Myelodysplastic syndromes (MDS) and MDS/myeloproliferative neoplasms (MPNs) are clonal diseases that differ in morphologic diagnostic criteria but share some common disease phenotypes that include cytopenias, propensity to acute myeloid leukemia evolution, and a substantially shortened patient survival. MDS/MPNs share many clinical and molecular features with MDS, including frequent mutations involving epigenetic modifier and/or spliceosome genes. Although the current 2016 World Health Organization classification incorporates some genetic features in its diagnostic criteria for MDS and MDS/MPN

BACKGROUND: Offspring with a genetic predisposition to hypertension may have higher blood pressure (BP) at rest compared with those without a genetic predisposition to hypertension. They are also expected to have a higher sympathetic component in the heart rate variability (HRV) which could be computed with signal processing algorithms.
OBJECTIVE: The purpose of this study is to design a wavelet-based system to estimate the heart rate variability that can be used to detect early cardiovascular changes in offspring with a genetic predisposition to hypertension. Early detection will help in the

We present an infant referred to Developmental Paediatrics for delays, slow growth, hypotonia, esotropia and spasticity. Over the course of 2 months, the infant's exam progressed, demonstrating worsening spasticity and tonal changes in the setting of a normal brain MRI with acquired microcephaly. Genetic testing demonstrated a pathogenic CTNNB1 nonsense mutation. Following the discovery of the underlying cause for the child's clinical picture, the child was evaluated by therapeutic services and neurology, which was initially only available via asynchronous telehealth, due to a resource limited

The ommochrome and porphyrin body pigments that give freshwater planarians their brown color are produced by specialized dendritic cells located just beneath the epidermis. During embryonic development and regeneration, differentiation of new pigment cells gradually darkens newly formed tissue. Conversely, prolonged light exposure ablates pigment cells through a porphyrin-based mechanism similar to the one that causes light sensitivity in rare human disorders called porphyrias. Here, we describe a novel program using image-processing algorithms to quantify relative pigment levels in live anima

The use of the Storm Water Management Model (SWMM) to simulate flows in urban river watersheds necessitates the proper calibration of the various parameters involved in the process. Back Propagation Neural Network (BPNN) is often used to establish relationship between two sets of multivariate variables, such as parameters and simulation results of SWMM. The aim of this study is to establish an improved BPNN to calibrate SWMM. It was found that when using gauged flow data obtained from the urban river management system as calibration data, only using BPNN was not sufficient. An improved BPNN fr

We present an improved inverse-design approach for automatically identifying molecular (or other) systems with optimal values for prechosen properties. The new approach uses SMILES (simplified molecular input line entry system) to describe molecular structures efficiently, a genetic algorithm to optimize the molecules automatically, and the DFTB+ (self-consistent charge density functional tight-binding) method to calculate electronic properties. Thereby, almost every class of materials─even macromolecules or monomers─can be studied easily. Without crossover operators but with only mutation ope

PURPOSE: For neurodevelopmental disorders (NDDs), etiological evaluation can be a diagnostic odyssey involving numerous genetic tests, underscoring the need to develop a streamlined algorithm maximizing molecular diagnostic yield for this clinical indication. Our objective was to compare the yield of exome sequencing (ES) with that of chromosomal microarray (CMA), the current first-tier test for NDDs.
METHODS: We performed a PubMed scoping review and meta-analysis investigating the diagnostic yield of ES for NDDs as the basis of a consensus development conference. We defined NDD as global dev

Pheochromocytoma is a rare but important tumor of chromaffin cells that is frequently considered in the evaluation of hypertension, arrhythmias, or panic disorder and in the follow-up of patients with particular genetic diseases. This report provides an update about the genetics, neurochemical diagnosis, localization by imaging, and surgical management of pheochromocytoma. Specific mutations of the RET proto-oncogene cause familial predisposition to pheochromocytoma in multiple endocrine neoplasia type II, and mutations in the von Hippel-Lindau tumor suppressor gene cause familial disposition

BACKGROUND: Defining the phenotype is critical for investigating the genetic etiology of asthma. As part of the Collaborative Study on the Genetics of Asthma (CSGA), the primary objective of which is to identify asthma susceptibility loci, an algorithm was designed to determine diagnoses of definite asthma, probable asthma, less than probable asthma, or no asthma. A respiratory questionnaire was designed to assist in the process of characterizing the asthma phenotype.
OBJECTIVE: This study was designed to determine the validity of the CSGA algorithm for the diagnosis of asthma, to determine a

BACKGROUND: Molecular/genetic methods are becoming increasingly important for surveillance of diseases like malaria. Such methods allow monitoring routes of disease transmission or the origin and spread of variants associated with drug resistance. A confounding factor in molecular disease surveillance is the presence of multiple distinct variants in the same infection (multiplicity of infection - MOI), which leads to ambiguity when reconstructing which pathogenic variants are present in an infection. Heuristic approaches often ignore ambiguous infections, which leads to biased results.

