Assessing the impact of perfluoroalkyl substances on liver health: a comprehensive study using multi-donor human liver spheroids.

BACKGROUND: Although per- and polyfluoroalkyl substances (PFAS) have been linked to chronic liver diseases, the specific cellular and molecular mechanisms by which different PFAS contribute to human liver dysfunction remain unclear. This study aims to elucidate those mechanisms. METHODS: We exposed a multi-donor human liver spheroid model composed of multiple cell types to 20 µM of PFHxS, PFOA, PFOS, or PFNA for seven days, followed by single-cell RNA sequencing and lipid staining. RESULTS: PFAS impacted liver spheroids in a compound- and sex-specific manner. PFOA and PFHxS increased lipid a

Microplastics and PFAS as ubiquitous pollutants affect potencies of highly toxic chemicals in mixtures.

Pollutants usually occur as mixtures in the environment, where they affect each other's toxicity even at non-toxic concentrations. Despite the environmental relevance of interactions in complex mixtures, they remain severely understudied. Therefore, this study aimed to assess how PFAS and microplastics (MPs) as ubiquitous pollutants at (non-)toxic concentrations affect the potencies of highly toxic chemicals in mixtures. Experiments were performed to assess the toxicity of perfluorobutane sulfonamide (FBSA), linear low-density polyethylene MPs (0-50 µm), chlorpyrifos and imidacloprid to the re

Perfluorooctane sulfonic acid (PFOS) perturbs skeletal muscle oxidative phosphorylation by a different mechanism than liver.

PFOS perturbs normal energy metabolism such as lipid metabolism; however, despite being a highly metabolic organ, skeletal muscle toxicity has not been widely studied. Skeletal muscle, a healthy body's largest metabolic organ, presents a target for PFOS-induced metabolic toxicity due to its high mitochondrial content. Therefore, we aimed to evaluate the response of skeletal muscle to PFOS in vivo and compare the responses to liver. Mice were treated with 0, 1, or 10 mg/kg/day PFOS for 21-days and gastrocnemius and liver collected. All mice exhibited a dose-dependent decrease in bodyweight foll

Influence of separator gel in blood sampling tubes for perfluoroalkyl and polyfluoroalkyl substances analysis.

INTRODUCTION: For several years, concerns about perfluoroalkyl and polyfluoroalkyl substances (PFAS) have been growing, and clinical laboratories may have to perform a growing number of PFAS analyses. The use of serum tubes without a separator gel is currently recommended for the quantification of PFAS due to the concern that the compounds may adsorb to the gel. The impact of gel adsorption on the accuracy of the results, however, has not been evaluated. METHODS: Aliquots from a pool of blood spiked with PFAS were stored in gel-free clot activator tubes (CATs) and gel-containing BD SST II Adv

Optimizing sterile filtration of nanoemulsions through proper choice of prefilter properties.

Nanoemulsions, with droplet sizes between 20 and 200 nm, have emerged as a promising vaccine adjuvant and drug delivery system, enhancing the solubility of hydrophobic drugs for diverse applications. Sterile filtration of nanoemulsions is particularly challenging due to the similar size between the nanodroplets and the 0.2 μm nominal pore size rating of sterile filters. One approach to reducing membrane fouling, and enhancing filtration capacity and yield, is to employ an appropriate prefilter, but there are currently no clear guidelines on how to select the prefilter pore size, chemistry, or

PFAS in biotic and abiotic matrices in coastal and estuarine ecosystems: Temporal and seasonal distribution, discharge and environmental impacts.

Besides freshwater ecosystems such as lakes and rivers, estuaries and coastal regions are crucial to the global distribution of per- and polyfluoroalkyl substances (PFAS) through the ocean and their impacts and transport throughout the food web. This review includes a comprehensive assessment of the concentration and distribution of legacy and emerging PFAS compounds in living species, such as plants and aquatic creatures, as well as in abiotic components, such as surface water and sediment within estuarine ecosystems. This paper also explores the temporal and seasonal patterns of PFAS emissio

Temporal effects of perfluorononanoic acid (PFNA) exposure on metabolic disorders and gut-liver axis injury in large yellow croaker (Larimichthys crocea).

