes.

Serum amyloid A3 (SAA3) is typically associated with inflammation, but its role in brown adipose tissue (BAT) thermogenesis remains elusive. Here, we identify SAA3 as a cold-inducible, locally acting amplifier of BAT thermogenesis. Using lentiviral modulation and BAT-specific knockout mice, we demonstrate that SAA3 is essential for cold tolerance and energy expenditure. Mechanistically, secreted SAA3 augments thermogenic signaling via a GPR3-dependent cAMP/PKA axis, driving robust UCP1 induction. Notably, this activation triggers transcriptional upregulation of endogenous Saa3 and Gpr3, establ

Functional annotation of insulin-responsive genes in human adipocytes reveals PLCXD1 as a lipid storage regulator.

Insulin-driven gene regulation is central to adipocyte function, but the roles of many of these genes in lipid metabolism remain unclear. Here, we integrate three transcriptomic datasets to identify insulin-responsive genes and define their functions in human adipocytes using a multiparametric lipid turnover screen. Our results reveal four major clusters involved in metabolic regulation, transcription, stress responses, and lipid metabolism. Among lipid-related hits, phospholipase C X domain-containing protein-1 (PLCXD1) emerges as a regulator of insulin-stimulated lipogenesis, without affecti…

Silencing of Homeodomain-Interacting Protein Kinase 2 (HIPK2) Induces Anti-Adipogenic Effects in 3T3-L1 Adipocytes.

Homeodomain-interacting protein kinase 2 (HIPK2) is an important regulator of various transcription factors and cofactors, which are involved in cell growth, cell death, and embryonic development. Previously, it has been reported that HIPK2 is upregulated during white fat development. However, the expression and functional role of HIPK2 during adipogenesis remain unclear. Here, we investigated the expression and biological function of HIPK2 during the adipogenesis of 3T3-L1 cells. Importantly, protein and mRNA expression of HIPK2 were significantly upregulated in a time-dependent manner during

TG-interacting factor 1 mediates TNF-induced Ccl2 expression in adipocytes to promote prostate cancer progression.

Periprostatic adipose tissue secretes chemokines and lipid mediators that, particularly in the context of obesity, enhance prostate cancer cell (PCa) migration and invasion. Treating 3 T3-F442A pre-adipocytes with conditioned medium (CM) from M1-like RAW264.7 cells resulted in a significant upregulation of TG-interacting factor 1 (Tgif1) expression. Using a Tgif1 promoter reporter assay, we showed that the tumor necrosis factor (TNF) transcriptionally upregulates Tgif1 promoter activity, consistent with increased mRNA and protein expression. We also demonstrated that Tgif1 contributes to the m…

Current and Future Perspectives on Adipo-Oncology: Roles of Adipocytes in Local and Systemic Tumor Regulation.

Adipose tissue has traditionally been regarded as an energy-storage organ; however, accumulating evidence indicates that adipocytes actively regulate cancer biology through local and systemic mechanisms. These findings have contributed to the emergence of adipo-oncology, an interdisciplinary field focused on interactions between adipose tissue and cancer. This review summarizes current evidence on the local and systemic roles of adipocytes in tumor progression and discusses how tissue heterogeneity and plasticity may influence these processes. Locally, adipocytes adjacent to tumors may be repr…

MTT-Based Normalization of Glycerol Release for Accurate Quantification of Lipolysis in Primary Rat Epididymal Adipocytes.

Lipolysis, a critical metabolic pathway, involves the hydrolysis of stored triglycerides into free fatty acids and glycerol. However, accurate quantification of glycerol output in lipid-rich primary adipocytes is often confounded by experimental artifacts, such as variability in cell viability and assay interference. To address these limitations, a straightforward method was established to normalize glycerol release measurements using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)-based cell viability assay. This protocol leverages the difference in specific gravity bet…

Wnt Activation in Mature Dermal Adipocytes Leads to Lipodystrophy and Skin Fibrosis via ATGL-Dependent Lipolysis.

Accumulation of extracellular matrix (ECM) and dermal adipocyte lipodystrophy occurs during skin fibrosis, which compromises the skin's flexibility and function. Sustained Wnt activation in dermal progenitor cells leads to fibrotic ECM thickening in the dermis and lipodystrophy of dermal white adipose tissue (DWAT). Mouse genetic models with lineage tracing demonstrate that Wnt activation in mature dermal adipocytes is sufficient to induce adipocyte lipodystrophy and fibrotic ECM remodeling in the skin. Upon withdrawal of adipocyte-restricted Wnt activation, lipodystrophy and fibrosis are reve…

Recent advances in osteonecrosis of the femoral head: a focus on mesenchymal stem cells and adipocytes.

Osteonecrosis of the femoral head (ONFH) is a debilitating orthopedic disease characterized by femoral head collapse and destruction of bone and articular cartilage, resulting in severe joint pain and loss of hip mobility. Bone marrow mesenchymal stem cells (BMSCs) exhibit multilineage differentiation potential, including osteoblasts, adipocytes, fibroblasts, chondrocytes, and neurocytes. The imbalance between osteogenesis and adipogenesis in BMSCs plays a critical role in ONFH pathogenesis. Factors such as alcohol consumption and glucocorticoid exposure promote adipogenic differentiation whil…

Effect of vitamin D3 on lipid droplet growth in adipocytes of mice with HFD-induced obesity.

Vitamin D-regulating action of PPARγ on obesity has been confirmed on adipocyte differentiation. However, it is not clear whether vitamin D affects the morphological size of mature adipocytes by influencing the expression of PPARγ in vivo. Our hypothesis was that Vitamin D3 (VitD3) inhibits the growth of adipocyte size by suppressing PPARγ expression in white adipocytes of obese mice. Five-week-old male C57BL/6J mice were randomly divided into normal diet and high-fat diet groups. After 10 weeks, the body weight between the two groups differed by 26.91%. The obese mice were randomly divided in…
