Assessing DNA damage in sentinel crabs as indicators of tourist impact in the Fernando de Noronha Archipelago, a Marine Protected Area and World Natural Heritage Site in Brazil.

This study investigates the genotoxic effects of tourism on two sentinel crab species (Johngarthia lagostoma and Grapsus grapsus) in the Fernando de Noronha Archipelago, Brazil, a Marine Protected Area and World Natural Heritage Site-sampling (October 2020-June 2022) covered periods of varying tourist influx due to COVID-19 restrictions. Genomic damage was assessed using micronucleus (macrolesions) and comet assays (microlesions) in hemolymph samples from crabs at two sites: Dolphin Trail (low visitation) and Porto de Santo Antônio Beach (high visitation). DNA damage increased with tourist num

Interspecies differences in mammalian susceptibility to legacy POPs and trace metals using skin fibroblast cells.

The susceptibility to trace metals and legacy POPs is different between terrestrial and marine mammals. In this study, we established the first cell line from Indo-Pacific finless porpoises and compared the cellular responses of skin fibroblast cells from Pygmy killer whales, Pantropic spotted dolphins, Indo-Pacific finless porpoises, mice, and humans following exposure to copper, methylmercury, cadmium, PCB126, PCB153, and BDE47 to better understand the interspecies sensitivities of mammals to chemical pollutants. We conducted a risk assessment by comparing no-observed effect concentrations (…

"Black Bone" MRI: a potential non-ionizing method for three-dimensional cephalometric analysis--a preliminary feasibility study.

OBJECTIVES: CT offers a three-dimensional solution to the inaccuracies associated with lateral cephalogram-based cephalometric analysis. However, it is associated with significant concerns regarding ionizing radiation exposure. MRI offers a non-ionizing alternative, but this has been less well investigated. We present a novel gradient echo MRI sequence ("Black Bone") and highlight the potential of this sequence in cephalometric analysis. METHODS: After regional ethics approval, "Black Bone" imaging was obtained in eight patients in whom lateral cephalograms were available. "Black Bone", T1 an

Lung size and thoracic morphology in shallow- and deep-diving cetaceans.

Shallow-diving, coastal bottlenose dolphins (Tursiops truncatus) and deep-diving, pelagic pygmy and dwarf sperm whales (Kogia breviceps and K. sima) will experience vastly different ambient pressures at depth, which will influence the volume of air within their lungs and potentially the degree of thoracic collapse they experience. This study tested the hypotheses that lung size will be reduced and/or thoracic mobility will be enhanced in deeper divers. Lung mass (T. truncatus, n = 106; kogiids, n = 18) and lung volume (T. truncatus, n = 5; kogiids, n = 4), relative to total body mass, were com…

Linear accuracy and reliability of cone beam CT derived 3-dimensional images constructed using an orthodontic volumetric rendering program.

OBJECTIVE: To compare accuracy of linear measurements made on cone beam computed tomographic (CBCT) derived 3-dimensional (3D) surface rendered volumetric images to direct measurements made on human skulls. MATERIALS AND METHODS: Twenty orthodontic linear measurements between anatomical landmarks on 23 human skulls were measured by observers using a digital caliper. The skulls were imaged with CBCT and Dolphin 3D (version 2.3) software used to generate 3D volumetric reconstructions (3DCBCT). The linear measurements between landmarks were computed by a single observer three times and compared

Development of a standardized nomenclature for bronchoscopy of the respiratory system of harbor porpoises (Phocoena phocoena).

Respiratory disease is common in captive and wild cetaceans. Bronchoscopy may permit early diagnosis of respiratory disease in dolphins and porpoises. Refinement of cetacean bronchoscopy requires development of a nomenclature system to facilitate description of the anatomic site at which lesions occur. A standard bronchoscopic nomenclature also permits serial evaluations of lesions and enhances communication between veterinarians. In this project, we adapted the bronchoscopic nomenclature devised by Amis and McKiernan for the dog and horse to the harbor porpoise (Phocoena phocoena). Silastic a…

Isolation of St. Louis encephalitis virus from a killer whale.

We report the isolation of St. Louis Encephalitis (SLE) virus from a mature male killer whale (Orcinus orca). This represents the first isolation of SLE virus from a marine mammal. The animal presented with reduced appetite, rapidly became lethargic and subsequently died. Virus-induced CPE was observed in a dolphin cell line, SP-1K (ATCC CCL 78), inoculated with brain, kidney, and lung tissues obtained at necropsy. Electron microscopy of infected SP-1K cells revealed the presence of virions having morphology and size resembling members of the Flaviviridae. Final identification as SLE virus was

Brain Structure of South African Children Born to Mothers on Dolutegravir Versus Efavirenz-Based Antiretroviral Therapy.