To investigate the toxic effects of PFNA on aquatic organisms, this study used large yellow croaker (L. crocea) as a model and examined the impacts of 1000 ng/L PFNA exposure for 3, 7, and 14 days on the hepatic and intestinal systems. Histopathological examination, transcriptomic profiling, and 16S rRNA gene sequencing were employed to evaluate tissue damage, gene expression changes, and gut microbial alterations. The results revealed that PFNA exposure induced progressive histopathological changes in the liver, including nuclear enlargement and vacuolization, with increasing severity over ti

Ultrasound Erosion of Rabbit Liver Induced by Locally Injected Phase-Shift Acoustic Droplets and With Lauromacrogol.

OBJECTIVES: Our previous studies have found that low-frequency, low-pressure, weakly focused ultrasound (FUS) can induce acoustic droplet vaporization (ADV) of perfluoropentane (PFP) droplets and result in localized liver and prostate tissue controllable cavitation resonance and mechanical damage. To further investigate the mechanical erosion induced by ultrasound and locally injected phase-shift acoustic droplets in rabbit liver. METHODS: The liver of each rabbit was treated with perfluoromethylcyclopentane (PFMCP) alone, FUS combined with PFMCP (FUS + PFMCP), and FUS combined with PFP (FUS

Analysis of Perfluoroalkyl Substances (PFASs) in Settled Dust by Liquid Chromatography with Tandem Mass Spectrometry: Method Validation and Application to Vietnamese House and Road Dusts.

Information about perfluoroalkyl substances (PFASs) like perfluoroalkyl carboxylic acids (PFCAs) and sulfonates (PFSs) in settled dust from emerging and developing countries is still limited, partly due to the lack of efficient analytical methods. In this study, a reliable, simple, and cost-effective analytical procedure was developed and validated to determine 12 PFCAs and 4 PFSs in dust samples. The samples were ultrasonicated with methanol, followed by a dispersive sorbent clean-up step with graphitized carbon and quantification by liquid chromatography with tandem mass spectrometry (LC-MS/

Morphological and cellular effects of lead, cadmium, and perfluorooctanoic acid on Gallus gallus embryonic development: Insights into cephalic region vulnerabilities.

Lead (Pb), cadmium (Cd), and perfluorooctanoic acid (PFOA) are environmental pollutants with bioaccumulative properties, posing risks to embryonic development, particularly through cephalic region malformations. Despite their significance, few studies have investigated their effects at realistic concentrations or in mixtures. This study assessed the individual and combined effects of Pb, Cd, and PFOA on Gallus gallus embryos at relevant concentrations, focusing on the cephalic region at stages HH10 and HH14. Exposure to contaminant mixtures, particularly Pb + Cd, Pb + PFOA, and the full combin

Comparing the effects of developmental exposure to alpha lipoic acid (ALA) and perfluorooctanesulfonic acid (PFOS) in zebrafish (Danio rerio).

Alpha lipoic acid (ALA) is a dietary supplement that has been used to treat a wide range of diseases, including obesity and diabetes, and have lipid-lowering effects, making it a potential candidate for mitigating dyslipidemia resulting from exposures to the per- and polyfluoroalkyl substance (PFAS) family member perfluorooctanesulfonic acid (PFOS). ALA can be considered a non-fluorinated structural analog to PFOS due to their similar 8-carbon chain and amphipathic structure, but, unlike PFOS, is rapidly metabolized. PFOS has been shown to reduce pancreatic islet area and induce β-cell lipotox

Low levels of per- and polyfluoroalkyl substances (PFAS) detected in drinking water in Norway, but elevated concentrations found near known sources.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous contaminants which are also found in drinking water. Concentration levels in drinking water vary widely and range from a very low contribution to total daily exposure for humans to being the major source of uptake of PFAS. PFAS concentrations in Norwegian drinking water has been rarely reported. We investigated concentrations of 31 PFAS in 164 water samples, representing both source water (i.e., before drinking water treatment) and finished drinking water. Samples were taken from 18 different water bodies across Norway. The 17 waterwork

Hydrophobic Sorption Properties of an Extended Series of Anionic Per- and Polyfluoroalkyl Substances Characterized by C18 Chromatographic Retention Measurement.

