Dietary Sheep Blood Zymolysate Modulates Gut Barrier Integrity, Microbiota Composition, and Nutritional Metabolism in Common Carp (Cyprinus carpio).

Sustainable protein sources are pivotal to the green development of aquaculture. As an underutilized abattoir byproduct, sheep blood holds great potential for valorization via enzymatic hydrolysis. This study evaluated the impacts of dietary sheep blood zymolysate (SBZ) supplementation (0.3%, 0.6%, and 1.0% dosages, denoted as TL, TM, and TH groups, respectively) on growth performance, serum biochemistry, intestinal barrier function, and gut microbiota of common carp (Cyprinus carpio). A total of 240 carp (initial weight: 48.0 ± 0.2 g) were randomly assigned to four groups (three replicates/gr

Physicochemical characteristics, gelation properties, and molecular mechanisms of grass carp (Ctenopharyngodon idella) myofibrillar proteins affected by the substitution of NaCl with CaHMB.

The construction of health-oriented fish protein products with desirable gel behavior as well as low salt content is urgently demanded by people nowadays. This study elucidated the substitution of sodium chloride (NaCl) with calcium β-hydroxy-β-methylbutyrate (CaHMB) in modulating the physicochemical and gel properties of grass carp myofibrillar proteins (gcMPs) at different substitution degrees. The results showed that the presence of CaHMB clearly reduced the fluorescence intensity and surface hydrophobicity of gcMPs, while increased their UV absorption intensity, representing the unfolding

Network meta-analysis of novel diagnostic biomarkers for rheumatoid arthritis: comparative performance of anti-CarP, anti-MCV, and emerging markers.

OBJECTIVE: Rheumatoid arthritis (RA) diagnosis remains challenging due to the limitations of conventional biomarkers. This network meta-analysis (NMA) aimed to compare the diagnostic performance of seven novel biomarkers for RA. METHODS: We systematically searched EMBASE, PubMed, and Cochrane databases, identifying 1,713 records. Twelve studies met inclusion criteria. Diagnostic accuracy metrics (sensitivity, specificity, log diagnostic odds ratio [lnDOR]) were extracted. A frequentist NMA was performed using the netmeta package in R, with biomarkers ranked via P-scores against the ACR/EULAR

Tauroursodeoxycholic acid (TUDCA) protects against soybean meal-induced hepatopancreatic steatosis and intestinal inflammation in grass carp (Ctenopharyngodon idellus).

This study evaluated the potential for tauroursodeoxycholic acid (TUDCA) to ameliorate the negative effects associated with high soybean meal (SBM) diets in the grass carp (Ctenopharyngodon idellus). Grass carp (initial weight 25.42 ± 0.16 g) were fed one of four diets for 8 weeks: a control diet (fish meal [FM]-based), Control+TUDCA (0.5 g/kg), a high SBM diet (100SBM, completely replacing FM), or 100SBM + TUDCA. The 100SBM diet reduced growth performance and feed efficiency. However, TUDCA supplementation did not reverse these growth deficits. The 100SBM diet induced hepatopancreatic lipid a

Multi-Omics Analysis Reveals Impacts of SCARB1, TYR, and TYRP1 Knockouts on Pigmentation and Metabolic Pathways in Oujiang Color Common Carp.

Gene knockout (KO) techniques are important for understanding gene function and their impact on organismal traits. This study investigates the effects of targeted KO of SCARB1, TYR, and TYRP1 genes in Oujiang Color common carp, focusing on pigmentation changes, metabolic dysregulation, and host-microbiome interactions. Our multi-omics approach revealed that SCARB1 KO led to complete loss of red pigmentation, while TYR and TYRP1 KOs resulted in hypopigmentation and altered pigmentation patterns. Transcriptomic analysis identified seven key genes (PLA2G4, C3, F2, ERN1, UGP2, purA, and GRIN2A), l

Transcriptomic and functional evidence reveals dual-module immune response of human-nasal staphylococcus aureus in Koi Carp (Cyprinus carpio).

