Detecting the Undetectable: Advances in Methods for Identifying Small Tau Aggregates in Neurodegenerative Diseases.

Tau, a microtubule-associated protein, plays a critical role in maintaining neuronal structure and function. However, in neurodegenerative d … u aggregates. Advanced single-molecule approaches, including single-molecule fluorescence resonance energy transfer (smFRET) and single-molecule pull-down (SiMPull), offer improved sensitivity for studying tau aggregation at the molecular level. These emerging tools provide critical insights into tau pathology, enabling earlier detection and characterisation of d …

Direct Optical Detection of Factor Xa Activity in Minimally Processed Whole Blood.

The ability to measure factor Xa activity directly in whole blood samples offers a path toward point-of-care monitoring and personalized anticoagulant dosage, potentially reducing bleeding risk and other anticoagulant-associated complications. We present a strategy to enable direct optical detection of factor Xa in minimally processed whole blood samples. Our strategy relies on a custom FRET-pair labeled DNA-peptide substrate, allowing FRET ratio to be monitored as an indicator of factor Xa activity. Substrates are tethered to a tapered-fiber sensor to allow evanescent detection of fluorescenc

Self-assembled nanoplatforms with ZIF-8 as a framework for FRET-based glutathione sensing in biological samples.

A glutathione (GSH) optical sensor with high sensitivity and exceptional selectivity was established for the first time. Zeolitic imidazolate framework-8 (ZIF-8) as a model was used for the first time to entrap Mn2+:ZnS quantum dots (QDs) and a rhodamine B derivative (RBD) by self-assembly. Benefiting from the confinement effect of ZIF-8, the loaded QDs and RBD can be brought into close proximity for energy transfer to occur. In the presence of GSH, the fluorescence resonance energy transfer (FRET) process from the QDs to RBD can be initiated, rendering the fluorescent nanoprobe to exhibit a r…

TP-FRET-Based Fluorescent Sensor for Ratiometric Detection of Formaldehyde in Real Food Samples, Living Cells, Tissues, and Zebrafish.

Formaldehyde (FA, HCHO) is a highly reactive carbonyl species, which is very harmful to humans and the environment as a tissue fixative and preservative. Therefore, developing some highly sensitive, selective, and rapid detection methods is significant for human health in food safety and environmental protection. Herein, a two-photon (TP) ratiometric sensor, CmNp-CHO, has been constructed by conjugating a TP donor (Π-push-pull-structure) with a FA off-on acceptor (functioned with hydrazide moiety) via a nonconjugated linker through the fluorescence resonance energy transfer mechanism. Such a s…

A simple FRET system using two-color CdTe quantum dots assisted by cetyltrimethylammonium bromide and its application to Hg(II) detection.

In this study, we developed a novel simple fluorescence resonance-energy transfer (FRET) system between two-color CdTe quantum dots (QDs) assisted by cetyltrimethylammonium bromide (CTAB). Mercaptopropionic (MPA)-capped CdTe QDs serving as both donors and acceptors were successfully synthesized by changing the refluxing time in aqueous solution. CTAB micelles formed in water and minimized the distance between the donors and acceptors significantly by electrostatic interactions, improving FRET efficiency. Several factors that affected the fluorescence spectra of the FRET system were investigate…

Photon cascade with clip-on fluorophores.

Bird on a wire: a recent study by Rant and Burley showed that it is possible to transfer excited-state energy along double-stranded DNA by arranging fluorophore relays with base-pair precision through Dervan-polyamides in a cascade-like process. The resulting assembly acts as the largest DNA-based photonic wire yet.

Conformations and electronic structures of axially coordinated fullerene-porphyrin-fullerene triad (C60CHCOO)2-Sn(IV) porphyrin.

