Detection of Fusarium solani using cutinase antibody and its application in diagnosing fungal keratitis in an animal model.

Fungal keratitis (FK) is a sight-threatening infectious disease that can result in blindness if not appropriately treated. Although keratitis associated with filamentous fungi was rarely reported in the past, the incidence of fungal keratitis due to contact lens usage has increased. The clinical manifestations of fungal keratitis often resemble those of other corneal infections, potentially delaying accurate diagnosis and treatment. In this study, we developed a Fusarium-specific polyclonal peptide antibody targeting the cutinase of F. solani and evaluated its diagnostic potential for FK in an…

Phase-Dependent Differential In Vitro and Ex Vivo Susceptibility of Aspergillus flavus and Fusarium keratoplasticum to Azole Antifungals.

Fungal keratitis (FK) is an invasive infection of the cornea primarily associated with Aspergillus and Fusarium species. FK is treated empirically with a limited selection of topical antifungals with varying levels of success. Though clinical infections are typically characterized by a dense network of mature mycelium, traditional models used to test antifungal susceptibility of FK isolates exclusively evaluate susceptibility in fungal cultures derived from asexual spores known as conidia. The purpose of this study was to characterize differences in fungal response when topical antifungal trea…

Molecular evidence for the involvement of a polygalacturonase-inhibiting protein, GhPGIP1, in enhanced resistance to Verticillium and Fusarium wilts in cotton.

Polygalacturonase-inhibiting protein (PGIP), belonging to a group of plant defence proteins, specifically inhibits endopolygalacturonases secreted by pathogens. Herein, we showed that purified GhPGIP1 is a functional inhibitor of Verticillium dahliae and Fusarium oxysporum f. sp. vasinfectum, the two fungal pathogens causing cotton wilt. Transcription of GhPGIP1 was increased in cotton upon infection, wounding, and treatment with defence hormone and H2O2. Resistance by GhPGIP1 was examined by its virus-induced gene silencing in cotton and overexpression in Arabidopsis. GhPGIP1-silenced cotton …

Fusarium falciforme infection of foot in a patient with type 2 diabetes mellitus: a case report and review of the literature.

Fungal infections of foot in patients with diabetes are not uncommon; however, foot infection due to Fusarium species has been rarely reported. We report here a case of a 50-year-old male with type 2 diabetes who developed multiple spontaneous nodular lesions on right foot without any systemic symptoms and signs for 6 months. The lesions were unresponsive to broad-spectrum antibacterial treatment. Fine needle aspiration cytology of nodular lesions revealed the presence of fungal hyphae, and Fusarium species was isolated from the same sample which was identified as Fusarium solani species compl…

Transgenic expression of polygalacturonase-inhibiting proteins in Arabidopsis and wheat increases resistance to the flower pathogen Fusarium graminearum.

Fusarium head blight (FHB), caused by Fusarium graminearum, is one of the most important diseases of wheat worldwide, resulting in yield losses and mycotoxin contamination. The molecular mechanisms regulating Fusarium penetration and infection are poorly understood. Beside mycotoxin production, cell wall degradation may play a role in the development of FHB. Many fungal pathogens secrete polygalacturonases (PGs) during the early stages of infection, and plants have evolved polygalacturonase-inhibiting proteins (PGIPs) to restrict pectin degradation during fungal infection. To investigate the r…

Expression of the cyanide hydratase enzyme from Fusarium lateritium in Escherichia coli and identification of an essential cysteine residue.

The filamentous fungus Fusarium lateritium is cyanide tolerant, due partly to the induction of the enzyme cyanide hydratase in the presence of cyanide. This enzyme catalyses the hydration of cyanide to formamide. The expression in Escherichia coli of a cDNA clone encoding cyanide hydratase is described. The cDNA cloned was expressed as a transcriptional fusion in the expression vector pKK233-2 and a high level of activity of cyanide hydratase was detected in E. coli. Site-directed mutagenesis of the cys-163 residue inactivated the enzyme.

Induction of tibial dyschondroplasia and suppression of cell-mediated immunity in chickens by Fusarium oxysporum grown on sterile corn.

