Leukocyte adhesion deficiency syndromes: adhesion and tethering defects involving beta 2 integrins and selectin ligands.

Leukocyte adhesion deficiency (LAD) syndromes are failures of innate host defenses against bacteria, fungi, and other microorganisms resulti … recognized by the selectin family of adhesion molecules as well as defective glycosylation of other glycoconjugates. The defect has recently been attributed to mutations in a novel fucose transporter localized to the Golgi apparatus. Establishing the molecular basis for LAD syndromes has generated insights into mechanisms of leukocyte accumulation relevant to a b …

Sialic acid derivatives and their distribution in rat sublingual gland acini during pre- and post-natal development.

Sialoglycoconjugates in rat sublingual gland acinar cells, at different stages of pre- and post-natal development, were investigated in situ with specific lectins and by the selective removal of terminal sialic acids. Cleavage of acetyl substituents sited in the pyranose ring and/or polyhydroxyl side chain was used as an additional means of characterising the glycoconjugates. The first expression of terminal sialic acid linked to beta-galactose was found at gestational day 17 and progressive different derivatives were observed. The terminal disaccharide sialic acid-N-acetylgalactosamine was co…

Measurement of antibody response to keyhole limpet haemocyanin was not adequate for early diagnosis of schistosomiasis in a group of Dutch visitors to Mali.

The detection of antibodies against keyhole limpet haemocyanin (KLH) (extracted from Megathura crenulata) has been described as a reliable t … s found with an immunofluorescence assay for determination of IgM antibodies against gut-associated glycoconjugates. There was no significant difference in the anti-KLH response before, and one year after, therapy. These results indicate that, for travellers from non-endemic areas like The Netherlands with generally light schistosome infections, detection of anti …

Endothelial-leukocyte adhesion molecules in human disease.

An effective host response to infection or tissue damage requires focal accumulation of leukocytes. Leukocyte adhesion to the vessel wall, a key step in this process, depends on the ordered expression of specific endothelial cell surface molecules. The endothelial molecules that support adhesion include selectins that recognize leukocyte cell surface glycoconjugates as well as members of the immunoglobulin superfamily that interact with leukocyte integrins. Although inflammation can occur with minimal damage to the vessel wall and surrounding tissues, control mechanisms sometimes appear to fai…

Selective binding of soybean agglutinin to the olfactory system of Xenopus.

The binding patterns of four different lectins conjugated to horseradish peroxidase were investigated in the nervous system of juvenile Xeno … rons. The selective binding of this lectin of olfactory neurons suggests that specific cell surface glycoconjugates binding soybean agglutinin may have either a functional or developmental role in the olfactory system of Xenopus.

Ganglioside changes in the regenerating goldfish optic system: comparison with glycoproteins and phospholipids.

Axonally transported radioactivity in sialoglycoconjugates, labeled by intraocular injection of [3H]N-acetylmannosamine, increased significantly during regeneration of goldfish optic axons at 30 degrees C. Ganglioside radioactivity showed the largest increase--approximately eightfold--in the optic nerve tract at 8 days after optic nerve crush while sialoglycoprotein radioactivity increased fourfold under the same conditions. As regeneration proceeded the magnitude of the increase in the nerve tract diminished for both glycoconjugates. In the optic tectum, however, transported radioactivities r…

The development of a broad-spectrum retaining β-exo-galactosidase activity-based probe.

Acid β-galactosidase (GLB1) and galactocerebrosidase (GALC) are retaining exo-β-galactosidases involved in lysosomal glycoconjugate metabolism. Deficiency of GLB1 may result in the lysosomal storage disorders GM1 gangliosidosis, Morquio B syndrome, and galactosialidosis, and deficiency of GALC may result in Krabbe disease. Activity-based protein profiling (ABPP) is a powerful technique to assess the activity of retaining glycosidases in relation to health and disease. This work describes the use of fluorescent and biotin-carrying activity-based probes (ABPs) to assess the activity of both GLB1

Galectin-1: A Traditionally Immunosuppressive Protein Displays Context-Dependent Capacities.

Galectin-Carbohydrate interactions are indispensable to pathogen recognition and immune response. Galectin-1, a ubiquitously expressed 14-kDa protein with an evolutionarily conserved β-galactoside binding site, translates glycoconjugate recognition into function. That galectin-1 is demonstrated to induce T cell apoptosis has led to substantial attention to the immunosuppressive properties of this protein, such as inducing naive immune cells to suppressive phenotypes, promoting recruitment of immunosuppressing cells as well as impairing functions of cytotoxic leukocytes. However, only in recent

Biochemical and biological characteristics of cross-reacting material 197 CRM197, a non-toxic mutant of diphtheria toxin: use as a conjugation protein in vaccines and other potential clinical applications.

The biochemical and biological characteristics of CRM(197) are reviewed. Polysaccharide protein conjugate vaccines represent an important technological advancement that allowed for protection against dangerous diseases in vulnerable populations such as infants. The first carrier proteins, diphtheria and tetanus toxoids, were chosen in the context of an extensive body of information describing their immunogenicity and safety profiles in clinical use. These carriers perform well, and they require detoxification. A non-toxic mutant of diphtheria toxin, cross-reacting material 197 (CRM(197)), is a

Group B Streptococcus bacteremia elicits beta C protein-specific IgMand IgG in humans.

