Normal pregnancy and successful delivery in myophosphorylase deficiency (McArdle's disease).

The progress in pregnancy of a female with myophosphorylase deficiency (McArdle's disease) is described. In spite of the increased muscular effort expended, both pregnancy and labour were normal and the muscle symptoms unchanged, suggesting that compensatory mechanisms might have operated. These possible mechanisms are discussed. Women suffering from the myopathy need not expect any deterioration during pregnancy.

Longitudinal Evidence of Sustained Taurine Deficiency in Inflammatory Bowel Disease.

Inflammatory Bowel Diseases (IBD), most notably ulcerative colitis (UC) and Crohn's disease (CD), are long-standing disorders driven by dysregulated immune responses within the gastrointestinal tract and characterized by several metabolic disturbances, which are believed to influence disease progression. We have recently shown that a systemic deficiency of taurine, a semi-essential amino acid with anti-inflammatory properties, marks IBD. To characterize the temporal dynamics and determinants of taurine deficiency in IBD, we conducted a prospective longitudinal study assessing serum taurine lev

A rare cause of kidney disease in two siblings: transaldolase deficiency.

Transaldolase is a key enzyme in the pentose phosphate pathway, an alternative pathway for glucose metabolism. Transaldolase deficiency leads to the accumulation of metabolites such as polyols and seven-carbon sugars, particularly in urine. Transaldolase deficiency is a rare disorder with multisystem involvement, including liver dysfunction, hepatosplenomegaly, anemia, thrombocytopenia, endocrine abnormalities and dysmorphic features. We now report on two siblings with kidney involvement due to transaldolase deficiency. Kidney dysfunction is common in transaldolase deficiency and may progress

Selective immunoglobulin A deficiency and coeliac disease: Immune response dynamics.

OBJECTIVE: To analyse the serological immune response in paediatric patients with selective immunoglobulin A deficiency (SIgAD) and coeliac disease (CeD), focusing on immunoglobulin G antibody dynamics after starting the gluten-free diet (GFD). METHODS: Data from SIgAD patients with a CeD diagnosis, age less than 18 years, was retrospectively collected by the participating centres in this observational, multicentre, pan-european registry, conducted by the European Society for Paediatric Gastroenterology, Hepatology and Nutrition (ESPGHAN) Coeliac Disease Special Interest group. The analysis o

Iron Deficiency and Anemia in Chronic Kidney Disease: Assessment and Management.

Patients with all stages of chronic kidney disease are at risk for developing anemia and iron deficiency with and without anemia. Therapies to manage these conditions and evidence-based guidelines are available to assist in evaluating and directing management. Patient history, laboratory evaluation, current therapy, and patient preference all assist in making an accurate plan of care.

Copper dysregulation in cardiometabolic disease: copper deficiency versus cuproptosis.

Copper is an essential micronutrient required for mitochondrial respiration, antioxidant defense, and metabolic homeostasis. Accumulating evidence demonstrates that dysregulated copper handling, including deficiency, redistribution, or overload, is a reproducible feature of multiple cardiometabolic disorders, including heart failure, diabetes mellitus, obesity, and NAFLD/MASLD. Human, animal, and cellular studies consistently implicate altered copper trafficking and compartmentalization in mitochondrial dysfunction, oxidative stress, and tissue remodeling across these conditions. The recent id

Sex-dependent impact of Il6 deficiency in Parkinson's disease mice.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder accompanied by neuroinflammation. Many studies have demonstrated that interleukin-6 (IL-6) exhibits both anti-inflammatory and pro-inflammatory effects in the central nervous system, yet its role in PD remains controversial. In this study, Il6 -/- (knockout), Il6 +/-, and wild-type mice were utilized to investigate the impact of IL-6 on the pathology of MPTP- or α-SynucleinA53T-induced PD mice. Our findings revealed that Il6 deficiency exacerbated motor dysfunction in both female and male mice. MPTP intoxication resulted in ear

Assessment of Bicarbonate Deficiency in Feline Acute and Chronic Kidney Disease.

Bicarbonate deficiency is a common complication of acute kidney injury (AKI) and chronic kidney disease (CKD), resulting from impaired renal bicarbonate handling. It promotes bone demineralization and calcium-phosphate (CaxP) imbalance. In dogs, elevated CaxP has been linked to increased frequency and severity of bicarbonate deficiency. This 10-year study evaluated the prevalence and degree of bicarbonate deficiency in 618 cats diagnosed with AKI, acute-on-chronic kidney disease (ACKD), and CKD, and its association with azotemia and mineral imbalance. A retrospective analysis was conducted on

PATJ deficiency leads to cystic kidney disease and related ciliopathies.

Cystic kidney disease and related ciliopathies are caused by pathogenic variants in genes that commonly result in ciliary dysfunction. For a substantial number of individuals affected by those cilia-related diseases, the causative gene remains unknown. Using massively parallel sequencing, we here identified a pathogenic bi-allelic variant in the gene encoding PALS1-associated tight junction protein ([PATJ] also known as inactivation-no-afterpotential D-like, INADL) in an individual with ciliopathy. The affected fetus carried the homozygous truncating PATJ nonsense variant c.830delC (p.Pro277fs
