The perspective of cAMP/cGMP signaling and cyclic nucleotide phosphodiesterases in aortic aneurysm and dissection.

Aortic aneurysm (AA) and dissection (AD) are aortic diseases caused primarily by medial layer degeneration and perivascular inflammation. They are lethal when the rupture happens. Vascular smooth muscle cells (SMCs) play critical roles in the pathogenesis of medial degeneration, characterized by SMC loss and elastin fiber degradation. Many molecular pathways, including cyclic nucleotide signaling, have been reported in regulating vascular SMC functions, matrix remodeling, and vascular structure integrity. Intracellular cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (c

Role of phosphodiesterases in the pathophysiology of neurodevelopmental disorders.

Phosphodiesterases (PDEs) are enzymes involved in the homeostasis of both cAMP and cGMP. They are members of a family of proteins that includes 11 subfamilies with different substrate specificities. Their main function is to catalyze the hydrolysis of cAMP, cGMP, or both. cAMP and cGMP are two key second messengers that modulate a wide array of intracellular processes and neurobehavioral functions, including memory and cognition. Even if these enzymes are present in all tissues, we focused on those PDEs that are expressed in the brain. We took into consideration genetic variants in patients af

ADH resistance of LLC-pk1 cells caused by overexpression of cAMP-phosphodiesterase type-IV.

The studies of animal models of nephrogenic diabetes insipidus (NDI) suggest that abnormally high activity of cAMP phosphodiesterase (cAMP-PDE), may cause unresponsiveness to the diuretic effect of AVP. We explored whether overexpression of one of the cAMP-PDE type isozymes, PDE-IV, in [8-Arg]-vasopressin (AVP) sensitive renal epithelial LLC-PK1 cells can prevent the hormone-elicited cAMP increase. LLC-PK1 cells were stably transfected with ratPDE3.1 cDNA (which encodes for rolipram-sensitive PDE-IV), inserted in plasmid pCMV5 and then were compared with sham-transfected LLC-PK1 cells and wild

Phosphodiesterase 1A overexpression contributes to the progression of renal fibrosis.

Renal fibrosis is a common histological feature of chronic kidney disease. The role of phosphodiesterase 1A (PDE1A), a crucial molecule regulating cyclic guanosine monophosphate (cGMP) signaling in tubular epithelial cells, in renal fibrosis is unknown. We found that the mRNA of PDE1A was elevated in the urinary exosomes from patients with rapidly progressing IgA nephropathy. The PDE1A protein in renal tubular epithelial cells increased as renal function decline in patients with IgA nephropathy or diabetic kidney disease. In mouse models of unilateral ureter obstruction (UUO) and chronic folic

cGAS deficiency mitigated PM2.5-induced lung injury by inhibiting ferroptosis.

Ferroptosis emerges as one of the pivotal types of cell death during fine particulate matter (PM2.5)-induced lung injury. The recently discovered cytosolic DNA sensor, cyclic GMP-AMP synthase (cGAS), triggers the activation of the downstream adaptor protein STING by synthesizing cyclic GMP-AMP, playing vital roles in innate immunity and cell death. Nonetheless, the specific function of cGAS in lung injury caused by PM2.5 remains to be elucidated. The present study aimed to explore the involvement of cGAS in the pathogenesis of PM2.5-induced lung injury and its potential mechanisms. The express…

Hypoxia inducible factor-1 improves the negative functional effects of natriuretic peptide and nitric oxide signaling in hypertrophic cardiac myocytes.

