Computational design of inhibitory peptides and an mRNA-Based multi-epitope vaccine targeting the MIC3 protein of Eimeriatenella.

This study addresses coccidiosis, a prevalent disease causing significant economic losses, and focuses on the EtMIC3 protein due to its critical role in Eimeria tenella pathogenesis. Our goal is to develop innovative therapeutic and preventive strategies leveraging the potential of the EtMIC3 protein. In this investigation, we evaluate the interaction of the EtMIC3 protein with its receptors, develop inhibitory peptides, and select epitopes from EtMIC3 using immunoinformatic tools. We assess the presentation of these epitopes to immune cells, model a multi-epitope protein, predict the mRNA str

High-strength, multi-mode processable bamboo molecular bioplastic enabled by solvent-shaping regulation.

The global reliance on petrochemical plastics has led to severe environmental crises, necessitating sustainable alternatives that combine high performance with circularity. While bioplastics derived from biomass show promise, their widespread adoption is hindered by inferior mechanical properties, limited processability, and reliance on food-competing feedstocks. Here, we present a molecular engineering strategy to fabricate high-strength bamboo molecular plastics (BM-plastics) through a solvent-regulated shaping process. By employing deep eutectic solvents to disassemble bamboo cellulose's hy

Targeted Weaving of Triazine Ionic Polymers Within the Pores of Covalent Organic Frameworks for Enhanced CO2 Conversion.

The chemical conversion of carbon dioxide (CO2) is crucial for mitigating global warming and creating economic value. A particularly promising approach is the cycloaddition of CO2 with epoxides to produce cyclic carbonates. Nevertheless, designing green and highly efficient heterogeneous catalysts remains a key challenge in this process. Based on a function-oriented molecular design strategy, this study developed a series of composite materials, COF-TIPs-X (X = 1, 2, 3, or 4), by precisely controlling the grafting density of triazine ionic polymers (TIPs) onto the framework of covalent organic

Immobilization of carbonyl reductase in engineered polyhydroxyalkanoate microspheres for asymmetric synthesis of (S)-CFPL.

An in vivo self-assembly strategy was developed to overcome the limitation of traditional enzyme immobilization methods. An engineered E. coli system was employed to immobilize carbonyl reductase (LcCR) onto intracellular polyhydroxyalkanoate (PHA) microspheres, which was realized by fusion expression of LcCR with covalent PhaC or non-covalent PhaP. The fusion of LcCR and PHA surface binding proteins (Phac, PhaP) was optimized by adjusting the fusion direction (C/N end) and replacing peptide linkers (GSA, (G4S)3, (G4S)4). The expression form for fusion of N-terminal of LcCR with three series c

[Proximal humerus fracture: old and new, established and doubtful, conservative and operative aspects].

Proximal humeral fractures are not only frequent injuries but also serve as an important indicator for a possibly underlying osteoporosis. Therefore, in addition to fracture management, timely diagnostics and treatment of the osteoporosis should also be prioritized. The decision between a conservative and surgical treatment approach requires a differentiated consideration of the fracture morphology, individual patient characteristics and functional expectations. Conservative treatment can be an adequate alternative in specific cases, particularly in geriatric patients with low functional deman

Matricellular Proteins (MCPs) in Rheumatoid Arthritis.

Rheumatoid arthritis is a chronic autoimmune disorder affecting approximately 230 per 100,000 people worldwide. It typically affects joints and bones but may involve other tissues and internal organs as well. Rheumatoid arthritis is twice as common in females compared to males and causes a significant psychological burden on patients and an economic burden on society. During the development of the disease, multiple cellular processes are involved, including the activation of JAK-STAT, MAPK, PI3KAKT, and Wnt signaling pathways, the subsequent production of cytokines, interleukins, and matrix me

Genetic Susceptibility to Sport-Related Muscle Injuries: Insights from the Literature and Novel Gene Candidates.

Physical activity is widely recognized for its health benefits; however, it also increases the risk of musculoskeletal soft tissue injuries, with muscle-related cases constituting a considerable proportion. These injuries impair well-being, athletic performance, and career longevity while creating substantial social and economic burdens. Their multifactorial etiology involves internal and external risk factors, and evidence suggests a heritable component influencing tissue integrity, recovery, and overall susceptibility. While genetic contributions to ligament and tendon damage are relatively

Model formulation and oscillatory patterns in immune-pathogen dynamics during Brucellosis infection.

Brucellosis, a global zoonosis imposing major health and economic burdens, is clinically marked by recurrent undulant fever linked to Brucella's persistence within macrophages. To decipher the immune-pathogen dynamics underlying this fever periodicity, this study develops a novel mathematical model that integrates macrophage self-renewal, logistic growth constrained by cellular carrying capacity, and intracellular Brucella replication. Stability and bifurcation analyses reveal two crucial thresholds: one for infection persistence, requiring R0>1, and another for the emergence of undulant fever

Oral Administration of a Tetrahedral Framework DNA-Based Nanoshot Attenuates Liver Fibrosis via Targeting Galectin-3.

Liver cirrhosis is a prevalent global health issue that has imposed a significant economic burden. Currently, there is a lack of effective treatment for liver cirrhosis due to poor patient compliance with injectable medications. Hepatic uncontrolled inflammation is the primary therapeutic target for liver cirrhosis. The oral administration of tetrahedral framework nucleic acid (tFNA) presents a promising approach due to its low hepatic toxicity and the ability to control inflammation, target the liver, and deliver small interfering RNAs (siRNAs). Here, we reported that tFNA could be effectivel

Research on inner membrane complex protein 1: a novel nanovaccines against Toxoplasma gondii.

Toxoplasma gondii (T. gondii) is a globally prevalent zoonotic parasite causing severe health and economic impacts. Despite decades of research, no commercial vaccine provides comprehensive protection against both acute and chronic toxoplasmosis. DNA vaccines represent a promising strategy, but their application is hindered by low delivery efficiency and limited immunogenicity. Here, we developed and evaluated pVAX1-TgIMC1-loaded PLGA and chitosan (CS) nanospheres as potential vaccine candidates. Immunization studies in mice showed that pVAX1-TgIMC1/PLGA and pVAX1-TgIMC1/CS nanospheres induced

Evaluating User Perceptions of a Vibrotactile Feedback System in Trunk Stabilization Exercises: A Feasibility Study.

Low back pain patients often have deficits in trunk stability. For this reason, many patients receive physiotherapy treatment, which represents an enormous socio-economic burden. Training at home could reduce these costs. The problem here is the lack of correction of the exercise execution. Therefore, this feasibility study investigates the applicability of a vibrotactile-controlled feedback system for trunk stabilisation exercises. A sample of 13 healthy adults performed three trunk stabilisation exercises. Exercise performance was corrected by physiotherapists using vibrotactile feedback. Th
