Associations of Romantic Partners' Characteristics with Alcohol Consumption and Binge Drinking: Examining Evidence for Gene-Environment Interaction.

We investigated associations of romantic partners' alcohol use, cigarette smoking, personality, and psychological distress with alcohol use and binge drinking within a sample of Finnish twins who have initiated alcohol use (N = 1620, 51% female, mean age = 33.6 years) and their romantic partners. We also used twin modeling to examine whether partner characteristics moderate genetic influences on drinking behavior. Having a romantic partner with more frequent alcohol use and smoking was consistently associated with greater alcohol consumption and binge drinking, and partner alcohol use and smok

Imposter syndrome and college students' drinking behaviors: The roles of negative affect and coping motivated alcohol use.

BACKGROUND AND OBJECTIVES: College students are known to frequently engage in alcohol consumption, and are at risk for developing dangerous drinking patterns (e.g., binge drinking). Since imposter syndrome occurs at high rates among undergraduate students, it is reasonable to assume that college students may experience its negative effects, such as anxiety and depression. Given the relation of anxiety and depression to worse drinking outcomes, imposter syndrome may also be related to greater drinking and related problems. Yet, this is the first known study to test whether imposter syndrome is

Goal-directed behavior and hippocampal activity predict real-life impact of drinking intentions in alcohol use disorder.

A shift away from goal-directed, model-based behavior is commonly viewed to characterize alcohol use disorder (AUD). Previous research, however, has failed to demonstrate differences between individuals with and without AUD regarding goal-directed control, operationalized as model-based behavior. Instead, findings suggest associations between model-based behavior and alcohol consumption patterns, but mechanistic insights into the link between model-based behavioral and neural signatures and longitudinal, real-life control over alcohol intake remain elusive. Here, we investigated whether experi

N6-methyladenosine RNA methylation is a novel epitranscriptomic regulator of excessive alcohol drinking and vulnerability to relapse.

While internal RNA modifications have been known for decades, the contribution of epitranscriptomics to alcohol use disorder (AUD) remains unexplored. Here we investigated, for the first time, the role of the most abundant mRNA modification, N6-methyladenosine (m6A) in the regulation of alcohol drinking and alcohol mediated gene expression. The fat mass and obesity associated gene (FTO) plays a key role in erasing m6A RNA methylation marks. We generated brain m6A hypermethylated mice with selective deletion of FTO in neurons by crossing FTO-floxed mice (FTO-fl/fl) with the synapsin-1-CRE delet…

The Effect of Serotonin 5-HT2C Receptor Modulation on Binge Drinking and Alcohol-Seeking in Female Mice.

Alcohol misuse remains a leading cause of preventable death worldwide, prompting research into novel pharmacotherapies for alcohol use disorder (AUD). This study investigated the therapeutic potential of full agonism or positive allosteric modulation of the serotonin 2C receptor (5-HT2CR) in addressing alcohol binge drinking and seeking behaviours in mice. Using a drinking-in-the-dark paradigm and a context-induced reinstatement model following punishment-imposed abstinence, we assessed the acute effects of 5-HT2CR ligands lorcaserin, CYD-1-79, VA012 and CTW0415 on alcohol intake and seeking b

Circulating Immune and Endocrine Markers in Currently Drinking and Abstinent Individuals With Alcohol Use Disorder and Controls.

Alcohol use disorder (AUD) is associated with changes in endocrine and immune system function. This study is a secondary analysis aimed at investigating changes in circulating immune and endocrine biomarkers in blood samples from three groups: (1) healthy controls (HC, N = 12), (2) AUD-currently drinking, nontreatment seeking (CD, N = 9), and (3) AUD-abstinent, treatment-seeking (AB, N = 10; abstinent for at least 6 weeks). We hypothesized that both immune and endocrine biomarker concentrations would be different in AUD groups compared to healthy controls. Immune biomarkers included IL-8, IL-1

A multi-omics Mendelian randomization study identifies new therapeutic targets for alcohol use disorder and problem drinking.

Integrating proteomic and transcriptomic data with genetic architectures of problematic alcohol use and alcohol consumption behaviours can advance our understanding and help identify therapeutic targets. We conducted systematic screens using genome-wise association study data from ~3,500 cortical proteins (N = 722) and ~6,100 genes in 8 canonical brain cell types (N = 192) with 4 alcohol-related outcomes (N ≤ 537,349), identifying 217 cortical proteins and 255 cell-type genes associated with these behaviours, with 36 proteins and 37 cell-type genes being new. Although there was limited overlap

Neurocognitive Markers during Maximum Alcohol Use in Late Adolescence as Predictors of Change in Later Drinking Behaviors.

BACKGROUND: The influence of alcohol use on later neurocognitive functioning is well researched, yet few studies have investigated whether neurocognition post-drinking initiation in adolescence predicts changes in later alcohol use. OBJECTIVE: Investigate neurocognitive task performance during maximum alcohol use in late adolescence as predictors of drinking behaviors 3-7 years later. METHODS: Analyses (n=105) were conducted on a longitudinal dataset involving adolescents (12-13 years-old) who were followed for 16 years. Time 1 (T1) was defined as the individuals' maximum drinking year withi

Race/Ethnicity and Contexts: Associations of Racial/Ethnic Discrimination with Underage Youth's Alcohol Use and Drinking Contexts.

