A fundamental difference in the capacity to induce proliferation of naive T cells between CD28 and other co-stimulatory molecules.

T cell activation requires two signals: a signal from the TCR and a co-stimulatory signal provided by antigen-presenting cells (APC). In addition to CD28, multiple molecules on the T cell have been described to deliver co-stimulatory signals. Here, we investigated whether there exist quantitative or qualitative differences in the co-stimulatory capacity between CD28 and other molecules. Anti-CD28 monoclonal antibody (mAb) and mAb against CD5, CD9, CD2, CD44 or CD11a all induced activation of naive T cells in the absence of APC when co-immobilized with a submitogenic dose of anti-CD3 mAb. In vivo synchronous membrane potential oscillations in mouse pancreatic beta-cells: lack of co-ordination between islets.

1. The properties of the oscillations in electrical activity of different beta-cells within the same islet of Langerhans, and of different islets within the same pancreas, recorded in vivo, are described. 2. Simultaneous recordings of two cells within the same islet showed that the oscillations were synchronous. A rapid increase in blood glucose led to the simultaneous appearance of a transitory phase of continuous electrical activity in both cells. These results indicate that under physiological conditions, the islets operate as a functional syncytium. 3. Simultaneous recordings of cells from

Co-amplification of the gene for parathyroid hormone-related protein (PTHRP) and KRAS2 in a human lung cancer cell line.

Parathyroid hormone-related protein (PTHRP) is expressed in a large number of tumors and is the mediator of parathyroid hormone-like effects seen in humoral hypercalcemia of malignancy. The gene coding for PTHRP has been localised to the short arm of chromosome 12. This is at the same region as the oncogene KRAS2, and amplification of KRAS2 has previously been found in human lung cancer. The BEN cell line which is known to express PTHRP was established from a patient who had squamous cell carcinoma of the lung with hypercalcemia. Cytogenetic analysis of the BEN cell line revealed a very comple

CAR-T cells co-expressing IL-7 and CCL19 promote epitope spreading to enhance antitumor immunity.

Antigen heterogeneity remains a major obstacle to the effective application of chimeric antigen receptor (CAR)-T cell therapy in solid tumors. We investigated the potential of epitope spreading as a strategy to overcome this limitation and examined whether CAR-T cells concomitantly producing IL-7 and CCL19 (7 × 19 CAR-T cells) could act as potent inducers of epitope spreading, as well as the underlying mechanisms. We used a murine model inoculated with a mixture of cancer cell lines-genetically modified to express the CAR target antigen-and their respective parental lines lacking target expres

Co-encapsulated resveratrol and paclitaxel in liposomes coating with RGDfC peptide for drug resistance reversal in renal cell carcinoma.

BACKGROUND: Drug resistance critically impedes renal cell carcinoma therapy. A dual-targeted liposomal system that codelivered resveratrol (RES) and paclitaxel (PTX) with αvβ3 integrin-targeting was developed to overcome multidrug resistance (MDR). METHODS: RGDfC peptide-functionalized liposomes (PTX/RES-RGDfC-Lip) were synthesized via film hydration-ultrasound methods. The particle size, zeta potential, drug encapsulation efficiency, and release kinetics were evaluated. P-glycoprotein (P-gp) modulation, cellular uptake, and chemosensitization in PTX-resistant A498 cells (PTX-res-A498) and in

Integrative multiomic analysis reveals co-ordinated alternative splicing in human bone marrow stromal stem cells.

Alternative splicing is essential for the production of diverse messenger RNAs from a single gene. However our understanding of alternative splicing and its regulation during processes such as osteoblast differentiation remains incomplete. Using differentiating hMSC-TERT4 cells as a model, we performed deep short- and long-read RNA sequencing, revealing a highly integrated multiphasic differentiation program. Analysis for splicing revealed extensive changes during lineage commitment, in genes associated with known transcriptional regulators including RUNX2, TEAD1, CTNNB1 and NFATC4. During lat

Dense and Uniform Co-Self-Assembled Monolayers for Modifying NiOX toward High-Performance Perovskite Solar Cells.

