Detecting replication-associated single-stranded DNA.

Single-stranded DNA (ssDNA) is used as a readout of replication stress, yet it arises in several contexts. It is a normal intermediate of DNA replication but can accumulate and persist after replication challenges and is also generated by replication-independent processes. These structures differ in strand origin, architecture, biological consequence, and therapeutic response, so ssDNA detection alone rarely identifies the underlying molecular intermediate or how it was generated. Here, we describe the principal sources of replication-associated ssDNA and review how different methods capture d

'Guide and Prejudice'- How Argonautes recognize targets across domains of life.

Argonaute proteins mediate RNA interference in eukaryotes and nucleic-acid defence in prokaryotes. Although these systems operate in distinct biological contexts and produce diverse downstream outcomes, they share a conserved structural architecture and mechanism of guide-dependent target recognition. Here, we argue that Argonautes function as programmable surveillance platforms that kinetically sample nucleic acids, engage targets through sequential pairing and conformational checkpoints, and couple recognition to functional outputs through regulated structural transitions. Rather than repres

Structure-Regulated Silk Fibroin Microneedle Patches for Minimally Invasive Glucose-Responsive Colorimetric Sensing.

Minimally invasive microneedle devices have emerged as highly promising platforms for localized interstitial fluid (ISF) sensing and biomarker detection. In this study, we present a bilayer silk fibroin microneedle system that leverages material structure regulation to achieve optimal mechanical strength, controlled swelling, and limited dissolution. Our dual-layer architecture spatially separates skin penetration and local interstitial-fluid access from the sensing region, improving reagent retention and enabling clear, localized colorimetric readouts. Furthermore, the incorporation of 2-hydr

Mechanochemistry-Accessed MOF Polymorphism Revealed by an Integrated 3DED/DFT-Assisted Rietveld Analysis.

A combined 3DED, plane-wave DFT, and Rietveld refinement workflow uncovers the first example of interlayer-twisting polymorphism in a pillared MOF. While the parent material, PUM198, a Zn-based mixed-ligand MOF, was previously synthesized under solvothermal conditions, mechanochemistry selectively affords a new polymorph, PUM198-m. Although the two phases share identical composition and connectivity, they differ in symmetry, layer arrangement, pore architecture, and topology. Accessible only as a microcrystalline powder, PUM198-m is unsuitable for conventional single-crystal X-ray diffraction,

Enantioselective Radical-Radical Cross-Couplings of β-Hydroxy Amides and N-Hydroxyphthalimide Esters via Ni/Photoredox Catalysis.

Enantioselective alkyl-alkyl cross-coupling is a powerful yet challenging strategy for constructing three-dimensional molecular architectures, which are essential in fields such as organic chemistry and pharmaceutical chemistry. While radical-radical cross-coupling offers a promising approach, achieving control over both cross- and enantioselectivity between two distinct alkyl radicals remains a formidable challenge due to their transient nature. In this article, we introduce a practical platform that combines photoredox and chiral nickel catalysis to tame transient primary and secondary alkyl

The late frontal positivity reflects incremental mental model updating: Graded predictability effects during authentic text reading.

High word predictability facilitates the access and integration of word meaning, indicated by a reduced N400 effect. However, whether word predictability affects later phases of reading comprehension (mental model updating), as assessed by a late positivity, remains unsettled. Some studies suggest that unexpected but plausible words increase positivity in frontal regions, while others do not. To gain clarity on this issue, we used two-sentence passages from The New York Times articles that do not contain implausible words. Using a language model with a transformer architecture, we assessed the

Predicting progression events in multiple myeloma from routine blood work.

This study introduces a system for predicting disease progression events in multiple myeloma patients from the CoMMpass study (N = 1186). Utilizing a hybrid neural network architecture, our model predicts future blood work from historical lab results with high accuracy, significantly outperforming baseline estimators for key disease parameters. Disease progression events are annotated in the forecasted data, predicting these events with significant reliability. We externally validated our model using the GMMG-MM5 study dataset (N = 504), and could reproduce the main results of our study. Our a

Distinct type I and II interferon responses direct cortical and medullary thymic epithelial cell development.

Advances in genomics have redefined our understanding of thymic epithelial heterogeneity and architecture, yet signals driving thymic epithelial differentiation remain incompletely understood. Here, we elucidated pathways instructing human thymic epithelial cell development in the context of other anterior foregut-derived organs. Activation of interferon response gene regulatory networks distinguished epithelial cells of the thymus from those of other anterior foregut-derived organs. Thymic cortex and medulla epithelia displayed distinctive interferon-responsive signatures defined by lineage-s

Tri- and Tetra-ortho-Substituted Biaryls via Lithium Cuprate-Catalyzed Reductive Desymmetrization of Isophthaldehydes.

Polyfunctionalized chiral biaryls, especially those with dense ortho substitution patterns, are important building blocks for catalysts, pharmaceuticals, and advanced materials. Stereoselective synthesis of these congested architectures while simultaneously introducing multiple sites for further modification is highly sought after. Here, we report that a combination of an aziridinyl methanol-derived tetradentate ligand and a lithium cuprate salt can differentiate between the pair of aldehydes in biaryl isophthaldehydes to enable a desymmetric hydrosilylation. DDX1 methylation mediated MATR3 splicing regulates intervertebral disc degeneration by initiating chromatin reprogramming.