The conformational (cis and trans) stability and electronic structures of (C(60)CHCOO)(2)-Sn(IV) porphyrin, recently synthesized as a novel fullerene-porphyrin-fullerene triad linked by metal axial coordination, have been studied by ab initio calculations. The cis conformer was found to be slightly more stable than the trans by 1.38 kcal/mol in the neutral compound. Upon the addition of an electron to the triad, the relative stability of the cis conformer was found to be higher (3.29 kcal/mol) than that in the neutral one. From the investigation of frontier molecular orbitals, for the cis conf

Computational insight into the antioxidant activity of thymol in diverse solvent environments: a DFT approach.

Natural phenolic compounds are a good source of antioxidant. Thymol, a naturally occurring substance containing a phenolic OH group and non-phenolic CHₓ (x = 1, 3) groups, are expectedly reported to possess antioxidant activity. In this study, its antioxidant potential was systematically studied using DFT along with the B3LYP hybrid functional and 6-311G ++ (d,p) basis set in gas and several variable solvent media. The major antioxidant mechanisms (HAT, SPLET, and SETPT) were calculated. Frontier molecular orbitals, energy gap, physico-chemical reactivity descriptors, Fukui functions, MEP surf

Recombination, dissociation, and transport of ion pairs across the liquid/liquid interface. implications for phase transfer catalysis.

Molecular dynamics simulations are used to calculate several free energy profiles relevant to the recombination/dissociation and transport of individual ions and ion pairs across the water/chloroform interface. Tetra methyl ammonium (TMA(+)) and tetra butyl ammonium (TBA(+)) (typically used as phase transfer catalysts) and a chloride ion (as an example of a transferred nucleophile) are considered. The free-energy profiles for the transfer of the three ions and the two ion pairs (TMA(+)Cl(-) and TBA(+)Cl(-)) across the interface, as well as the potential of mean force for the dissociation of th

On-Demand Charging Management Model and Its Optimization for Wireless Renewable Sensor Networks.

Nowadays, wireless energy transfer (WET) is a new strategy that has the potential to essentially resolve energy and lifespan issues in a wireless sensor network (WSN). We investigate the process of a wireless energy transfer-based wireless sensor network via a wireless mobile charging device (WMCD) and develop a periodic charging scheme to keep the network operative. This paper aims to reduce the overall system energy consumption and total distance traveled, and increase the ratio of charging device vacation time. We propose an energy renewable management system based on particle swarm optimiz…

An efficient dual-channel fluorescence probe for the detection of sulfur dioxide derivatives and viscosity.

In this work, a novel fluorescent probe (CBT) was developed by constructing an efficient fluorescence resonance energy transfer (FRET) and twisted intramolecular charge transfer (TICT) platform to identify SO2 and viscosity changes. CBT has excellent water solubility and exhibits superior fluorescence response performance without the involvement of organic solvents. The energy transfer efficiency (ETE) of CBT is quite high (99.2 %). Besides, CBT not only exhibits high selectivity (fluorescence intensity ratio enhanced 150-fold) and sensitivity (limit of detection: 24 nM) toward SO2 derivatives…

A Hydrogen Bonded Organic Framework Constructed from Mixed Valence Fe Clusters for Efficient H2O2 Photosynthesis.

A critical bottleneck in photocatalytic H2O2 production lies not only in the severe non-radiative energy losses incurred from enhancing light absorption, but also in a fundamental proton-electron kinetic imbalance. Here we introduce a light-heat-proton coupling strategy that harnesses the dissipated photothermal energy to activate proton dissociation from carboxylic acids, thereby creating a productive driving force for redox catalysis. A hydrogen-bonded organic framework (HOF-FJU-200) incorporating mixed-valence Fe2+/Fe3+ clusters is constructed via a metalloligand approach. The intervalence

Mechanisms and patterns of injury: the key to anticipation in trauma management.

Traumatic injuries are classified into blunt and penetrating injuries. Penetrating injuries are caused when an object such as a knife or bullet crushes and tears the tissues that it traverses. The energy transfer from the penetrating object to the tissues determines the degree of injury, and the wounding tract determines the pattern of injury. Blunt trauma produces injury by transferring energy through acceleration forces (+ and -) to the victim (usually from motor vehicles or falls). The pattern and severity of injury are determined by the magnitude and orientation of the acceleration change …