Partitioning from water to nonaqueous phases is an important process that controls the behavior of contaminants in the environment and biota. However, for ionic chemicals including many perfluoroalkyl and polyfluoroalkyl substances (PFAS), environmentally relevant partition coefficients cannot be predicted using the octanol/water partition coefficient, which is commonly used as a hydrophobicity indicator for neutral compounds. As an alternative, this study measured C18 liquid chromatography retention times of 39 anionic PFAS and 20 nonfluorinated surfactants using isocratic methanol/water elue

Polyvinylidene fluoride binder removal through subcritical methanol for efficient liberation of cathode materials from lithium-ion batteries.

Polyvinylidene fluoride (PVDF) binder removal is critical for the recovery of valuable metal materials during the treatment of spent lithium-ion batteries (LIBs). This study proposed a new PVDF removal method through subcritical methanol extraction. The optimal conditions and mechanism of the method for the liberation of cathode materials were explored, and the recovered cathode materials, aluminum foils (Al foils), and extracted binder were characterized. Experimental results on actual cathode sheets show that under the extraction temperature of 200 °C, after holding and stirring time for 10

Characterization of per- and polyfluoroalkyl substances (PFAS) concentrations in a community-based sample of infants from Samoa.

Per- and polyfluoroalkyl substances (PFAS) are persistent contaminants with documented harmful health effects. Despite increasing research, little attention has been given to studying PFAS contamination in low- and middle-income countries, including Samoa. Using data and biosamples collected through the Foafoaga o le Ola ("Beginning of Life") Study, which recruited a sample of mothers and infants from Samoa, we conducted an exploratory study to describe concentrations of 40 PFAS analytes in infant cord blood collected at birth (n = 66) and infant dried blood spots (DBS) collected at 4 months p

Effects of Perfluorooctane Sulfonate on Cerebellar Cells via Inhibition of Type 2 Iodothyronine Deiodinase Activity.

Perfluorooctane sulfonate (PFOS) has been used in a wide variety of industrial and commercial products. The adverse effects of PFOS on the developing brain are becoming of a great concern. However, the molecular mechanisms of PFOS on brain development have not yet been clarified. We investigated the effect of early-life exposure to PFOS on brain development and the mechanism involved. We investigated the change in thyroid hormone (TH)-induced dendrite arborization of Purkinje cells in the primary culture of newborn rat cerebellum. We further examined the mechanism of PFOS on TH signaling by re

Insight into the binding model of per- and polyfluoroalkyl substances to proteins and membranes.

Legacy per- and polyfluoroalkyl substances (PFASs) have elicited much concern because of their ubiquitous distribution in the environment and the potential hazards they pose to wildlife and human health. Although an increasing number of effective PFAS alternatives are available in the market, these alternatives bring new challenges. This paper comprehensively reviews how PFASs bind to transport proteins (e.g., serum albumin, liver fatty acid transport proteins and organic acid transporters), nuclear receptors (e.g., peroxisome proliferator activated receptors, thyroid hormone receptors and rep

A review of cardiovascular effects and underlying mechanisms of legacy and emerging per- and polyfluoroalkyl substances (PFAS).

Cardiovascular disease (CVD) poses the leading threats to human health and life, and their occurrence and severity are associated with exposure to environmental pollutants. Per- and polyfluoroalkyl substances (PFAS), a group of widely used industrial chemicals, are characterized by persistence, long-distance migration, bioaccumulation, and toxicity. Some PFAS, particularly perfluorooctane sulfonic acid (PFOS), perfluorooctanoic acid (PFOA) and perfluorohexanesulfonic acid (PFHxS), have been banned, leaving only legacy exposure to the environment and human body, while a number of novel PFAS alt