Low back pain (LBP), primarily driven by intervertebral disc degeneration (IVDD), has become a core challenge in public health. DDX1, an RNA-binding protein, plays key roles in RNA metabolism but its function in IVDD remains unclear. We identify DDX1 as a substrate of methyltransferase EZH2, which methylates DDX1 at lysine 234 (K234), promoting IVDD in vitro and in vivo. EZH2 inhibition restores matrix homeostasis in nucleus pulposus (NP) cells and slows IVDD progression. Methylation at DDX1 K234 disrupts its interaction with splicing factors and RNA targets, promoting exon 14 skipping in MATR

Gut microbe-derived metabolites drive psoriatic inflammation via modulation of skin Th17 cells.

The persistence of tissue-specific chronic inflammation results from an interplay of genetic and environmental factors. How these factors coordinate to sustain pathology in chronic conditions like psoriasis is not well resolved. Using a Card14E138A/+ murine model of psoriasis, we found that spontaneous skin inflammation reshaped not only the immune architecture in the skin but also systemic metabolites. We identified indole-producing microbiota in the gut-but not the skin-as drivers of psoriatic inflammation. Mechanistically, indole-producing intestinal microbes promoted host indoxyl sulfate (

Opportunities for Microphysiological Systems in Toxicity Testing of New Drug Modalities.

New drug modalities offer life-saving benefits for patients through access to previously undruggable targets. Yet these modalities pose a challenge for the pharmaceutical industry, as side effects are complex, unpredictable, and often uniquely human. With animal studies having limited predictive value due to translatability challenges, the pharmaceutical industry seeks out new approach methodologies. Microphysiological systems (MPS) offer important features that enable complex toxicological processes to be modeled in vitro such as (a) an adjustable complexity of cellular components, including

Lightweight convolutional neural network for chest X-ray images classification.

In this study, we developed a lightweight and rapid convolutional neural network (CNN) architecture for chest X-ray images; it primarily consists of a redesigned feature extraction (FE) module and multiscale feature (MF) module and validated using publicly available COVID-19 datasets. Experiments were conducted on multiple updated versions of the COVID-19 Radiography Database, a publicly accessible dataset on Kaggle. The database contained images categorized into three classes: COVID-19 coronavirus, viral or bacterial pneumonia, and normal. The results revealed that the proposed method achieve

Low Power In-Memory Computation with Reciprocal Ferromagnet/Topological Insulator Heterostructures.

The surface state of a 3D topological insulator (3DTI) is a spin-momentum locked conductive state, whose large spin hall angle can be used for the energy-efficient spin-orbit torque based switching of an overlying ferromagnet (FM). Conversely, the gated switching of the magnetization of a separate FM in or out of the TI surface plane can turn on and off the TI surface current. By exploiting this reciprocal behavior, we can use two FM/3DTI heterostructures to design an integrated 1-transistor 1-magnetic tunnel junction random access memory unit (1T1MTJ RAM) for an ultra low power Processing-in-

Peculiarities of Plasmodium falciparum Gene Regulation and Chromatin Structure.

The highly complex life cycle of the human malaria parasite, Plasmodium falciparum, is based on an orchestrated and tightly regulated gene expression program. In general, eukaryotic transcription regulation is determined by a combination of sequence-specific transcription factors binding to regulatory DNA elements and the packaging of DNA into chromatin as an additional layer. The accessibility of regulatory DNA elements is controlled by the nucleosome occupancy and changes of their positions by an active process called nucleosome remodeling. These epigenetic mechanisms are poorly explored in

Treponema pallidum Disrupts VE-Cadherin Intercellular Junctions and Traverses Endothelial Barriers Using a Cholesterol-Dependent Mechanism.

Treponema pallidum subspecies pallidum, the causative agent of syphilis, traverses the vascular endothelium to gain access to underlying tissue sites. Herein, we investigate the mechanisms associated with T. pallidum traversal of endothelial barriers. Immunofluorescence microscopy reveals that a subpopulation of T. pallidum localizes to intercellular junctions and that viable T. pallidum, as well as a T. pallidum vascular adhesin (Tp0751), disrupts the architecture of the main endothelial junctional protein VE-cadherin. Intriguingly, in this study we show that T. pallidum traverses endothelial

Junction Mapper is a novel computer vision tool to decipher cell-cell contact phenotypes.

Stable cell-cell contacts underpin tissue architecture and organization. Quantification of junctions of mammalian epithelia requires laborious manual measurements that are a major roadblock for mechanistic studies. We designed Junction Mapper as an open access, semi-automated software that defines the status of adhesiveness via the simultaneous measurement of pre-defined parameters at cell-cell contacts. It identifies contacting interfaces and corners with minimal user input and quantifies length, area and intensity of junction markers. Its ability to measure fragmented junctions is unique. Im

Genetic architecture and adaptive significance of the selfing syndrome in Capsella.

The transition from outcrossing to predominant self-fertilization is one of the most common evolutionary transitions in flowering plants. This shift is often accompanied by a suite of changes in floral and reproductive characters termed the selfing syndrome. Here, we characterize the genetic architecture and evolutionary forces underlying evolution of the selfing syndrome in Capsella rubella following its recent divergence from the outcrossing ancestor C. grandiflora. We conduct genotyping by multiplexed shotgun sequencing and map floral and reproductive traits in a large (N= 550) F2 populatio

Microfluidic serial dilution circuit.

In vitro evolution of RNA molecules requires a method for executing many consecutive serial dilutions. To solve this problem, a microfluidic circuit has been fabricated in a three-layer glass-PDMS-glass device. The 400-nL serial dilution circuit contains five integrated membrane valves: three two-way valves arranged in a loop to drive cyclic mixing of the diluent and carryover, and two bus valves to control fluidic access to the circuit through input and output channels. By varying the valve placement in the circuit, carryover fractions from 0.04 to 0.2 were obtained. Each dilution process, wh
