Gasdermin D-mediated keratinocyte pyroptosis as a key step in psoriasis pathogenesis.

Gasdermin D (GSDMD)-mediated pyroptosis has a significant pro-inflammation characteristic due to dramatic secretion of pro-inflammatory subs … he background of Gsdmd-/- mice (not limited in transplanting area). In M5-stimulated keratinocytes, disulfiram or GSDMD siRNA transfection can inhibit pyroptosis and eliminate disproportionate increases of Ki-67 and PI. We further validated that topically application of disulfiram (pyroptosis inhibitor) also alleviated IIPLD in mice. These findings indicate …

Pharmacologic interventions for alcohol use disorder.

Alcohol use disorder (AUD) has devastating effects, and its prevalence in the US is growing. It is therefore important to examine multiple avenues for successful cessation and continued abstinence. Disulfiram, oral naltrexone, naltrexone IM, and acamprosate are currently the only four medications approved for the treatment of AUD by the FDA. This review aims to examine the efficacy of the approved medications as well as explore the use of off-label pharmacologic treatments used for AUD treatment (including gabapentin, baclofen, oxytocin, N-acetylcysteine, calcium carbonate, and psilocybin). Th…

The Role of Repetitive Transcranial Magnetic Stimulation in Treating Alcohol Use Disorder: Neural Mechanisms, Efficacy, and Future Directions.

Alcohol Use Disorder (AUD) is a global health challenge, affecting 10-15% of the population, with significant social, health, and economic consequences. Although pharmacotherapies such as disulfiram, naltrexone, and acamprosate are available, their effectiveness is limited and patient adherence is often poor. Repetitive transcranial magnetic stimulation (rTMS), a non-invasive neuromodulation technique that targets neural circuits implicated in addiction. Emerging evidence suggests that rTMS may reduce alcohol craving and consumption, although results have been mixed. This review examines the n…

Characterizing the influence of structure and route of exposure on the disposition of dithiocarbamates using toluene-3,4-dithiol analysis of blood and urinary carbon disulfide metabolites.

Differences in the toxicities observed for dithiocarbamates have been proposed to result from the influence of nitrogen substitution, oxidation state, and route of exposure. To better characterize the fate of dithiocarbmates in vivoas a function of structure and route of exposure, rats were administered equimolar doses of carbon disulfide (CS2), N-methyldithiocarbamate, pyrrolidine dithiocarbamate, N,N-diethyldithiocarbamate, or disulfiram daily for five days, either po or ip, and sequential blood samples obtained. Protein dithiocarbamates formed by the in vivo release of CS2, parent dithiocar…

The measurement of 2-thiothiazolidine-4-carboxylic acid as an index of the in vivo release of CS2 by dithiocarbamates.

Investigation of the DSF/Cu Reaction Using HR LCMS and Reactivity Studies.

Cuprotosis is a unique mode of cell death being explored in many new therapeutic anticancer studies. A widely used approach is the combination of Cu(II) salts with diethyldithiocarbamate disulfide, disulfiram (DSF), also known as the alcohol-sensitizing drug Antabuse, which generates the toxic Cu(II)bis(diethyldithiocarbamate), Cu(DTC)2, in a complex redox reaction. The mechanism for the generation of Cu(DTC)2 in this reaction, which we term the DSF/Cu reaction, has been examined using high-resolution liquid chromatography-mass spectroscopy (HR LCMS) by monitoring the reaction at different sam…

Excessive alcohol use is a leading cause of preventable death in the United States, with alcohol-related deaths increasing during the pandem … ntly abstinent; moderate evidence supports the use of topiramate in decreasing heavy-drinking days. Disulfiram has been commonly prescribed, but little evidence supports its effectiveness outside of supervised settings. Identification of novel dithiocarbamate-copper complexes targeting p97/NPL4 pathway in cancer cells.

Dithiocarbamates (DTCs) are simple organic compounds with many applications in industry and medicine. They are potent metal chelators forming complexes with various metal ions, including copper. Recently, bis(diethyldithiocarbamate)-copper complex (CuET) has been identified as a metabolic product of the anti-alcoholic drug Antabuse (disulfiram, DSF), standing behind DSF's reported anticancer activity. Mechanistically, CuET in cells causes aggregation of NPL4 protein, an essential cofactor of the p97 segregase, an integral part of the ubiquitin-proteasome system. The malfunction of p97/NPL4 cau…

Medication Development for Alcohol Use Disorder: A Focus on Clinical Studies.

Compared to other medical disorders, including other brain diseases, the number of medications approved for alcohol use disorder (AUD) is very small. Disulfiram, naltrexone (oral and long-acting), and acamprosate are approved by the US Food and Drug Administration (FDA) to treat patients with AUD. These medications are also approved in other countries, including in Europe, where the European Medicines Agency (EMA) also approved nalmefene for AUD. Furthermore, baclofen was recently approved for AUD in France. These approved medications have small effect sizes, which are probably the consequence…

Antabus in the treatment of alcoholism in a private general hospital.

In 35 consecutive cases in which patients were admitted to a private hospital for alcoholism, antabus was contraindicated for eight patients and administered to 27. Sixteen patients, followed three to nine months, cooperated fully with treatment and did not return to use of alcohol. In four cases there was at least one episode of drinking, but the patients continued to cooperate with treatment and attained partial success. In seven cases, including three in which antabus was stopped because of organic disease, treatment was unsuccessful. Insufficient insight and lack of family cooperation acco

A nonhypoglycemic chlorpropamide analog that inhibits aldehyde dehydrogenase.

Chlorpropamide (CP), a sulfonylurea-type oral hypoglycemic agent, is known to provoke a flushing reaction reminiscent of the disulfiram-ethanol reaction in certain individuals. This is manifested in rodents by an increase in blood acetaldehyde levels after ethanol administration. When the sulfonamide N1-nitrogen of CP was substituted with an ethyl group, the product, N1-ethylchlorpropamide, was found to be three times as active as CP in raising ethanol-derived blood acetaldehyde. However, whereas CP lowered fasting blood glucose in rats measured over 6 h, N1-ethylchlorpropamide was devoid of h…

Protection by diethyldithiocarbamate, a CS2-liberating agent, against different models of experimentally-induced liver injury.

In experimental animals, diethyldithiocarbamate as well as carbon disulfide exert antihepatotoxic effects in different models of chemically-induced liver injury like carbon tetrachloride, chloroform, bromobenzene, dimethyl nitrosamine, furosemide, D-galactosamine, paracetamol, thioacetamide and trichloroethylene. On the other hand, both dithiocarb and CS2 are strong inhibitors of the microsomal mixed-function oxidase system which is involved in the bioactivation of these hepatotoxic agents. It is concluded that CS2 and CS2-producing agents as dithiocarb and disulfiram may inhibit metabolism of…

Pharmacotherapy of alcoholism: gaps in knowledge and opportunities for research.

During the past decade, renewed interest in medications to prevent relapse in alcoholics has yielded a number of promising candidates. Although two of these medications, naltrexone and acamprosate, are currently in clinical use in a number of countries, overall, their effectiveness appears to be limited. Disulfiram, the deterrent medication that was approved 50 years ago for the treatment of alcoholism, has not consistently been shown to be efficacious. However, since inadequate dosing and other modifiable factors may limit its deterrent effects, the identification of a more potent metabolite …
