Natural History of Prenatally Identified Children with 48,XXYY Syndrome in Infancy and Early Childhood.

BACKGROUND: 48,XXYY syndrome is a rare sex chromosome aneuploidy (SCA) characterized by neurodevelopmental deficits and medical comorbidities. The limited information available in the literature is almost exclusively limited to postnatally diagnosed cases. This study aims to describe the early medical and developmental features of prenatally identified 48,XXYY infants, with comparisons to 47,XYY, 47,XXY cohorts, and typical populations, as well as previously reported postnatally diagnosed 48,XXYY cases. METHODS: The eXtraordinarY Babies Study prospectively follows children prenatally identifi

Diverse virulence traits underlying different clinical outcomes of Salmonella infection.

Salmonella strains have evolved to infect a wide variety of reptiles, birds, and mammals resulting in many different syndromes ranging from colonization and chronic carriage to acute fatal disease. Adaptation to a large number of different evolutionary niches has undoubtedly driven the high degree of phenotypic and genotypic diversity in Salmonella strains. Differences in LPS and flagellar structure generate the antigenic variation that is reflected in the more than 2,000 known serotypes. Moreover, variations of LPS structure affect the virulence of the strain. The differential expression of v

Hantavirus: A Comprehensive Narrative Review of Virology, Epidemiology, Pathogenesis, Clinical Manifestations, Diagnostics, and Emerging Therapeutic Strategies.

Hantaviruses are a genus of negative-sense, single-stranded, tri-segmented RNA viruses that are distributed worldwide. They cause two severe zoonotic diseases in humans: haemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS), also known as hantavirus cardiopulmonary syndrome (HCPS). Hantaviruses are primarily spread by inhaling aerosolised excreta from infected rodent reservoirs. Over 50 rodent species on six continents have been found to carry hantaviruses, with only one reported case of human-to-human transmission, involving the Andes virus (ANDV) in South Amer

The role of stress and the hypothalamic-pituitary-adrenal axis in the pathogenesis of the metabolic syndrome: neuro-endocrine and target tissue-related causes.

The stress system coordinates the adaptive response of the organism to real or perceived stressors. The main components of the stress system are the corticotropin-releasing hormone (CRH) and locus ceruleus-norepinephrine/ autonomic (LC/NE) systems and their peripheral effectors, the hypothalamic-pituitary-adrenal (HPA) axis, and the limbs of the autonomic system. Activation of the stress system leads to behavioral and peripheral changes that improve the ability of the organism to adjust homeostasis and increase its chances for survival. Thus, CRH and the LC/NE system stimulate arousal and atte

PLGA-particle-based vaccine induces SARS-CoV-2-specific antibody and T-cell responses.

BACKGROUND AND PURPOSE: Current coronavirus disease 2019 (COVID-19) vaccines effectively prevent severe disease but induce primarily systemic immunity without mucosal protection in the respiratory tract, which is mandatory for protection from severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and the prevention of viral transmission. Vaccination strategies capable of inducing local immunity at the site of infection are therefore needed. Here, we evaluated a poly (lactic-co-glycolic acid) (PLGA) microparticle-based vaccine co-encapsulating the receptor binding domain (RBD) o

Bioluminescent label-free molecular switch for SARS-CoV-2 detection.

BACKGROUND: Simple, single-step platforms for virus detection are still important for improving diagnostic testing beyond laboratory settings. Molecular switch platforms, which generate measurable signals upon specific target binding, represent a promising strategy for virus detection due to their modularity, high sensitivity, and potential adaptability to emerging pathogens. RESULTS: Here, we present CovSwitch, a novel one-step bioluminescent molecular switch platform engineered for rapid, sensitive, and potentially point of care detection-compatible detection of severe acute respiratory syn

ICA-based Individualized Differential Structure Similarity Networks for Predicting Symptom Scores in Adolescents with Major Depressive Disorder.

Major depressive disorder (MDD) is a complex mood disorder characterized by persistent and overwhelming depression. Previous studies have identified large scale structural brain alterations in MDD, yet most are group analyses with atlas-parcellated anatomical regions. Here we proposed a method to measure individual difference by independent component analysis (ICA)-based individual difference structural similarity network (IDSSN). This approach provided a data-adaptive, atlas-free solution that can be applied to new individual subjects. Specifically, we constructed individualized whole-brain s

Antiviral therapy for COVID-19: Derivation of optimal strategy based on past antiviral and favipiravir experiences.

Favipiravir, a broad-spectrum RNA-dependent RNA polymerase inhibitor, inhibits the replication of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) at significantly lower concentrations than the plasma trough levels achieved by the dosage adopted for influenza treatment and exhibits efficacy against coronavirus disease 2019 (COVID-19) pneumonia. Although high doses of favipiravir are required due to the molecule being a purine analog, its conversion into the active form in infected cells with active viral RNA synthesis enhances the antiviral specificity and selectivity as a chain te

Integrated Engineering of CAR-T Cells for Solid Tumours.

Chimeric antigen receptor (CAR) T-cell therapy has achieved durable efficacy in hematologic malignancies but encounters persistent obstacles in solid tumours, including antigen heterogeneity, a suppressive tumour microenvironment (TME), and intrinsic T-cell dysfunction. This review examines the transition from single-axis engineering to an integrated framework that addresses these hurdles in sequence. We delineate how next-generation CAR-T cells are designed for precise spatiotemporal activation through logic-gated and pharmacologically regulatable receptors, while being reinforced by metaboli

Charting new territories in cardiac care intensive care rehabilitation and the enduring impact on post-acute cardiac event outcomes.

Cardiac intensive care unit (CICU) survivors face significant morbidity and mortality after hospital discharge. Many develop new or worsening physical, cognitive, or psychological impairments consistent with post-intensive care syndrome (PICS). Our current knowledge regarding the epidemiology of and risk factors for PICS is largely derived from general intensive care unit (ICU) cohorts. CICU care includes distinct exposures and pathophysiologic characteristics, including cardiogenic shock, cardiac arrest, temporary mechanical circulatory support, and complex cardiovascular interventions that m

A dynamic ensemble model for short-term forecasting in pandemic situations.

During the COVID-19 pandemic, many hospitals reached their capacity limits and could no longer guarantee treatment of all patients. At the same time, governments endeavored to take sensible measures to stop the spread of the virus while at the same time trying to keep the economy afloat. Many models extrapolating confirmed cases and hospitalization rate over short periods of time have been proposed, including several ones coming from the field of machine learning. However, the highly dynamic nature of the pandemic with rapidly introduced interventions and new circulating variants imposed non-t