BACKGROUND: Clustering of sequences into operational taxonomic units (OTUs) and denoising methods are a mainstream stopgap to taxonomically classifying large numbers of 16S rRNA gene sequences. Environment-specific reference databases generally yield optimal taxonomic assignment.
RESULTS: We developed SpeciateIT, a novel taxonomic classification tool which rapidly and accurately classifies individual amplicon sequences ( https://github.com/Ravel-Laboratory/speciateIT ). We also present vSpeciateDB, a custom reference database for the taxonomic classification of 16S rRNA gene amplicon sequence

BACKGROUND AND AIMS: The early diagnosis of familial hypercholesterolaemia is associated with a significant reduction in cardiovascular disease (CVD) risk. While the recent use of statistical and machine learning algorithms has shown promising results in comparison with traditional clinical criteria, when applied to screening of potential FH cases in large cohorts, most studies in this field are developed using a single cohort of patients, which may hamper the application of such algorithms to other populations. In the current study, a logistic regression (LR) based algorithm was developed com

Peripheral T cell lymphomas (PTCL) comprise a diverse group of aggressive T-cell and NK-cell lymphomas with many subtypes sharing same treatment algorithms despite having different pathobiology and responses to treatment. The molecular advances made in discovery of genetic mutations that disrupt epigenetic modulation in some subtypes of PTCL such as angioimmunoblastic T cell lymphoma and PTCL-not otherwise specified (NOS) may explain the poor outcomes and unsatisfactory responses to frontline line CHOP and CHOP-like therapy seen in this group of lymphomas. In this article, we address the main

There is a great clinical and genetic heterogeneity in congenital myopathies. Myo-MRI with pattern recognition has become a first-line complementary tool in clinical practice for this group of diseases. For diagnostic purposes, whole-body imaging techniques are preferred when involvement is axial or diffuse, as in most congenital myopathies, because of involvement of the tongue, masticator, neck or trunk muscles. Myo-MRI is widely used to identify abnormalities in muscle signal, volume or texture. Recognizable profiles or patterns have been identified in many of these genetic myopathies. The r

Selecting informative genes is essential for building accurate and efficient diagnostic models, especially for high-dimensional gene expression data such as those related to Alzheimer's disease. These datasets typically involve thousands of genes with limited samples, increasing the risk of overfitting and computational complexity. To address this, we propose a LASSO-HHO Gene Selection (LHGS) framework. LASSO is first applied to reduce dimensionality by filtering irrelevant genes. A conditional HHO-based optimization stage is then applied only when the LASSO-selected subset does not achieve su

DNA rearrangements contribute to the formation of new chromosomes, which are often the foundation of speciation. Deciphering the order of DNA rearrangements facilitates the reconstruction of evolutionary trajectories from extant to ancestral chromosomes, a task that is computationally NP-hard. Here, a mathematically rigorous chromosome-rearrangement model is integrated with the "Chromosomal Inversion Path Exploration via Monte-Carlo Tree Search (CIPE-MCTS)" framework in a deeply coupled manner. This integration yields a robust analytical framework that ensures both global optimality and comput

Accurate identification of disease-associated microRNAs (miRNAs) is crucial for elucidating pathogenic mechanisms and advancing therapeutic discovery. Although computational methods, particularly those based on biological networks, have become essential tools for predicting miRNA-disease associations, existing approaches often struggle to comprehensively learn from heterogeneous data and optimize feature representations. To overcome these limitations, we propose the Multi-view Hybrid Attention Graph Convolutional Network (MV-HAGCN). This framework constructs a comprehensive heterogeneous netwo

Reticular materials have come to the fore of chemistry with exceptional potential in applications ranging from CO2 capture and chemical separations to catalysis and drug delivery. However, due to the vast combinatorial space of molecular building blocks that can form these materials, designing high-performing reticular materials for applications remains a considerable challenge. Here, we present a computational approach that combines a library of molecular fragments suitable for constructing organic building units, template-based reassembly, and evolutionary optimization to accelerate the disc

Normal development of the immune system is essential for overall health and disease resistance. Bony fish, such as the zebrafish (Danio rerio), possess all the major immune cell lineages as mammals and can be employed to model human host response to immune challenge. Zebrafish neutrophils, for example, are present in the transparent larvae as early as 48 hours post fertilization and have been examined in numerous infection and immunotoxicology reports. One significant advantage of the zebrafish model is the ability to affordably generate high numbers of individual larvae that can be arrayed in