BACKGROUND: This study examined how racial/ethnic discrimination may shape alcohol use behaviors, related problems, and contexts of alcohol use among underage youth (<21 years old). Age group differences were also explored. METHODS: Cross-sectional data were obtained from California youth identified as racial and ethnic minoritized youth (n=605; ages 14-20). An adapted version of the Everyday Discrimination Scale was used to assess racial/ethnic discrimination. Demographics were also assessed. Outcomes included drinking status, past-6-month alcohol frequency and quantity, number of types of a

Influence of Drinking Context and Age of Onset on Alcohol Consumption Patterns and Motivations Among Uruguayan Adults.

OBJECTIVES: An early age of drinking onset is linked to a greater likelihood of alcohol-related problems. Alcohol use occurs in places featuring characteristic social groups, and different drinking contexts are associated with different levels of alcohol-related outcomes. Drinking context may affect drinking motives, expectations or alcohol-related outcomes in concert with individual-level variables. The study aimed to examine how the preferred drinking context and age of the first drink affects the occurrence and volume of alcohol use, drinking motives and alcohol-related expectations. METHO

Associations of habitual alcohol drinking with HDL cholesterol and D-dimer in patients with peripheral arterial disease.

BACKGROUND: The risk of cardiovascular disease is lower in light-to-moderate alcohol drinkers than in nondrinkers in the general population. However, it remains to be clarified whether these beneficial effects of alcohol are shown in patients with peripheral arterial disease (PAD). METHODS: The subjects were 153 male outpatients with PAD and they were divided by frequency of drinking into nondrinkers, occasional drinkers (1-4 days/wk) and regular drinkers (5-7 days/wk). Relationships of alcohol drinking with variables related to progression of atherosclerosis and cardiovascular risk were inve…

Alcohol-Drinking Under Limited-Access Procedures During Mature Adulthood Accelerates the Onset of Cognitive Impairment in Mice.

A history of heavy drinking increases vulnerability to, and the severity of, Alzheimer's disease (AD) and related dementias, with alcohol use disorder identified as the strongest modifiable risk factor for early-onset dementia. Heavy drinking has increased markedly in women over the past 10 years, particularly in mature adult women during the coronavirus (COVID-19) pandemic. This is concerning as women are more sensitive to many alcohol-related disease states, including AD and related dementias. Herein, we conducted two studies to determine if a 1-month period of binge drinking during mature a

Daily relationships between posttraumatic stress symptoms, drinking motives, and alcohol consumption in trauma-exposed sexual minority women.

Objective: Sexual minority women (SMW) evidence elevated rates of trauma exposure, posttraumatic stress (PTS), and alcohol consumption. Self-medication models of drinking suggest that individuals may drink to cope with symptoms of PTS, but this possibility remains untested among SMW. Method: The current study used data from drinking days collected via daily diary assessments from 81 trauma-exposed SMW. Participants were mostly White (72.8%) and ranged in age from 18 to 25 (M = 23.8). Participants were followed over 2 14-consecutive-day measurement bursts (720 person-days reported). Analyses we

Efficacy and safety of sodium oxybate in alcohol-dependent patients with a very high drinking risk level.

Medication development for alcohol relapse prevention or reduction of consumption is highly challenging due to methodological issues of pharmacotherapy trials. Existing approved medications are only modestly effective with many patients failing to benefit from these therapies. Therefore, there is a pressing need for other effective treatments with a different mechanism of action, especially for patients with very high (VH) drinking risk levels (DRL) because this is the most severely affected population of alcohol use disorder patients. Life expectancy of alcohol-dependent patients with a VH DR

Precision Medicine in Alcohol Dependence: A Controlled Trial Testing Pharmacotherapy Response Among Reward and Relief Drinking Phenotypes.

Randomized trials of medications for alcohol dependence (AD) often report no differences between active medications. Few studies in AD have tested hypotheses regarding which medication will work best for which patients (ie, precision medicine). The PREDICT study tested acamprosate and naltrexone vs placebo in 426 randomly assigned AD patients in a 3-month treatment. PREDICT proposed individuals whose drinking was driven by positive reinforcement (ie, reward drinkers) would have a better treatment response to naltrexone, whereas individuals whose drinking was driven by negative reinforcement (i

Safety Assessment of Liver Injury with Quetiapine Fumarate XR Management in Very Heavy Drinking Alcohol-Dependent Patients.

BACKGROUND: Studies have reported liver injury as a consequence of antipsychotic treatment. Very heavy alcohol drinking (ten or more drinks/day for men and eight for women) also causes liver injury. This study aims to evaluate liver injury with quetiapine extended release (XR) in very heavy drinking alcohol-dependent (AD) patients. METHODS: Two hundred and eighteen AD patients, 18-65 years of age, received 12 weeks of quetiapine XR or placebo treatment in a dose-escalated manner reaching the full dose of 400 mg/day during week 4. Blood chemistry and hematology were assessed at baseline (W0), …

Patterns of use of other drugs among those with alcohol dependence: Associations with drinking behavior and psychopathology.

INTRODUCTION: Alcohol dependence (AD) presents with substantial clinical heterogeneity, including concurrent use of non-alcohol drugs. Here, we examine specific patterns of concurrent non-alcohol substance use during the previous year among a nationally representative sample of adults with DSM-IV AD, and estimate their population prevalence in the U.S. We then evaluate alcohol use behavior and comorbid psychopathology among respondents with AD according to their patterns of concurrent non-alcohol substance use. METHODS: These analyses utilized data from Waves 1 and 2 of the National Epidemiol