Self-assembled molecules (SAMs) have emerged as an efficient strategy for addressing buried nickel oxide (NiOX) interface defects in inverted perovskite solar cells (PSCs). Nevertheless, conventional carbazole-based SAMs suffer from weak intermolecular interactions, leading to film aggregation that hinders the formation of dense and uniform SAMs, which in turn causes discontinuities in the contact between NiOX and the perovskite film, thereby inducing interfacial defects and nonradiative recombination. Herein, 1-(2-carboxyethyl)-2,3,3-trimethyl-3H-indol-1-ium (CTI) was combined with 2PACz to p

An oxygen-glucose co-releasing platform fostering dental pulp regeneration by driving metabolic recovery of stem cells.

In dental pulp regeneration, the ischemic microenvironment within the root canal severely compromises the survival and function of transplanted human dental pulp stem cells (hDPSCs). Here, we developed a PDMS-based core-shell oxygen-glucose delivery platform (P-C@P-G) that continuously released oxygen and glucose for up to 40 and 29 days, respectively, while minimizing peroxide-associated cytotoxicity through diffusion-controlled regulation without the need for exogenous enzymes. Under oxygen-glucose deprivation (OGD, 0.1% O2, glucose-free conditions) conditions, P-C@P-G improved cellular meta

Identification of Human Transferrin Receptor as an Entry Co-receptor for Parvovirus B19 Infection of Human Erythroid Progenitor Cells.

UNLABELLED: Parvovirus B19 (B19V), a member of the genus Erythroparvovirus within the Parvoviridae family, infects human erythroid progenitor cells (EPCs) of bone marrow and fetal liver, and causes various hematological disorders. The minor capsid protein VP1 of B19V contains a unique N-terminal region (VP1u) that facilitates virus binding and internalization into EPCs via its receptor-binding domain (RBD). We previously identified tyrosine protein kinase receptor UFO (AXL) as a proteinaceous receptor for B19V infection of EPCs. In this study, we employed an ascorbate peroxidase 2 (APEX2)-base

Microfluidic Fibroblast Cell Culture Chip for Embryo Co-Culture: Analysis of Preimplantation Embryo Viability and Development Potential.

Preimplantation embryo development requires a tightly regulated microenvironment that is not fully reproduced by conventional static culture. We developed a polydimethylsiloxane-based microfluidic embryo co-culture platform integrating compartmentalized architecture with dynamic perfusion to simulate physiological conditions. The study included two stages. First, NIH/3T3 mouse fibroblasts were evaluated as helper cells under three culture conditions after transition to embryo culture medium. Helper-cell viability in the dynamic chip was 84.72%, compared with 71.91% after manual medium replacem

A high-throughput co-culture CRISPR array for rapid fitness hypothesis testing and growth engineering in CHO cells.

Cell growth and fitness shape mammalian biomanufacturing performance, but subtle genetic effects are difficult to resolve with conventional growth assays. We developed a high-throughput competitive co-culture CRISPR array in Chinese hamster ovary (CHO) suspension cells that pairs a CRISPR-competent population with a CRISPR-inactive green fluorescent protein (GFP) reference pool and tracks GFP-ratio dynamics by flow cytometry. Guided by a prior genome-wide CRISPR screen, we tested 235 single-gene knockouts and 155 multiplex gene sets across seven batches and >100 days of cultivation. The assay

Network-informed deconvolution of bulk immune gene co-expression reveals single-cell programs and spatial organization.

INTRODUCTION: Single-cell RNA sequencing (scRNA-seq) has opened unprecedented possibilities to explore the complexity of the immune system. However, existing methods primarily rely on expression-based clustering analysis, which lacks mechanistic explanations for immune cell states and encounters challenges in integrating multi-scale data. METHODS: We developed a network-informed deconvolution framework that constructs Bayesian network-derived regulatory structures using immune-related genes from context-matched bulk RNA-seq datasets. Network markers were extracted from these structures and pr

Co-designing a Novel Ordinal Endpoint for an Adaptive Platform Trial, BANDICOOT, in Pediatric Hematopoietic Stem Cell Transplant.