Staphylococcus aureus is a significant pathogen that poses a threat to both human and animal health. Its pathogenicity in humans has been extensively studied, however, the signaling pathways and key genes in Koi Carp responding to S. aureus from human rhinitis remain unclear. In this study, we established an intraperitoneal infection model in koi carp (Cyprinus carpio) using an S. aureus isolate from patients with rhinitis and integrated RNA-seq, qPCR, and ELISA to dissect the host response. Our findings reveal a dual-module immune evasion strategy employed by S. aureus in koi carp. Module I:

Hsd3b7 regulates ovarian development and maturation via steroidogenesis and lipid metabolism in Qihe gibel carp (Carassius gibelio var. Qihe).

Steroid hormones regulate ovarian development, but hsd3b7 function in fish remains unclear. In this study, we cloned full-length hsd3b7 cDNA from Qihe gibel carp (Carassius gibelio var. Qihe) and generated a polyclonal antibody. Real-time PCR, Western blotting, and immunohistochemistry revealed widespread hsd3b7 expression in brain, heart, spleen, muscle and ovary. During ovarian development and maturation, hsd3b7 mRNA expression peaked at 150 days after hatching (dah) and remained at a relatively high level until 720 dah. Immunohistochemical analysis further revealed that Hsd3b7 protein was l

Characterization of Keap1a and Keap1b in grass carp (Ctenopharyngodon idella) and Keap1-Nrf2 pathway-related transcriptional responses under heat stress.

The Keap1-Nrf2 pathway is a critical pathway for cellular antioxidant defense, with Keap1 acting as its central regulator. Unlike mammals, which possess only a single Keap1 gene, fish have two keap1 paralogs, keap1a and keap1b, which may have functionally diverged. In this study, we cloned and characterized keap1a and keap1b genes in grass carp (Ctenopharyngodon idella) and investigated their responses to heat stress through both in vivo and in vitro experiments. Our results showed that keap1a was downregulated and nrf2 was upregulated in both the spleen and liver during heat stress. In contra

Effect of quorum quenching enzyme YtnP on immune response and intestinal microbiota in Aeromonas hydrophila-infected grass carp (Ctenopharyngodon idella).

Aeromonas hydrophila induces tissue swelling, necrosis, ulcers, and hemorrhagic septicemia in grass carp, causing significant economic losses. Quorum sensing, a communication mechanism among microorganisms, plays a key role in microbial ecological competition and dynamic equilibrium. Quorum quenching has emerged as a hot spot in anti-bacterial infection research. In this study, grass carp (n = 75 per group) were intraperitoneally injected with A. hydrophila and the quorum quenching enzyme YtnP. The expression levels of immune-related genes were assessed by qPCR. The intestinal microbiota was s

Effects of dietary quality on melatonin in gut, serum and ovary in mediating growth, digestive and ovarian physiology in juvenile carp (Catla catla).

The dietary protein and tryptophan of fish feed have vital roles in the growth performance, stress management, and digestive physiology of fish. However, in this context, the functions of gut melatonin, which depend on the availability of food, the timing of food supply, the frequency of feeds/day, the quality of food, and the growth stages of carp, still need to be clarified. Hence, the present study aimed to investigate the impact of dietary quality on specific growth rate (SGR), residual gut content (RGC), feeding intensity (FI), gut melatonin, gut oxidative stress and its major antioxidant

Analysis of star promoter in common carp and catfish testis: Role of c-jun and its association with testicular function as a transcription factor.

Steroidogenic acute regulatory protein (Star) is a key factor that predominantly mediates the rate-limiting step of steroidogenesis which involved in the translocation of cholesterol from the outer to inner mitochondrial membrane in most of the vertebrates. To understand the transcriptional regulation of star (star1) in fish, the 5' upstream region of the star was cloned from both common carp and catfish genomic libraries and in silico promoter motif analysis predicted putative binding sites of C-jun, AP-1, Gata2, and Oct1 in common carp while C-jun, Gata1, Gata2, and Oct1 binding sites were p

Type II interferons activate MHC-I pathway to enhance antigen presentation of grass carp reovirus VP35 DNA vaccine.

