Metabolomics and image interpolation reveal secondary metabolite regulation in co-cultures of endophytic Penicillium cataractarum and Sclerotinia sclerotiorum.

Competition among microorganisms can activate cryptic genes, leading to the production of secondary metabolites that play roles in microbial defense and communication. Co-culture is an efficient strategy to induce metabolite biosynthesis, modulate biosynthetic gene clusters, and change metabolite abundance, thereby providing insights into microbial interactions with phytopathogens and potential approaches for agricultural pest control research. In this study, we investigated the co-culture interaction between the endophytic fungus Penicillium cataractarum 5IP2F4 (PC) and the phytopathogen Scle

Why Peacock Eye Severity Varies with Olive Tree Orientation: Defining Criteria to Predict Infection Events.

Peacock eye disease, caused by the hemibiotrophic fungus Venturia oleaginea, is the most destructive and widespread disease of olive in Israel. In a national project to develop an environmentally friendly peacock eye disease-management protocol for the local olive industry, it was found that one or a few fungicide applications in the autumn effectively suppressed disease development in the concurrent season. Since fungicide application timing should be based on environmental criteria rather than a predefined seasonal schedule, the goal of the current study was to define criteria for predicting

Natural cross-kingdom transmission of a novel ssDNA mycovirus confers broad-spectrum resistance to plant diseases.

Cross-kingdom virus transmission between plants and fungi remains rare in nature. Here, a novel circular ssDNA virus, Diaporthe pseudophoenicicola DNA virus 1 (DpDV1), was identified from the phytopathogenic fungus Diaporthe pseudophoenicicola. DpDV1 attenuates the pathogenicity of both its original and experimental host fungi, demonstrating its potential as a biocontrol agent against fungal phytopathogens. Remarkably, DpDV1 displays bidirectional plant-fungal transmission capability without inducing observable phenotypic changes in plant hosts, overcoming the limitations imposed by vegetative

Nicotinamide and Sodium Butyrate Double the Pool of Treatment-Exclusive Metabolites in the Endophytic Fungus Curvularia sp. G6-32.

Endophytic fungi are prolific sources of bioactive natural products, but many biosynthetic gene clusters remain silent under laboratory conditions. Following the OSMAC approach, the epigenetic modifiers nicotinamide and sodium butyrate (10 µM) were added to triplicate liquid cultures of Curvularia sp. G6-32, an endophyte of Sapindus saponaria, and the crude extracts were analyzed by UHPLC-MS/MS with Mzmine 3 processing and GNPS molecular networking. In total, 712 features were detected: 474 common to all treatments, 65 exclusive to the control, and 154 occurring only in the presence of the mod

Co-Infecting Mycoviruses VdPV1 and VdMoV1 Attenuate Verticillium dahliae and Are Transmitted Vertically and Horizontally.

Cotton verticillium wilt caused by Verticillium dahliae severely restricts global cotton production. Mycovirus-induced hypovirulence provides a promising strategy for sustainable disease management. In this study, V. dahliae isolates from Xinjiang were screened by virome sequencing and RT-PCR, and strain 121-11C-1 co-infected with Verticillium dahliae partitivirus 1 (VdPV1) and Verticillium dahliae magoulivirus 1 (VdMoV1) was identified. Virus-free isogenic strains were obtained via single-conidium purification, verifying that both viruses can be vertically transmitted through fungal conidia.

Protease-Mediated Dual Action Against Rice Blast: Suppression of Magnaporthe oryzae Ergosterol Biosynthesis Pathway and Enhancement of Rice Disease Resistance by Bacillus amyloliquefaciens SN16-1.

Rice blast, caused by the fungal pathogen Magnaporthe oryzae, poses a severe threat to global rice production. While biological control represents a promising sustainable strategy, the molecular mechanisms by which biocontrol agents suppress M. oryzae remain poorly understood. Our findings elucidate a novel mechanism of bacterial biocontrol, wherein a secreted bacterial protease modulates fungal signalling pathways to disrupt its environmental fitness and pathogenic structures, while enhancing host resistance to blast disease. We demonstrate that the Bacillus amyloliquefaciens SN16-1 exhibits

Re-defining the culturing method for Pseudogymnoascus destructans incorporating viability detection.

