Stability and potential degradation of the α',β'-epoxyketone pharmacophore on ZnO nanocarriers: insights from reactive molecular dynamics and density functional theory calculations.

We investigate the structure and dynamics of a zinc oxide nanocarrier loaded with Carfilzomib, an epoxyketone proteasome inhibitor developed for treating multiple myeloma. We demonstrate that, even though both bare and functionalized zinc oxide supports have been used for drug delivery, their interactions with the reactive functional groups of the ligands could be detrimental. This is because pharmacophores like α',β'-epoxyketones should preserve the groups required for the drug activity and be capable of leaving the vehicle at the target site. Earlier studies showed that even when ZnO is func

Formation and evolution of C-C, C-O, C[double bond, length as m-dash]O and C-N bonds in chemical reactions of prebiotic interest.

A series of prebiotic chemical reactions yielding the precursor building blocks of amino acids, proteins and carbohydrates, starting solely from HCN and water is studied here. We closely follow the formation and evolution of the pivotal C-C, C-O, C[double bond, length as m-dash]O, and C-N bonds, which dictate the chemistry of the molecules of life. In many cases, formation of these bonds is set in motion by proton transfers in which individual water molecules act as catalysts so that water atoms end up in the products. Our results indicate that the prebiotic formation of carbon dioxide, formal

Novel triazole-sulfonamide bearing pyrimidine moieties with carbonic anhydrase inhibitory action: Design, synthesis, computational and enzyme inhibition studies.

A series of new triazole-sulfonamide bearing pyrimidine derivatives were designed and synthesized via click chemistry. All new compounds (SH-1 to SH-28) were validated by 1HNMR, 13CNMR, HRMS, and SH-3 was further structurally validated by X-Ray single diffraction study. These compounds (SH-1 to SH-28) were tested as inhibitors of human carbonic anhydrase (hCA) isoforms, such as hCA I, II, IX and XII, using a stopped flow CO2 hydrase assay. Most of the compounds exhibited significant inhibitory activity against hCA II and weak inhibitory activity against hCA I. The target compounds also display

Molecular diagnostics of injury and repair responses in critical illness: what is the future of "monitoring" in the intensive care unit?

OBJECTIVE: To identify potential future means of monitoring injury and repair in critical illness. DESIGN: Review of the literature. RESULTS: Critically ill patients are monitored extensively and intensively by such means as hemodynamics, laboratory values, and radiologic studies. In general, however, the goal of monitoring has been to measure the degree of injury and to prevent further injury, rather than to measure repair. Measures of repair have been limited to phenotypic end points such as return of organ function, as measured by blood chemistry. In this article, I examine how it may be

Molecular Toxicological Mechanisms of Pharmaceutical Nitrosamines: DNA Damage, Metabolic Activation, Carcinogenesis, and Advanced Analytical Detection Strategies.

In light of the valsartan recalls in 2018, nitrosamine impurities have become some of the most important pharmaceutical quality concerns to be seen in recent years, as NDMA and other nitrosamines have been discovered in angiotensin receptor blockers, metformin, ranitidine, and many other medicines. Nitrosamine impurities fall under the IARC classification of probable human carcinogens (Group 2A), and although present in very small amounts, these have prompted stricter standards for their presence, as set by the FDA, EMA, MHRA, PMDA, and Health Canada. The present review provides an overview of

Supramolecular recognition on the surface of two-dimensional polyketones.

Two-dimensional polymers constitute an emerging class of materials with structurally defined, atomically thin architectures that impart unique optical, electronic, and host-guest properties. Here, we report the synthesis of a crystalline two-dimensional polyketone (2D-PK) via a Friedel-Crafts acylation reaction at a solid-liquid interface on aluminum foil. This interfacial approach yields micrometer-sized sheets composed of highly ordered, stacked layers, as confirmed by SEM, TEM, AFM, and HRTEM analyses. Spectroscopic and diffraction studies-including characteristic shifts in the carbonyl str

Machine-learned interatomic potentials for battery materials: from fundamental methodology to emerging applications in electrodes, electrolytes, and interfaces.

The rapid expansion of battery technologies beyond conventional lithium-ion systems has created an urgent demand for predictive, atomistic-scale simulation tools capable of addressing increasingly complex materials and interfaces. While first-principles calculations have played a central role in elucidating fundamental properties such as redox potentials, phase stability, and ion diffusion, its computational cost severely limits accessible system sizes and timescales. As a consequence, large-scale phenomena including defect-mediated transport, interfacial reactions, and mesoscale structural ev

Targeting virus-glycan recognition in influenza viruses and coronaviruses: from the molecular principles to glycomimetic antivirals strategies.

Glycans constitute the first molecular landscape encountered by viruses during infection and play fundamental roles in viral attachment, host adaptation, tissue tropism, and immune evasion. Far from being passive components of the cell surface, host glycans actively regulate virus-host interactions by functioning as attachment factors, entry receptors, or modulators of receptor engagement, while viral glycans contribute to immune escape and infectivity. Consequently, the molecular recognition of glycans has emerged as a major determinant of viral evolution and pathogenicity. This review presen

Calculation of ion-ion mutual neutralization rate constants using Landau-Zener theory coupled with trajectory simulations for Ar+-Cl-, Br-, I.