AIMS: Both natriuretic peptides and nitric oxide may be protective in cardiac hypertrophy, although their functional effects are diminished in hypertrophy. Hypoxia inducible factor-1 (HIF-1) may also protect in cardiac hypertrophy. We hypothesized that upregulation of HIF-1 would protect the functional effects of cyclic GMP (cGMP) signaling in hypertrophied ventricular myocytes. MAIN METHODS: A cardiac hypertrophy model was created in mice by transverse aorta constriction. HIF-1 was increased by deferoxamine (150 mg/kg for 2 days). HIF-1alpha protein levels were examined. Functional parameter…

Signalling pathways in the nitric oxide donor-induced dopamine release in the striatum of freely moving rats: evidence that exogenous nitric oxide promotes Ca2+ entry through store-operated channels.

We showed previously, using in vitro microdialysis, that the activation of the soluble guanylate cyclase (sGC)/cyclic GMP pathway was the underlying mechanism of the extracellular Ca(2+)-dependent effects of exogenous NO on dopamine (DA) secretion from PC12 cells. In this study, the co-infusion of the sGC inhibitor 1H-[1,2,4]oxadiazolo[4,3] quinoxalin-1-one (ODQ) failed to affect the NO donor 3-morpholinosydnonimine (SIN-1, 5.0 mM)-induced DA increase (sevenfold baseline) in dialysates from the striatum of freely moving rats. Ca(2+) omission from the perfusion fluid abolished baseline DA relea…

Dual effects of NMDA-induced intracellular Ca2+ elevations on cGMP levels in cultured cerebellar granule neurons.

1. Cyclic GMP (cGMP) levels were markedly elevated by N-methyl-D-aspartate (NMDA) within 1-3 min of incubation, then gradually decreased with incubation time. 2. The NMDA-induced intracellular Ca2+ elevations showed maximal levels just after adding NMDA and were maintained for 60 min. 3. NMDA did not show augmentation of cGMP elevation with sodium nitroprusside (SNP), rather it decreased the SNP-induced cGMP elevation after exposure for 60 min. 4. The NMDA-induced elevation of cGMP was remarkably augmented with the phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX, 1mM), after 60 …

Suramin and its analogues are multi-functional inhibitors of cGAS.

Upon sensing double-stranded DNA, cyclic GMP-AMP synthase (cGAS) catalyzes ATP and GTP to synthesize the cyclic dinucleotide 2',3'-cGAMP, which subsequently activates stimulator of interferon genes (STING)-mediated immune responses. Modulation of cGAS catalytic activity is crucial for treating diseases associated with the cGAS-STING pathway. Here, we found that suramin and its analogues, such as NF023, can inhibit 2',3'-cGAMP synthesis by competitively binding to cGAS. Structural analysis of the cGAS-NF023 complex reveals that NF023 binds to the active-site cleft and DNA-binding sites of cGAS,…

The STING in Non-Alcoholic Fatty Liver Diseases: Potential Therapeutic Targets in Inflammation-Carcinogenesis Pathway.

Non-alcoholic fatty liver disease (NAFLD), an important chronic disease, is one of the major causes of high mortality and creates a substantial financial burden worldwide. The various immune cells in the liver, including macrophages, NK cells, dendritic cells, and the neutrophils involved in the innate immune response, trigger inflammation after recognizing the damage signaled from infection or injured cells and tissues. The stimulator of interferon genes (STING) is a critical molecule that binds to the cyclic dinucleotides (CDNs) generated by the cyclic GMP-AMP synthase (cGAS) to initiate the…

A non-canonical, interferon-independent signaling activity of cGAMP triggers DNA damage response signaling.

Cyclic guanosine monophosphate-adenosine monophosphate (cGAMP), produced by cyclic GMP-AMP synthase (cGAS), stimulates the production of type I interferons (IFN). Here we show that cGAMP activates DNA damage response (DDR) signaling independently of its canonical IFN pathways. Loss of cGAS dampens DDR signaling induced by genotoxic insults. Mechanistically, cGAS activates DDR in a STING-TBK1-dependent manner, wherein TBK1 stimulates the autophosphorylation of the DDR kinase ATM, with the consequent activation of the CHK2-p53-p21 signal transduction pathway and the induction of G1 cell cycle ar…
