Advancements in the treatment of hypothyroidism with L-T4 liquid formulation or soft gel capsule: an update.

The most recent advance concerning levothyroxine (L-T4) therapy is the development of novel oral formulations: the liquid preparation, and the soft gel capsule. Areas covered: This review evaluates the most recent clinical studies about these new formulations. The liquid formulation has been shown to overcome: the food and beverages intereference with L-T4 tablets absorption, caused by food or coffee at breakfast; malabsorption induced by the increased gastric pH, resulting from atrophic gastritis, or due to proton-pump inhibitors; and malabsorption after bariatric surgery. The use of liquid L

A two-factor model of rotation-induced motion sickness syndrome in squirrel monkeys.

Nonrestrained squirrel monkeys were rotated repeatedly in the horizontal plane once per day for preset times or until a vomiting response occurred. Emesis latencies and frequencies were recorded. Some subjects were fed fresh banana immediately before and after each rotation. The amount consumed provided a measure of conditioned food aversion or feeding suppression. A two-factor model of the motion-sickness syndrome was formulated to account for the data. According to the model, appropriate stimulation of the semicircular canals accompanied by visual and proprioceptive stimulation generates two

Enhancing Bioavailability: Advances in Oral Isotretinoin Formulations.

Oral isotretinoin continues to be unsurpassed in efficacy for acne. However, it is associated with potential adverse events including risk of fetal defects, necessitating appropriate mitigation strategies. Furthermore, the variance in bioavailability of the original formulation when ingested in fed versus fasted conditions can lead to differences in daily dosing and duration of exposure. Advances in formulation, with lidose encapsulation and subsequently with micronization, have led to iterative improvements in reducing bioavailability variation between fed and fasted conditions. Differences i

Can time-dependent optimal interventions reduce Salmonella spread? A multi-pathway ODE model using real data from EU.

Salmonella remains a major public health concern worldwide due to its complex transmission pathways involving humans, food, the environment, and vectors, making efficient allocation of limited control resources essential for effective disease management. In this study, we develop a comprehensive compartmental ordinary differential equation (ODE) model to investigate optimal intervention strategies for reducing Salmonella transmission. The model incorporates multiple transmission routes and is calibrated using reported Salmonella case data from the EU/EEA spanning 2007-2023. Global sensitivity

Pharmacokinetics and Pharmacodynamics of Posaconazole.

Posaconazole is typically used for preventing invasive yeast and mold infections such as invasive aspergillosis in high-risk immunocompromised patients. The oral suspension was the first released formulation and many pharmacokinetic and pharmacodynamic studies of this formulation have been published. Erratic absorption profiles associated with this formulation were widely reported. Posaconazole exposure was found to be significantly influenced by food and many gastrointestinal conditions, including pH and motility. As a result, low posaconazole plasma concentrations were obtained in large grou

Bioavailability of aspirin in fasted and fed states of a novel pharmaceutical lipid aspirin complex formulation.

Dyspeptic symptoms are common with aspirin and clinicians frequently recommend that it be taken with food to reduce these side effects. However, food can interfere with absorption, especially with enteric-coated aspirin formulations. We evaluated whether food interferes with the bioavailability of a new, pharmaceutical lipid-aspirin complex (PL-ASA) liquid-filled capsule formulation. In this randomized, open label, crossover study, 20 healthy volunteers fasted for ≥ 10 h and then randomized as either "fasted", receiving 650 mg of PL-ASA, or as "fed", with a standard high-fat meal and 650 mg of

A household-level sweet potato-based infant food to complement vitamin A supplementation initiatives.

Vitamin A deficiency (VAD) prevalence in Sub-Saharan Africa is high in spite of vitamin A supplementation programmes among children in most countries. Plant-based complementary foods remain the key source of nutrients in addition to breast milk for infants in lower income countries. Cereal-legume blends are superior in protein and energy densities compared with maize, millet or sorghum-only porridge. However, unfortified cereal-legume and cereal-only porridges are low in vitamin A. A household-level sweet potato-based infant food, rich in vitamin A, has been developed to complement vitamin A s

A Review of the Risk of Cow's Milk Protein Allergy in Oral Medication.

Immunoglobulin E (IgE)-mediated cow's milk allergy (CMA) is a common food reaction resulting from the consumption of cow's milk protein (CMP). The clinical manifestations of CMA include mild to severe urticaria, skin-manifested hypersensitivity reactions, and anaphylaxis. Food allergies may affect 8% of children and 10% of adults. The Federal Food, Drug, and Cosmetic Act (FD&C Act) requires that the label of a food must declare the presence of a "major food allergen" (MFA) contained in the food or ingredient. The Food and Drug Administration (FDA) generally regards milk protein concentrate (MP

Simple Strategy to Protect Lactase Activity in Solid Formulation.

BACKGROUND: Lactose intolerance is characterized by the absence of the enzyme lactase (beta-galactosidase) and affects two thirds of the world adult population. Our aim was to evaluate a lactase gastro-resistant formulation to ensure increased activity in the action site of the enzyme (lumen of the small intestine). Simultaneously, we also evaluated the commercial product stability and enzyme activity, because the product containing beta-galactosidase is classified as food supplement according to the Food and Drug Administration (FDA), so it is free to pass quality testing, efficacy and stabil

Long-Acting Growth Hormone Preparations in the Treatment of Children.

