Engineering Escherichia coli Nissle 1917 for the highly efficient biosynthesis of food-grade ergothioneine.

Ergothioneine (EGT) is a natural sulfur-containing amino acid possessing potent antioxidant activity. It demonstrates a range of beneficial biological functions, including anti-inflammatory, neuroprotective, and cardioprotective effects. Microbial fermentation presents a promising alternative to traditional extraction from edible fungi for EGT production, offering greater scalability, though its feasibility hinges critically on production titer and cost. In this study, to achieve food-grade EGT synthesis, the probiotic Escherichia coli Nissle 1917 was employed as the host, and the heterologous

In vitro detoxification of aflatoxin B1 by Lactiplantibacillus plantarum isolated from the north of Iran: A pioneering insights into the origin of fermented beverages.

The contamination of food and animal feeds with mycotoxions, particularly aflatoxin B1 (AFB1), poses significant risks to human health and causes economic losses. This study investigated bacteria from various fermented milk products to assess their ability to detoxify AFB1. A variety of household fermented kefir milk, kefir-like beverages, and kefir grains were collected from rural areas and subjected to microbiological analysis. Gram-positive bacterial isolates were further identified based on the 16S rRNA gene homology analysis. Seven bacterial isolates that were initially identified as lact

Current Perspectives on the Physiological Activities of Fermented Soybean-Derived Cheonggukjang.

Cheonggukjang (CGJ, fermented soybean paste), a traditional Korean fermented dish, has recently emerged as a functional food that improves blood circulation and intestinal regulation. Considering that excessive consumption of refined salt is associated with increased incidence of gastric cancer, high blood pressure, and stroke in Koreans, consuming CGJ may be desirable, as it can be made without salt, unlike other pastes. Soybeans in CGJ are fermented by Bacillus strains (B. subtilis or B. licheniformis), Lactobacillus spp., Leuconostoc spp., and Enterococcus faecium, which weaken the activity

Organic potassium salts or fibers effects on mineral balance and digestive fermentations in rats adapted to an acidogenic diet.

BACKGROUND: Fibers and potassium (K) organic salts in plant foods are liable to affect Ca and Mg balance at digestive and renal levels, respectively. K organic salts could counteract the acidifying effects of western diets and consequences of excess NaCl. AIM OF THE STUDY: To study this question, male rats were adapted to a basal acidifying low-K (LK) diet, or to diets supplemented with a fiber mix (LK/F), or K citrate (HK) or both (HK/F). RESULTS: HK and HK/F diets displayed a marked alkalinizing effect in urine and promoted citraturia, but this effect was not modulated by fibers. The effec

An outbreak of norovirus infection associated with fermented oyster consumption in South Korea, 2013.

An acute gastroenteritis (AGE) outbreak was reported in May 2013 in Gyeonggi Province, South Korea. Eight students who had eaten breakfast on 21 May 2013 at a high-school restaurant exhibited AGE symptoms. Our case-control study showed that a strong association was observed between AGE symptoms and fermented oyster consumption. Virological studies also indicated that noroviruses (NoVs) were detected from both clinical samples and fermented oyster samples, and multiple different genotypes (genogroups GII.4, GII.11 and GII.14) of NoVs were present in both samples. The nucleotide sequence similar

Microecological succession and quality enhancement of fermented tomato sour soup by synthetic functional microbiota.

Tomato sour soup, a traditional fermented food with health benefits, often exhibits inconsistent quality during natural fermentation. Three microbial consortia (yeasts, Eurotium, and their combination) were engineered to enhance fermentation. Physicochemical parameters, antioxidant activity, microbial communities, and metabolomic profiles were monitored throughout fermentation. Eurotium inoculation produced the strongest effects, including the highest total acid (21.10 g/kg), total phenolics (75.41 mg/g), and total flavonoids (29.41 mg/g), the lowest nitrite accumulation, and superior DPPH/ABT

Suppressing isovaleric acid off-flavor in Bacillus subtilis fermentation and its consequential effects on secondary metabolism.

Bacillus subtilis-fermented soybean foods (BFSFs) are nutritious East Asian staples. However, off-flavors dominated by isovaleric acid (IVA), a branched-chain fatty acid, limit consumer acceptance and industrial adoption. Although leucine- and acetolactate-derived pathways have been proposed, key regulatory nodes for IVA in soymilk remain unclear. This study systematically evaluated the key gene nodes controlling IVA biosynthesis during soymilk fermentation by B. subtilis, including bcd (leucine dehydrogenase, Bcd), bkdAB (branched-chain α-keto acid dehydrogenase complex, Bkd), ptb (phosphotra

Novel biosurfactants produced from food waste: a variability and validation study.

One-third of global food production is wasted, much of which is composted, gasified or landfilled. This food waste (FW) represents an untapped resource, particularly towards the production of biosurfactants. Genetically engineered Starmerella bombicola yeast strains offer a route to produce novel biosurfactants on FW with enhanced structural diversity, such as bola sophorosides. However, FW streams are variable, and often contain inhibitory compounds (e.g. lactic acid (LA), ethanol, etc.), posing bioprocessing challenges. Here we show that hydrolyzed FW consistently supports S. bombicola growt
