Acral lentiginous melanoma is a rare variant of melanoma that is associated with a relatively low survival rate. The latter is partly due to the advanced stage in which the tumor is usually diagnosed. The diagnostic delay is mainly due to difficulties in identifying the very early histopathological signs of acral melanoma. The current article is a review of diagnostic clues, concepts, and definitions from the literature, as well as illustrating examples from our own archives. We have sought to provide an article that can be easily consulted in difficult cases of acral lentiginous melanoma.

The excavation of the mass grave from the Monastery of the Observance dating from the Great Plague of Marseilles revealed the first evidence of an autopsy on the skull of a 15-year-old boy, performed during the spring of 1722. The reconstruction of the skull allows us to reconstitute the anatomic technique used, which is identical to those described in a surgical book dating from 1708. This research highlights the interest in correlating the biological and historical archives, and brings new anthropological data to the debate on the contagious nature of the plague.

Formalin-fixed paraffin-embedded (FFPE) tissues are archived resources for biomedical research but challenging for single-cell transcriptomic analysis. Here, we present a protocol for isolating high-quality nuclei from FFPE pancreatic cancer sections. We describe steps for deparaffinization, enzymatic digestion, nuclei purification, and library construction using the 10× Genomics Fixed RNA Profiling platform. BACKGROUND: Ageing and insulin resistant states such as diabetes mellitus frequently coexist and increase the risk of cardiovascular disease development among older adults. Here we investigate metabolic differences in amino acid profiles between ageing and diabetes mellitus, and their associations with cardiovascular function.

The purpose of this study was to describe the frequency of absent, unrecognized, or minimal myotonic discharges (MDs) in myotonic dystrophy type 2 (DM2). We performed a retrospective review of needle electromyography (EMG) data prior to genetic diagnosis in 49 DM2 patients at the Mayo Clinic.