In this computational study, we self-consistently calculate the rate constants of mutual neutralization reactions by incorporating the electron transfer probability, using Landau-Zener state transition theory with inputs derived from ab initio quantum chemistry calculations, into classical trajectory simulations. Electronic structure calculations are done using correlation consistent basis sets with multi-reference configuration interaction to map all the molecular electronic states below the ion-dissociation limit as a function of the distance between the reacting species. Our electronic stru

Design and synthesis of natural antibacterial derivatives for ocular tuberculosis: a comprehensive review.

Ocular tuberculosis (TB) is an underrecognized extrapulmonary manifestation of Mycobacterium tuberculosis (M. tuberculosis) infection that can result in irreversible vision loss. Current systemic therapies, including isoniazid, rifampicin, pyrazinamide, and ethambutol, are often inadequate in achieving therapeutic intraocular concentrations and may pose ocular toxicity risks. The eye's unique anatomical and physiological barriers, including the cornea, blood-aqueous, and blood-retinal barriers, limit drug penetration, particularly to the posterior segment. This paper explores the potential of

Fundamental Invariant Neural Network (FI-NN) Potential Energy Surface for the OH + CH3OH Reaction with Analytical Forces.

The hydrogen abstraction reaction of OH + CH3OH plays a great role in combustion and atmospheric and interstellar chemistry and has been extensively studied theoretically and experimentally. Theoretically, the numerical gradients with respect to the Cartesian coordinates of atoms in molecular simulations on our recent potential energy surface (PES) for the title reaction trained using the permutationally invariant polynomial neural network (PIP-NN) approach hinder the extensive calculation because of the unaffordable computation cost. To address this issue, we in this work report a new full-di

Mechanism of Nitrogen-Carbon Bond Formation from Iron(IV) Disilylhydrazido Intermediates during N2 Reduction.

We recently reported a reaction sequence that activates C-H bonds in simple arenes as well as the N-N triple bond in N2, delivering the aryl group to N2 to form a new N-C bond (Nature 2020, 584, 221). This enables the transformation of abundant feedstocks (arenes and N2) into N-containing organic compounds. The key N-C bond forming step occurs upon partial silylation of N2. However, the pathway through which reduction, silylation, and migration occurred was unknown. Here, we describe synthetic, structural, magnetic, spectroscopic, kinetic, and computational studies that elucidate the steps of

Enantio- and Diastereoenriched Enzymatic Synthesis of 1,2,3-Polysubstituted Cyclopropanes from (Z/E)-Trisubstituted Enol Acetates.

In nature and synthetic chemistry, stereoselective [2+1] cyclopropanation is the most prevalent strategy for the synthesis of chiral cyclopropanes, a class of key pharmacophores in pharmaceuticals and bioactive natural products. One of the most extensively studied reactions in the organic chemist's arsenal, stereoselective [2+1] cyclopropanation, largely relies on the use of stereodefined olefins, which require elaborate laboratory synthesis or tedious separation to ensure high stereoselectivity. Here we report engineered hemoproteins derived from a bacterial cytochrome P450 that catalyze the

Turbulent burning velocity and thermodiffusive instability of premixed flames.

Reported in the paper are results of unsteady three-dimensional direct numerical simulations of laminar and turbulent, lean hydrogen-air, complex-chemistry flames propagating in forced turbulence in a box. To explore the eventual influence of thermodiffusive instability of laminar flames on turbulent burning velocity, (i) a critical length scale Λ_{n} that bounds regimes of unstable and stable laminar combustion is numerically determined by gradually decreasing the width Λ of computational domain until a stable laminar flame is obtained, and (ii) simulations of turbulent flames are performed b

Cooperative Activation of CO2 and Epoxide by a Heterobinuclear Al-Fe Complex via Radical Pair Mechanisms.

Activation of inert molecules like CO2 is often mediated by cooperative chemistry between two reactive sites within a catalytic assembly, the most common form of which is Lewis acid/base bifunctionality observed in both natural metalloenzymes and synthetic systems. Here, we disclose a heterobinuclear complex with an Al-Fe bond that instead activates CO2 and other substrates through cooperative behavior of two radical intermediates. The complex Ldipp(Me)AlFp (2, Ldipp = HC{(CMe)(2,6-iPr2C6H3N)}2, Fp = FeCp(CO)2, Cp = η5-C5H5) was found to insert CO2 and cyclohexene oxide, producing LdippAl(Me)(

Biosynthesis of Chiral Amino Alcohols via an Engineered Amine Dehydrogenase in E. coli.

Chiral amino alcohols are prevalent synthons in pharmaceuticals and synthetic bioactive compounds. The efficient synthesis of chiral amino alcohols using ammonia as the sole amino donor under mild conditions is highly desired and challenging in organic chemistry and biotechnology. Our previous work explored a panel of engineered amine dehydrogenases (AmDHs) derived from amino acid dehydrogenase (AADH), enabling the one-step synthesis of chiral amino alcohols via the asymmetric reductive amination of α-hydroxy ketones. Although the AmDH-directed asymmetric reduction is in a high stereoselective

Computational planning of the synthesis of complex natural products.

Training algorithms to computationally plan multistep organic syntheses has been a challenge for more than 50 years1-7. However, the field has progressed greatly since the development of early programs such as LHASA1,7, for which reaction choices at each step were made by human operators. Multiple software platforms6,8-14 are now capable of completely autonomous planning. But these programs 'think' only one step at a time and have so far been limited to relatively simple targets, the syntheses of which could arguably be designed by human chemists within minutes, without the help of a computer.