An adaptive platform trial (APT) offers the ability to incorporate several research questions in the same target population across multiple domains (interventions), with the ability to add new questions in a perpetual manner. An APT is particularly appealing for pediatric hematopoietic stem cell transplant (HCT); an area of high heterogeneity, limited trial availability, and high mortality. Ideally, all domains in an APT would have the same primary endpoint. Therefore, an ordinal endpoint with multiple categories that combines various clinical outcomes into a single outcome measure is particul

Superior preclinical efficacy of co-treatment with BRG1/BRM and FLT3 inhibitor against AML cells with FLT3 mutations.

Although treatment with standard frontline therapies, including a FLT3 inhibitor (FLT3i) reduces AML burden and achieves clinical remissions, most patients with AML with FLT3 mutation relapse due to therapy-resistant stem/progenitor cells. The core ATPases, BRG1 (SMARCA4) and BRM (SMARCA2) of the canonical (c) BAF (BRG1/BRM-associated factor) complex is a dependency in AML cells, including those harboring FLT3 mutations. We have previously reported that treatment with FHD-286, a BRG1/BRM ATPases inhibitor, induces differentiation and loss of viability of AML stem/progenitor cells. Findings of

Illuminating radionuclide therapy response: Monitoring cancer cell and CAF survival in a direct 2D co-culture model.

BACKGROUND: Targeted radionuclide therapy (TRT) directed at fibroblast activation protein (FAP), highly expressed on cancer-associated fibroblasts (CAFs), is a promising approach for treating stroma-rich tumors such as pancreatic ductal adenocarcinoma (PDAC) and breast cancer (BC). To better assess the efficacy of FAP-TRT and to get more insight into the potential of crossfire effects from target expressing CAFs to neighboring cancer cells, more representative preclinical models are needed. Therefore, we established a direct 2D co-culture model consisting of both CAFs and cancer cells. MATERI

Co-Delivery of Multiple RNAs via Lipid Nanoparticles Enables Precise Gene Editing of CAR-T Cells.

Over the past decade, CAR-T cell therapy has achieved remarkable success in treating hematological malignancies. However, traditional CAR-T cell engineering employs viral vectors, which has several limitations. Additionally, the immunosuppressive tumor microenvironment, particularly mediated by the PD-1/PD-L1 pathway, significantly restricts CAR-T cell efficacy. CRISPR/Cas9-mediated PD-1 knockout can enhance CAR-T cell anti-tumor activity, but traditional electroporation (EP) method often damages T cells. Herein, a novel lipid nanoparticles (LNPs)-mediated delivery technology are introduced to

Metabolic reprogramming by chemo-gene co-delivery nanoparticles for chemo-immunotherapy in head and neck squamous cell carcinoma.

The therapeutic effects of platinum-based drugs are closely linked to the dysregulation of tumor metabolic-immune microenvironment, particularly aberrant lactate accumulation. Herein, we engineered multifunctional nanoparticles (PPPtIV NPs) through electrostatic self-assembly of poly(β-amino ester) to co-encapsulate a cisplatin prodrug (PtIV) and CRISPR/Cas9-PKM2 plasmids. Mechanistically, PPPtIV NPs efficiently entered cells via endocytosis, followed by escape from lysosomal degradation and cargo release. The reduction of PtIV prodrug to active PtII via GSH depletion induced DNA damage and RO

Non-coding RNA modulation in osteoclasts and its implications for osteoblast lineage cell behavior in a co-culture system.

The interplay between osteoblasts and osteoclasts (OCs) is a highly regulated and coordinated process essential for maintaining bone skeletal integrity and health. Disruption of this balance marks the onset of various bone disorders, such as osteoporosis. In our previous study, we demonstrated that non-coding RNAs (ncRNAs) were able to regulate OC behaviour. Specifically, the long ncRNA DLEU1 and the small ncRNA miR-16 independently control OCs multinuclearity, fusion and resorption. Herein, we investigate whether OCs modulated with ncRNAs impact the behaviour of osteoblast lineage cells (OBs)