An isolate of Fusarium oxysporum from corn associated with Kaschin-Beck disease in humans was tested for its ability to induce tibial dyschondroplasia (TD) and toxicity in chicks. Both leghorn and broiler chicks were fed diets in which corn was replaced with varied amounts (0% to 50%) of the F. oxysporum culture grown on sterile corn, or with known TD-inducing agents. F. oxysporum did not affect body weight in either type of chicks. In leghorn chicks, neither F. oxysporum nor the known TD-inducing agents (F. equiseti, 4%; tetramethylthiuram disulfide [Thiram], 35 ppm) caused TD. However, F. ox…

Suppression of immune response in the B6C3F1 mouse after dietary exposure to the Fusarium mycotoxins deoxynivalenol (vomitoxin) and zearalenone.

The effect that dietary exposure to the naturally-occurring Fusarium graminearum toxins deoxynivalenol (DON) and zearalenone (ZEA) may have on immune function was assessed in the B6C3F1 mouse. Dietary DON depressed the plaque-forming response to sheep red blood cells, the delayed hypersensitivity response to keyhole limpet haemocyanin and the ability to resist Listeria monocytogenes. Listerial resistance was similarly decreased in control mice fed restricted diets comparable to the dietary restriction caused by DON-induced feed refusal, whereas equivalent food restriction did not decrease the …

Fusarium asiaticum FaSdhC2 Point Mutation A83V Confers Resistance to Pydiflumetofen and Development of a Rapid LAMP Detection Method.

Fusarium head blight (FHB) caused by Fusarium graminearum species complex (FGSC) is a devastating disease worldwide. The new succinate dehydrogenase inhibitor pydiflumetofen is highly effective against FHB, but its growing use raises resistance concerns. In this study, we identified four resistant Fusarium asiaticum isolates from 4088 field isolates collected across eight provinces in China in 2025. Gene replacement tests confirmed that the single-point mutation A83V of FaSdhC2 confers resistance. Notably, the resistant isolates did not exhibit significant fitness costs related to pydiflumetof…

Genome-Wide Analysis of Fusarium oxysporum f. sp. cubense Peroxidases Reveals Oxidative-Stress Responses and Infection-Associated Expression.

Banana Fusarium wilt, caused by the soil-borne fungus Fusarium oxysporum f. sp. cubense (Foc), is a devastating disease and a major threat to global banana production. Here, we identified 24 genes predicted to encode heme-dependent peroxidase- or catalase-peroxidase-related proteins in Foc race 4 (Foc4), whereas nine genes encoding thiol-dependent peroxide-reducing proteins were catalogued separately. Phylogenetic, synteny, and Ka/Ks analyses indicated a conserved peroxidase repertoire under strong purifying selection, without substantial lineage-specific expansion. Motif, domain, gene-structu…

Estimation of Phenylalanine Ammonia-Lyase Activity for Assessing Resistance of Muskmelon Against Fusarium oxysporum f. sp. melonis Infection.

Phenylalanine ammonia-lyase is a key enzyme in the phenylpropanoid pathway, responsible for the conversion of L-phenylalanine into trans-cinnamic acid. This step is crucial for the biosynthesis of lignin, phytoalexins, and other phenolic compounds that play a significant role in plant defense responses. This chapter describes a detailed protocol for the extraction and quantitative estimation of phenylalanine ammonia-lyase activity in muskmelon (Cucumis melo L.) tissues in response to Fusarium oxysporum f. sp. melonis (FOM) infection. The method enables researchers to compare phenylalanine ammo…

From sea to cure: Discovery of marine-derived therapeutics against Fusarium solani in shrimps for enhancing aquaculture sustainability.

Fusarium solani, an Ascomycota filamentous fungus species, causes shell disease or mycotic infections in wild and farmed shrimps. It causes black gill disease in shrimps, which has no specific treatments, so cutting-edge pharmaceutical research to prevent glutamine synthetase is needed to stop it and reduce its negative effects on aquaculture productivity and health. In silico drug design has been evaluated as an innovative treatment for black gill disease in shrimps caused by F. solani. Initially, molecular docking targeted the Glutamine synthetase (AF-Q9UUN6-F1-v4), utilising a set of 1,191 …

Analysis of Wheat Grain Infection by Fusarium Mycotoxin-Producing Fungi Using an Electronic Nose, GC-MS, and qPCR.

Fusarium graminearum and F. culmorum are considered some of the most dangerous pathogens of plant diseases. They are also considerably dangerous to humans as they contaminate stored grain, causing a reduction in yield and deterioration in grain quality by producing mycotoxins. Detecting Fusarium fungi is possible using various diagnostic methods. In the manuscript, qPCR tests were used to determine the level of wheat grain spoilage by estimating the amount of DNA present. High-performance liquid chromatography was performed to determine the concentration of DON and ZEA mycotoxins produced by t…