BACKGROUND: The impact of in utero exposure to specific antiretroviral therapy (ART), particularly integrase strand transfer inhibitors, on early brain development, remains poorly understood. We used magnetic resonance imaging (MRI) to compare brain structure in children who are HIV-exposed and uninfected (CHEU) with prenatal dolutegravir (DTG) vs efavirenz (EFV) exposure. METHODS: DolPHIN-2 was a randomized trial of pregnant women initiating DTG- vs EFV-based ART in the third trimester. At 3-4 years of age, a subgroup of their children from the South African cohort, along with HIV-unexposed

Three-Dimensional Changes in the Upper Airway of Skeletal Class III Patients After Different Orthognathic Surgical Procedures.

PURPOSE: We hypothesized that 3-dimensional changes in the upper airway of patients with skeletal Class III malocclusion are different after mandibular setback and maxillary advancement (MSMA) compared with mandibular setback (MS) and that upper airway changes and the amount of jaw movement are correlated. PATIENTS AND METHODS: Skeletal Class III patients who underwent MS surgery or MSMA surgery were included in this retrospective cohort study. Cone beam computed tomography scans were acquired before and 3 months after surgery. Changes in the upper airway space were assessed as primary outcom

Evaluating the performance of automated detection systems for long-term monitoring of delphinids in diverse marine soundscapes.

There is an increasing reliance on passive acoustic monitoring (PAM) as a cost-effective method for monitoring cetaceans, necessitating robust and efficient automated tools for extracting species presence. This work compares two methods, one based on the 'off-line' analysis of raw PAM data, using Convolutional Neural Networks (CNNs), and the second based on in-situ detections, implemented within the C-POD. The C-POD is a rapid, low-cost choice for monitoring of odontocetes, while CNNs, requiring large efforts to train, are gaining traction within bioacoustics as they offer performance benefits

High resolution visualisation of tiemannite microparticles, essential in the detoxification process of mercury in marine mammals.

The North Sea is an ecologically rich habitat for marine wildlife which has also been impacted by industrial developments and anthropogenic emissions of contaminants such as mercury. Marine mammals are particularly susceptible to mercury exposure, due to their trophic position, long lifespan, and dependence on (increasingly contaminated) aquatic prey species. To mitigate impact, marine mammals can detoxify methylmercury by binding it to selenium-containing biomolecules, creating insoluble mercury selenide granules. Here, liver, kidney, muscle, and brain samples from an adult male bottlenose do

First Report of Paralytic Shellfish Toxins in Marine Invertebrates and Fish in Spain.

A paralytic shellfish poisoning (PSP) episode developed in summer 2018 in the Rías Baixas (Galicia, NW Spain). The outbreak was associated with an unprecedentedly intense and long-lasting harmful algal bloom (HAB) (~one month) caused by the dinoflagellate Alexandrium minutum. Paralytic shellfish toxins (PSTs) were analyzed in extracts of 45 A. minutum strains isolated from the bloom by high-performance liquid chromatography with post-column oxidation and fluorescence detection (HPLC-PCOX-FLD). PSTs were also evaluated in tissues from marine fauna (invertebrates and fish) collected during the e

[Invasive airway management update 2011].

Invasive airway management techniques are still challenging even for skilled anesthesiologists, intensivists and emergency physicians. All current percutaneous tracheostomy techniques are based on Seldinger's principle. Using the recent introduced Blue Dolphin Kit is feasible and safe, but without clear advantage compared to other kits. There is no data available to generally support performing early (<8 days) tracheostomy in intensive care patients requiring mechanical ventilation. Experimental data show promising results of supporting expiration through a small bore catheter using ejectors b

A novel cetacean adenovirus in stranded harbour porpoises from the North Sea: detection and molecular characterization.

Harbour porpoises (Phocoena phocoena) are the most prevalent cetaceans in the North Sea. The fecal viral flora of 21 harbour porpoises stranded along the Dutch coastline was analyzed by a metagenomics approach. Sequences of a novel cetacean mastadenovirus, designated harbour porpoise adenovirus 1 (HpAdV-1), were detected. The sequence of a 23-kbp genomic region, spanning the conserved late region, was determined using primer walking. Phylogenetic analysis indicated that HpAdV-1 is most closely related to bottlenose dolphin adenovirus and clusters with Cetartiodactyla adenoviruses. The prevalen