Group B Streptococcus (GBS) beta C protein elicits protective antibodies in experimental animals, making beta C protein an attractive component of a human GBS glycoconjugate vaccine. We determined whether natural exposure to beta C protein elicits antibodies in humans. Geometric mean concentrations (in micrograms per milliliter) of beta C-specific immunoglobulin (Ig) M and IgG as determined by enzyme-linked immunosorbent assay were similar in serum from 16 colonized (0.82 and 0.76, respectively) and 48 age-matched noncolonized (0.96 and 0.74, respectively) pregnant women. Size determination of bacterial capsular oligosaccharides used to prepare conjugate vaccines against Neisseria meningitidis groups Y and W135.

The glycoconjugate vaccines against Neisseria meningitidis groups Y and W135 consist of pools of selected oligosaccharides conjugated to the protein carrier (CRM197). Consistent production of these vaccines requires control and thus determination of the average degree of polymerisation of the oligosaccharides used for conjugation. Acid hydrolysis generates group Y and W135 oligosaccharides with N-acetylneuraminic acid at the reducing end. A method, involving NaBH4 reduction and quantification of this terminal N-acetylneuraminic acid by use of high performance anion-exchange chromatography with

Incidental presence of antibodies against gangliosides in Graves' disease.

In Graves' disease an increased immunological activity against glycoconjugate antigens is observed. Since gangliosides seem to be important for thyroid stimulation we searched for the presence of antiganglioside antibodies in Graves' patients sera by the immunoenzymatic method. Antibodies against some of 12 different gangliosides used in this study were found in 22% of the patients. However, in most cases the titer of antiganglioside antibodies was very low. Only the serum from one patient reacted with FucGM1 ganglioside at the dilution up to 1:1300. Anti-FucGM1 antibodies were present in seru

Infection microenvironment-responsive guanidine-modified nanostructures for precision photothermal therapy of diabetic wounds.

Diabetic wounds are among the most challenging complications of diabetes, particularly when persistent bacterial biofilms sustain infection, oxidative stress, and inflammation. Here, we developed an infection-microenvironment-responsive nanotherapeutic platform (AuNCs@PGD) by associating guanidine-rich PGD polymers with gold nanoclusters (AuNCs). The cationic PGD coating promotes interaction with negatively charged bacterial and biofilm components, while phenylboronic-acid motifs provide additional affinity for diol-containing surface glycoconjugates. Under 808-nm irradiation, AuNCs@PGD produc…

AgOTf-Assisted β-Glucuronidation Enables Convergent Synthesis of β-(1 → 4)-Linked Polyglucuronates up to the 32-Mer.

β-Glucuronides are widespread motifs in biologically important glycoconjugates and oligo/polysaccharides. However, the C-5 carboxylate of glucuronic acid renders glucuronosyl donors poor electrophiles and glucuronosyl acceptors weak nucleophiles, often leading to unproductive glycosylation. We disclose a robust, general gold(I)-catalyzed β-glucuronidation using glucuronosyl 8-alkynyl-1-naphthoate donors with the assistance of AgOTf catalysis. Mechanistic studies reveal the role of AgOTf: it promotes orthoester-to-β-glycoside conversion, likely through trace acid generated from the interaction …

Discovery of molecular glue degrader for treatment of IMiDs-resistant multiple myeloma based on GLUT-mediated tumor targeting strategy.

Based on the tumor Warburg effect and the strategy of GLUT-mediated drug transport, we have successfully discovered a promising molecular glue immunomodulatory drug (IMiD) candidate that exhibits excellent dual-targeting capability in tumor selective drug uptake, immunotherapy and combating lenalidomide (LEN) and pomalidomide (POM) resistance in multiple myeloma. The structure-activity relationship study culminated in the identification of the lena- and pomalidomide-glycoconjugates (1 and 20) with potent antitumor activity in cancer cells overexpressing GLUT1, especially in the highly resistan…

Synthesis of oligosaccharides from terminal B. pertussis LPS pentasaccharide and definition of the minimal epitope recognized by anti-pertussis antibodies.

Pertussis vaccines have been very effective in controlling whooping-cough epidemics but are ineffective in controlling circulation in older children and adults, thus facilitating the onset of future outbreaks. Antibodies against the lipopolysaccharide could reduce the carriage of the bacteria, its circulation, and transmission. The oligosaccharide fragments from the lipopolysaccharide may become a potential complement to existing vaccines in the form of protein glycoconjugates. An important step in the development of this type of vaccine is defining the minimal oligosaccharide epitope recogniz…

Brain glycogen serves as a critical glucosamine cache required for protein glycosylation.

Glycosylation defects are a hallmark of many nervous system diseases. However, the molecular and metabolic basis for this pathology is not fully understood. In this study, we found that N-linked protein glycosylation in the brain is metabolically channeled to glucosamine metabolism through glycogenolysis. We discovered that glucosamine is an abundant constituent of brain glycogen, which functions as a glucosamine reservoir for multiple glycoconjugates. We demonstrated the enzymatic incorporation of glucosamine into glycogen by glycogen synthase, and the release by glycogen phosphorylase by bio…