Pseudogymnoascus destructans (Pd) is a psychrophilic fungus whose genus is commonly found in soils of colder environments, including stable habitats such as caves and mines. The fungus causes the epizootic disease white-nose syndrome, which is responsible for the mortality of millions of bats (Chiroptera) in North America through disruption of the hibernation cycle and of gas/water exchange processes that take place across wing membranes. To understand pathogenicity and fungal ecology with the goal of intervention, cultivation of Pd is essential. However, previous research on the fungus does n

The biocontrol effect and mechanism of the marine-derived fungus Epicoccum nigrum against Alternaria alternata, the causal agent of tomato black spot disease.

Tomato black spot disease, caused by the fungus Alternaria alternata, leads to plant wilting and chlorosis and results in sunken, black lesions on harvested fruit, thereby causing significant economic losses in the tomato industry. To mitigate issues related to environmental contamination and the emergence of resistant pathogens, this study investigated a high-efficacy, low-toxicity biocontrol strategy. The marine-derived fungus Q8-1 inhibited the growth of A. alternata by up to 87.04 %. Based on morphological features and ITS rDNA sequencing, the strain was identified as Epicoccum nigrum. Gen

The In Vitro and In Vivo Fungal Volatile Organic Compounds Associated with Rapid 'Ōhi'a Death and the Response of Xyleborine Ambrosia Beetles to those Compounds.

Rapid 'ōhi'a death (ROD), caused by fungal pathogens Ceratocystis lukuohia and Ceratocystis huliohia, is devastating 'ōhi'a (Metrosideros polymorpha), the keystone forest tree species of Hawai'i. Ceratocystidaceae fungi produce fruity volatile organic compounds (VOCs) and sticky sexual spores, suggesting they are adapted for insect dispersal. In this study, we explored chemical signaling of the causal agents of the ROD disease complex and how those VOCs mediate ambrosia beetle (Coleoptera: Curculiondiae: Scolytidae)-plant interactions. Four species of ambrosia beetles (Xyleborinus saxesenii, X

Extraction and Identification of the Bioactive Metabolites Produced by Curvularia inaequalis, an Endophytic Fungus Collected in Iran from Echium khuzistanicum Mozaff.

Endophytic fungi (EF) are microorganisms that colonize the internal tissues of host plants, providing a range of benefits to them. In this symbiosis, they act as a reservoir of bioactive metabolites that are important for enhancing the host's defense mechanisms as a resistance against pathogens. These molecules usually possess antimicrobial properties that can be exploited for application in agriculture and medicine. In this context, the current work was designed to evaluate the phytotoxic and antimicrobial properties of the endophytic fungus Curvularia inaequalis, isolated for the first time

An efficient harvesting strategy for agarwood based on the correlation analysis of resin formation and leaves dynamic changes induced by integrated induction method.

Agarwood is a resin produced by wounded Aquilaria plants. Aquilaria sinensis (Lour.) Gilg  is the original plant source of agarwood in China. Formic acid combined with Botryosphaeria rhodina A13 (FAA13) induces the formation of artificial agarwood as an effective integrated induction method. However, its formation mechanism is still unclear, and the harvesting time of agarwood has not been elucidated. In this work, we analyzed FAA13-induced artificial agarwood and leaves at different time points within one year based on endophytic fungal community, expression of related genes, and secondary me

Design, synthesis and bioactivity of a new class of antifungal amino acid-directed phthalide compounds.

BACKGROUND: Peanut southern blight disease, caused by Sclerotium rolfsii, is a destructive soil-borne fungal disease. The current control measures, which mainly employ succinate dehydrogenase inhibitors, are prone to resistance and toxicity to non-target organisms. As a result, it is necessary to explore the potential of eco-friendly fungicides for this disease. RESULTS: Fourteen novel phthalide compounds incorporating amino acid moieties were designed and synthesized. The in vitro activity of analog A1 [half maximal effective concentration (EC50) = 332.21 mg L-1] was slightly lower than that
