Evaluation of the inactivation of avian influenza virus in duck meat through electron beam irradiation.

The global spread of highly pathogenic avian influenza (HPAI) among poultry, wild birds, and mammals has increased the risk of infection in felines, including domestic and stray cats. Wild felids are particularly susceptible to H5N1 HPAI virus infection because they may consume the carcasses of infected birds. Fatalities associated with the consumption of raw pet foods prepared from poultry meat have been reported in both the United States and Korea. Given that high titers of avian influenza (AI) viruses have been detected in poultry meat, effective sterilization methods are essential to ensur

IP3R3 gene expression in laying duck tissues and its polymorphisms associated with eggshell quality.

Inositol triphosphate receptor type III (IP3R3) is a key second messenger that mediates the release of intracellular Calcium ion (Ca2+). It binds to inositol triphosphate (IP3) to form a calcium channel, regulating intracellular calcium homeostasis. This study aimed to analyze IP3R3 mRNA expression in duck tissues and its single nucleotide polymorphism (SNP) association with eggshell quality. Forty-five-week-old Guizhou Sansui ducks were used as experimental subjects. The real-time fluorescence quantitative PCR (qRT-PCR) assessed gene expression, while SNPs were identified via PCR products seq…

Construction of an infectious clone of novel duck picornavirus using the infectious subgenomic amplicon method.

A picornavirus strain of Duck/FC22/China/2017 (FC22) with an unclear taxonomic status was isolated in our laboratory in 2017. To study the biological properties of the virus, an infectious clone of the FC22 strain was successfully rescued by the infectious subgenomic amplicon method. The FC22 gene was amplified in two segments and fused with CMV and HDR after the addition of marker sites. The fusion fragment was transfected into LMH cells and passaged, and the obtained virus was named rFC22 and identified by PCR, IFA and electron microscopy. The biological characteristics of strain rFC22 were

Revealing the spatial heterogeneity of the gut microbiota of red-feather ducks and their interaction with environmental microorganisms.

Ducks are one of important economic waterfowl species, and their gastrointestinal microbiota play crucial roles in nutrient metabolism, immune regulation, and host health maintenance. In this study, we collect 436 gastrointestinal content samples from red-feather ducks, including samples from the glandular stomach, muscular stomach, duodenum, ileum and cecum, as well as 10 environmental samples (water and soil). We then employed 16S rRNA gene sequencing to explore the gastrointestinal microbial communities of ducks and their interactions with environmental microbes from soil and water. The gas…

Friend Turned Foe: Pasteurella multocida Bacteremia Following a Scratch by an Adopted Pekin Duck.

Pasteurella multocida is a gram-negative coccobacillus that colonizes the aerodigestive tracts of cats, dogs, birds, and wild animals. This veterinary pathogen spreads to humans via licking, biting, or scratching. Pekin ducks are known carriers of multiple strains of Pasteurella bacteria that can manifest as fowl cholera, eye infection, or duck septicemia. This bacterium is a farmer's nightmare, as it is associated with high mortality rates. Herein, we report a unique case of P. multocida bacteremia in an immunocompetent host that shared a house with adopted Pekin ducks.

Threonine modulates the STAT3-SCD1 pathway to reduce fatty acid metabolism in duck hepatocytes.

Dietary threonine (Thr) is known to influence fat deposition in poultry, but the precise mechanisms behind its regulatory effects on hepatic lipid metabolism remain elusive. Prior research indicated that including supplemental Thr in the feed may influence STAT3 (Signal Transducer and Activator of Transcription 3) levels in the liver of meat ducks. Numerous studies have recorded the function of STAT3 in regulating fatty acid (FA) metabolism in mammals. The primary objective of this study was to investigate whether Thr influences FA metabolism and triglycerides (TG) deposition in duck liver by …

miR-317 regulates the proliferation and apoptosis of duck follicle granulosa cells by targeting VIPR1.

VIPR1 can specifically bind VIP, a PRL release factor, which promotes the secretion of PRL from the pituitary gland, and participates in the regulation of bird nesting behavior. The purpose of this study was to investigate the effects of miR-317 overexpression or silencing on VIPR1 gene and protein expression in duck follicle granulosa cells. The ovaries of Muscovy ducks were collected during the nesting and laying periods, and histological differences were analyzed via HE staining. Duck primary follicle granulosa cells were isolated and identified by immunofluorescence staining, after which t…

West Nile Virus Infection in a Pekin Duck ( Anas platyrhynchos domesticus ) Developer Flock.

Developer ducks are ducks being reared for breeding. Like breeder candidate chickens, they are raised with appropriate light and feed programs. A commercial Pekin duck (Anas platyrhynchos domesticus) developer flock experienced an extraordinary, elevated mortality event at 6 wk of age. Weekly mortality rate that week was 162 ducklings out of a flock of 6420 (2.5%). Mortality jumped to 988 (15.4%) ducklings the next week. On first elevated mortality, six dead ducks from that flock were submitted for diagnostic investigation at Michigan State University Veterinary Diagnostic Laboratory. Myocardi…

N-glycosylation of the envelope glycoprotein I is essential for the proliferation and virulence of the duck plague virus.

Duck plague virus (DPV) causes the highly pathogenic duck plague, and the envelope glycoprotein I (gI), as one of the key virulence genes, h … ental virus, the mutated virus shows a 66.3% reduction in intercellular transmission capability. In ducks, the deglycosylation of gI significantly reduces DPV replication in vivo, thereby weakening the virulence of DPV. This study represents the first successful creation of a weak DPV virus strain by specific mutation at the N-glycosylation site. The fi …

Slightly different metabolomic profiles are associated with high or low weight duck foie gras.

Understanding the evolution of fatty liver metabolism of ducks is a recurrent issue for researchers and industry. Indeed, the increase in weight during the overfeeding period leads to an important change in the liver metabolism. However, liver weight is highly variable at the end of overfeeding within a batch of animals reared, force-fed and slaughtered in the same way. For this study, we performed a proton nuclear magnetic resonance (1H-NMR) analysis on two classes of fatty liver samples, called low-weight liver (weights between 550 and 599 g) and high-weight liver (weights above 700 g). The …

Role of the homologous MTase-RdRp interface of flavivirus intramolecular NS5 on duck tembusu virus.

Flavivirus nonstructural protein 5 (NS5) harbors the N-terminal methyltransferase (MTase) and C-terminal polymerase RNA-dependent RNA polymerase (RdRp). The intramolecular NS5 features an integral MTase and RdRp interface with two components: a six-residue hydrophobic network and a GTR linker. Herein, the determinants of the MTase-RdRp interface and flavivirus substituted GTR linker were explored in TMUV replication and proliferation. First, the NanoLuc® Binary Technology (NanoBiT) and coimmunoprecipitation assays (Co-IP) methods confirmed the interaction between the MTase and RdRp domains of
