Cell-mediated immunity to myoglobin in polymyositis.

Using the leucocyte migration inhibition (LIF) test we looked for evidence of cell-mediated hypersensitivity against myoglobin in 8 patients with polymyositis (PM) or dermatomyositis (DM). The migration index for PM-DM patients was 47.5 +/- 17%, while in the controls the index was 86 +/- 12% (p less than 0.001). The presence of serum antibodies against myoglobin was also investigated by passive haemagglutination (PH) and counterimmunoelectrophoresis (CIE). By PH the reciprocal titre of antimyoglobin antibodies was 150 +/- 28 in PM-DM compared with 16.2 +/- 15.9 and 8.7 +/- 7.7 in control patie

Improved leucocyte migration inhibition response of leucocytes from lepromatous leprosy patients with hapten modified M. leprae.

Two acetoacetylated derivatives of Mycobacterium leprae with variable hapten groups and a conjugate with tetanus toxoid were prepared. These were tested as antigens along with unmodified M. leprae in the leucocyte migration inhibition response of leucocytes from clinically, bacteriologically and histopathologically confirmed cases of lepromatous leprosy. LMI response was poor with M. leprae, but was significantly enhanced with acetoacetylated M. leprae.

Leucocyte migration agarose test to study cell-mediated immunity in amoebiasis.

The leucocyte migration agarose test (LMAT) was used to measure quantitatively the magnitude of cell-mediated immunity (CMI) in 35 patients with amoebic liver abscess and 22 healthy controls. LMAT was positive in 65.7% and 85.7% of patients with amoebiasis in the presence of 200 microgram and 400 microgram of the amoeba extract respectively, whereas the test in all 22 healthy controls was negative. Time course studies showed that within 10 days after the onset of clinical illness, only 1 of 4 patients was positive. Thereafter the percentage positivity was raised, especially when 400 microgram

Optic neuritis in the Landry-Guillain-Barré-Strohl syndrome.

A patient with typical Landry-Guillain-Barré-Strohl syndrome (LG-BS) developed bilateral optic neuritis. Laboratory studies showed hypersensitivity to both central and peripheral nervous tissue myelin. The occurrence of optic neuritis is presumably due to autohypersensitivity to central nervous tissue myelin. The initial lesions of the LG-BS syndrome in the peripheral nerves might have liberated sequestered antigens that cross-reacted with central nervous system myelin.

The effect of salicylate on the leucocyte migration inhibition test.

The effect of sodium salicylate on peripheral blood leucocyte migration is described. At a concentration of 3,5 mmol/ml, no effect on leucocyte migration was seen, but this concentration appears to influence the leucocyte migration in the presence of specific antigen, either by increasing or by decreasing leucocyte migration inhibition.

Rebound from nitric oxide inhibition triggers enhanced monocyte activation and chemotaxis.

Exposure of human peripheral blood monocytes to the NO donor S-nitroso-N-acetyl-DL-penicillamine (SNAP) resulted in a rapid shift in cellular conformation of spontaneously activated cells from ameboid to round. The population of activated cells, approximately 7. 1 +/- 1.2%, was reduced 7-fold to 1.1 +/- 0.4% following 0.5 h exposure to SNAP. Observation of monocytes for 6 h demonstrated a gradual release from NO inhibition initiating at 2.5 h following SNAP treatment and a period of hyperactivity that was maximal at approximately 5 h following SNAP exposure. During the rebound from the NO inhi

Cytophilic antibodies cause leucocyte migration inhibition in coeliac disease.

Leucocyte migration inhibition in response to challenge with gluten fraction III was prevented by pretreatment of coeliac leucocytes with pronase, an enzyme that removes cytophilic antibodies. Leucocytes from healthy volunteers preincubated with serum from untreated and treated coeliac patients showed marked migration inhibition when incubated with gluten fraction III. No migration inhibition was observed with leucocytes from healthy volunteers preincubated with serum from patients with small bowel diseases other than coeliac disease and from other healthy volunteers. Untreated and treated coe

Retrobulbar neuritis. In vitro evidence of sensitization to myelin basic protein in patients without multiple sclerosis.

Thirty-six normal subjects and 34 patients with retrobulbar neuritis were studied with use of the technique of macrophage migration inhibition factor assay and myelin basic protein as antigen. Serial studies were carried out when possible. Normal subjects gave a mean migration index of 100.9+/-9. Eleven patients with retrobulbar neuritis alone gave a mean migration index of 55+/-16 in the first 3 weeks of illness, 89+/-17.3 during the fourth to the twenty-fourth weeks, and 100.9+/-9.0 after the twenty-fourth week. Ten multiple sclerosis patients with retrobulbar neuritis gave values of 61+/-21

Clearance of activated stellate cells for hepatic fibrosis regression: molecular basis and translational potential.

Hepatic fibrosis, characterized by abnormal accumulation of extracellular matrix (ECM), is a common pathological process of many chronic liver diseases. A growing number of studies have shown that the activation of hepatic stellate cells (HSCs) plays an important role in the pathogenesis of hepatic fibrosis. Inhibiting the activation of HSCs and accelerating the clearance of activated HSCs may be effective strategies for resolution of hepatic fibrosis. Therefore, understanding the underlying mechanisms of clearance of activated HSCs and the therapeutic implications is an active subject of rese

Intracellular localization of the migration inhibitory factor (MIF) in a long-term human lymphoid cell line.

Subcellular fractions of the human lymphoid cell line PGLC-33H were obtained by N2 cavitation and differential centrifugation. The purity of the fractions was assessed by the use of the following marker enzymes: beta-glucuronidase for the lysosomal-intermediate fraction; a nonspecific esterase for the microsomal fraction; and LDH for the supernatant fraction. These subcellular fractions were studied for MIF activity utilizing human lymphoid cells from established lines as target cells. MIF activity was most consistently found in the microsomal fraction. Also, MIF activity was closely associate

Effect of levamisole on experimental paracoccidioidomycosis in the Syrian hamster: immunologic and histopathologic correlation.

The effect of levamisole (LMS) was studied in hamsters inoculated with live yeast phase culture of Paracoccidioides brasiliensis by intratesticular route. One group started LMS therapy at an early stage of infection (LMS3 group), when the animals were immunocompetent, and another group was treated in a later stage, when the immune response was already depressed (LMS12 group). As control, one group was not treated. The alterations induced by levamisole were studied by immunologic and histopathologic parameters. Compared to controls, the LMS3 group presented normal levels of cellular immune resp

Enhanced elimination of Listeria monocytogenes at the site of delayed footpad reaction.

The protective activity against a challenge infection with Listeria monocytogenes was investigated at the site of a delayed footpad reaction in mice immunized with viable or killed listeria. Delayed footpad reactivity was induced only in mice immunized with viable bacteria. Rapid and marked elimination of challenge bacteria was observed only at the site of reaction in mice immunized with viable bacteria but not in mice immunized with killed bacteria. Macrophage migration inhibitory activity was observed equally in both groups of mice. These results suggest that the delayed footpad reaction con