Human growth hormone (hGH), which had been in use since 1958, was supplanted by recombinant human growth hormone (rhGH) in 1985 for those with growth hormone deficiency (GHD). Adherence to daily subcutaneous growth hormone is challenging for patients. Thus, several companies have pursued the creation of long acting rhGH. These agents can be divided broadly into depot formulations, PEGylated formulations, pro-drug formulations, non-covalent albumin binding GH and GH fusion proteins. Nutropin Depot is the only long acting rhGH ever approved by the U.S. Food and Drug Administration, and it was re

Abuse-deterrent formulations, an evolving technology against the abuse and misuse of opioid analgesics.

The increased use of opioid pain medication has been mirrored by the increased misuse and abuse of these drugs. As part of a multidisciplinary approach to this epidemic, pharmaceutical companies, with the encouragement of the Food and Drug Administration, have increased the development of abuse-deterrent formulations. While all have the goal of treating pain while mitigating misuse and abuse, there are different technologies utilized to impart the abuse-deterrent properties. The goal of this paper is to review the basis of abuse-deterrent formulations, the different types and approaches of som

Dietary sources of copper.

The dietary intakes of copper by children and adults who consume free diets are often significantly lower than the recommended daily allowances of the National Academy of Sciences (USA) or the World Health Organization. These lower-than-recommended intakes of copper appear to be adequate for healthy individuals since states of copper deficiency have not been observed in the absence of an accompanying metabolic disorder. Copper deficiencies have arisen in pre-term infants of low birth weight as a result of breast-milk diets, and in children and adults as a result of fluids that are used for tot

Food insufficiency exists in the United States: results from the third National Health and Nutrition Examination Survey (NHANES III).

OBJECTIVES: The purpose of this study was to estimate the prevalence of food insufficiency in the United States and to examine sociodemographic characteristics related to food insufficiency. METHODS: Data were analyzed from the third National Health and Nutrition Examination Survey, a cross-sectional representative sample of the civilian noninstitutionalized population living in households. Individuals were classified as "food insufficient" if a family respondent reported that the family sometimes or often did not get enough food to eat. RESULTS: From 1988 through 1994, the overall prevalenc

Background: German chamomile (Matricaria chamomilla L.) and Roman chamomile (Chamaemelum nobile (L.) All.) are widely consumed botanical materials increasingly used in nutraceuticals and functional foods. Although both are marketed as "chamomile", they differ in phytochemical composition, sensory profile, and potential health-promoting properties. This review compares both species as dietary and functional ingredients. Methods: This narrative review summarises current evidence on the botanical characteristics, phytochemical profiles, dietary forms, biological activities, safety aspects, and fu

Microbial Protein as a novel ingredient in pet food: A food preference study in dogs and cats.

The increasing demand for protein sources in pet nutrition has driven growing interest in microbial proteins (MP) as viable alternatives to traditional animal proteins. However, their successful incorporation into pet food formulations depends not only on nutritional adequacy but also on short-term food preference. This study evaluated the palatability of dry extruded diets containing 10% microbial protein in dogs and cats using an in-home two-bowl preference test and complemented these findings with independent laboratory characterization to support interpretation of the observed preferences.

Pharmacologic and clinical comparison of cefaclor in immediate-release capsule and extended-release tablet forms.

A new controlled-delivery, extended-release 500-mg formulation of cefaclor that is administered twice daily may improve patient compliance compared with the older, immediate-release 250-mg formulation that is administered three times daily. When the extended-release tablet is administered with food, peak plasma cefaclor concentrations are achieved about 2.5 hours after the dose compared with about 1 hour after a dose with the immediate-release capsule. However, the two formulations have an equivalent extent of absorption and equivalent pharmacokinetics after absorption. Cefaclor has retained i

Soy protein formulas in children: no hormonal effects in long-term feeding.

Recently, the finding of high plasma concentration of phyto-oestrogens in soy protein formula (SPF) fed children has focused scientific attention on the phyto-oestrogens (isoflavones genistein, daidzein, and their glycosides) contained in SPFs. The aim of this study was to evaluate some hormonal and metabolic effects of long-term (more than 6 months) SPF feeding. We enrolled 48 children, mean age 37 months (range 7-96 months), 27 males and 21 females. All children underwent physical examination. Bone age, urinary markers of bone metabolism, serum levels of bone alkaline phosphatase, osteocalci

Multiple-locus variable number of tandem repeat analysis (MLVA) of Listeria monocytogenes directly in food samples.

Listeria monocytogenes is the etiologic agent of listeriosis responsible for severe and fatal infections in humans. Listeria contamination occurs quite often in a wide range of foods due to its ubiquitous nature. Isolates need to be characterized to a strain level for accurate diagnosis of Listeria infection, epidemiological studies, investigation of outbreaks and effective prevention and control of food-borne listeriosis. The purpose of this research was to evaluate the multiple-locus variable number of tandem repeat analysis (MLVA) for sub-typing L. monocytogenes isolates in pure cultures an
