Nucleotide substitutions are common in cancer cells, and they occur in both protein coding regions and non-coding regions (5' and 3' UTRs and introns). Although substitutions in non-coding regions have the potential to alter gene expression, it is the alteration of coding regions that affects protein function and has the most drastic effect on cellular transformation. Mutations in certain genes (e.g., TP53, KRAS) are common to nearly all cancers, but most cancers are characterized by specific gene mutation signatures. In this report, we investigated nucleotide substitution signatures in coding

The storage and transfusion of red cells resuspended in an additive solution has resulted in a substantial decrease in the use of whole blood. In the Auckland transfusion region, 87% of red cells transfused are now in this new form. This has not resulted in a commensurate increase in the use of plasma volume expanders, total usage of which has remained relatively static. The improved availability of plasma for blood component production is an important side benefit of the procedure which has also gained satisfactory clinical acceptance. The routine use of whole blood is a wasteful and ineffici

AIM: Lumbricus terrestris (earthworm) erythrocruorin (LtEc) is a naturally occurring extracellular hemoglobin (Hb) with high molecular weight (3.6MDa), low autoxidation rate, and limited nitric oxide (NO) dioxygenation activity. These properties make LtEc a potential candidate for use as red blood cell (RBC) substitute, i.e. Hb-based oxygen carrier (HBOC). Previous studies have shown that small amounts of LtEc can be safely transfused into mice, rats, and hamsters without eliciting major side effects. Therefore, this study was designed to understand oxygen (O(2)) transport to tissues and syste

We studied the effects of rat stroma-free hemoglobin (rSFH), human stroma-free hemoglobin (hSFH), rat polyhemoglobin (rPoly), and human polyhemoglobin (hPoly) on coagulation factors in rats. Albumin and saline infused rats were controls. The infusion volume was 10% of the rat's blood volume. The concentrations of hemoglobin in this study were 7 g/dl. Measurements for prothrombin time (PT) and activated partial thromboplastin time (PTT) were at 5 minutes, 2, 6, 24 and 72 hours after infusion. Factor X, fibrinogen, plasminogen, antithrombin III, and antiplasmin were followed at 24 and 72 hours a

BACKGROUND: In this study, we prepared acetated-Ringer containing 10% dextran, and evaluated its efficacy in a rat model of acute normovolemic hemodilution (ANH).
METHODS: Under sevoflurane anesthesia, 21 rats (350-400 g) underwent ANH. In all rats 10 ml of blood was withdrawn, and they were infused with 10 ml of acetated-Ringer containing 10% dextran (Group-A, n = 11) or lactated-Ringer containing 10% dextran (Group-L, n = 10). For each rat, ANH procedure took 20 minutes. Hemodynamics and arterial blood gas data before and after ANH were compared.
RESULTS: All results but for blood lactate

Pathomorphological changes in the lungs in the period of clinical death due to acute blood loss and within 1 1/2--2 hours of resuscitation using intraarterial, intravenous, and intraosteal routes for blood loss substitution were studied in dogs. In the period of clinical death significant changes in the lung morphology were observed (capillary plethora, sludge, leukostasis, hemorrhages, fat embolism, irregular aeration of pneumonic tissue), in the early postresuscitation period the intensity of the changes varied in relation to the method of blood loss substitution. The above-mentioned methods
