Coxiella burnetii in free-living feral pigs (Sus scrofa) in Brazil.

BACKGROUND: Coxiella burnetii, the etiological agent of coxiellosis in animals and Q fever in humans, is a zoonotic pathogen of global relev … spleen sample (1/36 - 2.8%; 95% CI: 0.1% - 14.5%). Only Coxiella-like endosymbiont was detected in Amblyomma sculptum ticks (9/23 - 39.13%; 95% CI: 22.2% - 59.2%). MAIN CONCLUSIONS: These results represent the first detection of C. burnetii in free-living feral pigs in Brazil and suggest potential exposure of this invasive mammal species to the pathogen. …

Diversity, Host Attachment Preferences and Role of Tick Rhipicephalus microplus in the Transmission of Bartonella spp. and Rickettsia spp. in Cattle From Southern Benin.

INTRODUCTION: Ticks, particularly Rhipicephalus microplus, are major blood-sucking ectoparasites of livestock, causing considerable economic … ventional PCR. RESULTS: The tick fauna was dominated by R. microplus, with additional detection of Amblyomma variegatum and other Hyalomma, Haemaphysalis and Rhipicephalus species. Ticks were mainly attached to ears, axillae and trunk. Spatial variation was observed but should be considered exploratory given the limited number of sampling sites. Molecular …

A Francisella tularensis Chitinase Contributes to Bacterial Persistence and Replication in Two Major U.S. Tick Vectors.

Nearly 100 years after the first report of tick-borne tularemia, questions remain about the tick vector(s) that pose the greatest risk for transmitting Francisella tularensis (Ft), the causative agent of tularemia. Additionally, few studies have identified genes/proteins required for Ft to infect, persist, and replicate in ticks. To answer questions about vector competence and Ft transmission by ticks, we infected Dermacentor variabilis (Dv),Amblyomma americanum (Aa), and Haemaphysalis longicornis (Hl; invasive species from Asia) ticks with Ft, finding that although Aa ticks initially become i…

Identification of Theileria fuliginosa-like species in Ixodes australiensis ticks from western grey kangaroos (Macropus fuliginosus) in Western Australia.

Piroplasms, including the genera Babesia and Theileria, are intra-erythrocytic protozoa that are generally transmitted by ticks and are the aetiological agents for piroplasmosis in animals, as well as humans, worldwide. In Australia, numerous studies have been conducted on piroplasms in domestic animals; however, less is known about these protozoa in ticks from native wildlife. The present study characterised piroplasms in Ixodes australiensis (n = 119) and Amblyomma triguttatum (n = 35) ticks collected from kangaroos in Western Australia (WA). Approximately 7.6% (9/119) (95% CI 2.8-12.2) of t…

Proteasomal degradation of intracellularly expressed Amblyomin-X limits suicide gene therapy potential in melanoma cells.

Amblyomin-X is a complex Kunitz-type protease inhibitor from the tick Amblyomma sculptum, with selective pro-apoptotic effects in tumor cells. While promising when applied exogenously, its large-scale recombinant production is hindered by structural complexity. This study evaluated the feasibility of expressing Amblyomin-X via a suicide gene therapy approach and investigated its intracellular fate following gene delivery in human melanoma and nontumoral cell lines. Amblyomin-X transcripts were equally detected in both cell types; however, protein detection was restricted to nontumoral cells. T…

Heartland Virus Disease-An Underreported Emerging Infection.

First recognized 15 years ago, Heartland virus disease (Heartland) is a tickborne infection contracted from the transmission of Heartland virus (HRTV) through tick bites from the lone star tick (Amblyomma americanum) and potentially other tick species. Heartland symptoms include a fever <100.4 °F, lethargy, fatigue, headaches, myalgia, a loss of appetite, nausea, diarrhea, weight loss, arthralgia, leukopenia and thrombocytopenia. We reviewed the existing peer-reviewed literature for HRTV and Heartland to more completely characterize this rarely reported, recently discovered illness. The absenc…

Molecular methods routinely used to detect Coxiella burnetii in ticks cross-react with Coxiella-like bacteria.

BACKGROUND: Q fever is a widespread zoonotic disease caused by Coxiella burnetii. Ticks may act as vectors, and many epidemiological studies aim to assess C. burnetii prevalence in ticks. Because ticks may also be infected with Coxiella-like bacteria, screening tools that differentiate between C. burnetii and Coxiella-like bacteria are essential. METHODS: In this study, we screened tick specimens from 10 species (Ornithodoros rostratus, O. peruvianus, O. capensis, Ixodes ricinus, Rhipicephalus annulatus, R. decoloratus, R. geigy, O. sonrai, O. occidentalis, and Amblyomma cajennense) known to …