Hemorrhagic disease caused by grass carp reovirus (GCRV) poses a significant threat to the health and sustainability of grass carp (Ctenopharyngodon idella) farming. There are no effective measures to control the outbreaks of the disease. While DNA vaccines have proved to be promising to enhance the survival of vaccinated fish to GCRV infection, the protective efficacy is not maximized, and necessitates further improvement. This study explores the immunomodulatory potential of type II interferons (IFNs), including IFN-γ and IFN-γ related molecule (IFN-γrel), as adjuvants for GCRV-VP35 DNA vacc

Recirculating aquaculture-fasting strategy (RASF) to modulate gut microbiota and enhance the fish meat quality of bighead carp (Aristichthys nobilis).

Intensive aquaculture often leads to a decline in fish quality, and requires a strategy to restore the post-culture quality of fish. This study presents a novel recirculating aquaculture-fasting strategy (RASF) and assesses its impact on meat quality and gut microbiota of bighead carp. Results revealed RAS-F notably reduced body lipid and protein, achieving weight loss and body reshaping. RAS-F decreased the hardness, increased the springiness, and effectively improved the texture. Off-flavor compounds, especially aldehydes and ketones, were diminished, while umami amino acids (UAA) increased

Epigallocatechin-3-gallate supplementation regulates glucose redistribution through its potential endogenous receptor in grass carp (Ctenopharyngodon idella).

Exploring exogenous additives to enhance non-protein energy ingredient utilization in fish is highly anticipated. Epigallocatechin-3-gallate (EGCG) is a prominent polyphenol found in green tea. To assess the regulatory effect of EGCG on metabolic disorders induced by an excess of non-protein energy sources in grass carp (Ctenopharyngodon idella), five isonitrogenous diets were formulated: a control diet (23% digestible carbohydrates, 4.7% crude lipids), a high-carbohydrate (HC; 48% digestible carbohydrates, 4.7% crude lipids) diet, a high-fat (HF; 23% digestible carbohydrates, 9.6% crude lipid

Differences in flavor profiles and meat quality of grass carp from China's jingpo lake across four thermal processing methods.

This study examined the effects of four thermal processing methods (steaming, boiling, air frying, and roasting) on the flavor and meat quality of Grass Carp from Jingpo Lake, China (a natural water system). A total of 98 volatile substances were identified in Jingpo Lake Grass Carp (JPGC) meat after heat treatment. In the four treatments, the air frying could significantly decrease the content of unpleasant odor compounds (octanol, 1-octen-3-ol, 1-heptanol, hexanal, nonanal, and octanal). Electronic tongue analysis suggested a more appealing taste profile for air-fried samples compared to oth

Tissue Mosaicism in Xenogenic Common Carp (Cyprinus carpio) Injected with Unsorted Gonadal Cells from Blue Catfish (Ictalurus furcatus).

Xenogenic individuals are sterilized hosts that are colonized by germ cells transplanted from a donor species and can subsequently produce donor-derived gametes. In our case, unsorted gonadal cells from a donor species injected near the genital ridge of a sterile surrogate should colonize only that region, eventually becoming donor gametes. Unsorted gonadal cells from blue catfish (Ictalurus furcatus) injected into triploid common carp (Cyprinus carpio) embryos and fry were detected by PCR assays predominantly in recipient gonads and, in a subset of individuals, occasionally in DNA extracts fr

Nuclear scaffold attachment factor A functions as a potential viral recognition receptor involved in the antiviral immunity of grass carp (Ctenopharyngodon idella).

As one of the most primitive vertebrates, fish have evolved a distinct repertoire of viral recognition receptors. However, the existence of nuclear viral recognition receptors in fish remains uncertain. In this study, we identified a scaffold attachment factor A (SAFA), CiSAFA, as a potential nuclear recognition receptor for grass carp reovirus (GCRV) in grass carp (Ctenopharyngodon idella). CiSAFA shares high amino acid sequence similarity (70.3-99.6 %) and identity (56.1-98.6 %) with its counterparts from other vertebrates and contains three conserved domains, namely, SAP, SPRY, and AAA_33.
